Læring og hukommelse er en evolusjonært gammel tilpasningsmekanisme for miljøet, bevart fra Drosophila (D.) til human1. Menn var imidlertid litt raskere mens de kom inn i det øvre rommet i den første studien, mens kvinner fikk et litt høyere antall støt i hver oppbevaringsstudie. Det vestlige kostholdet (WD) svekket læring og minne betydelig hos mannlige fluer mens flyøvelse motvektet denne effekten. Samlet sett tilbyr den passive unngåelsesatferden i fluer en enkel og reproduserbar analyse som kan brukes til å studere grunnleggende mekanismer for læring og minne.

Den passive unngåelse ble studert i D. melanogaster (Canton-S) og D. simulans. Eksperimentene sammenlignet ventetidene og antall mottatte sjokk mellom påfølgende studier. Denne protokollen beskriver en enkel atferdsanalyse som gjør det mulig å studere negativ assosiativ læring og minnekonsolidering i fluer, inkludert effekter av genmiljøinteraksjoner. Analysen som presenteres her er en enkel, reproduserbar og kostnadseffektiv prosedyre for å studere minnemekanismer som enkelt kan monteres med noen få tilgjengelige forsyninger. Denne analysen kan gi innsikt i grunnleggende mekanismer som ligger til grunn for læring og hukommelseshemminger som følge av ulike genetiske, farmakologiske og kostholdsmanipulasjoner.Å demonstrere denne prosedyren vil være Morgan Tedder og Steven Bradley, studenter fra laboratoriet mitt. Til å begynne med, bor et fire millimeter hull vinkelrett rundt fire millimeter fra bunnen innenfor et 14 milliliter polypropylenkulturrør. Fjern den øvre delen av kulturrøret for å lage et 45 millimeter bunnfragment som brukes som det nedre rommet.Klipp av toppen av en 1000 mikroliter pipettespiss for å lage et 12 millimeter fragment. Sett fragmentet inn i kulturrørets fire millimeter hull som en lastedokk for å overføre fluene. Klipp et 15 millimeter stykke gjennomsiktig vinylrør og sett det nedre og øvre rommet fra motsatte ender inn i slangen.Fest enheten vertikalt ved hjelp av en justerbar klemme med to pinner. Suge forsiktig en fly inn i munnen aspirator ved å tegne luft.Deponer fluen ved å blåse lett inn i lastedokken og start umiddelbart en ett-minutts timer og stoppeklokke. Slå på stimulatoren og lever et elektrisk støt når flyet kommer inn i støtrøret. Trykk på stoppeklokken for å registrere den første ventetiden.Hvis flyet reenters sjokkrøret, levere ekstra støt. Bruk en opptelling eller en Arduino-basert teller til å registrere antall støt i løpet av en ett-minutts prøveperiode. Overfør forsiktig flyet tilbake til hetteglasset på slutten av en ett-minutts prøveperiode.Registrer ventetiden, antall mottatte sjokk og eventuelle merkbare endringer i atferd. Bruk 70% etanol, rengjør nedre og støtrom. Tørk av med et lofritt rengjøringsvev og tørk det med en hårføner.Gjenta rettssaken med neste flue. Rengjør det nedre rommet med vann og luktfritt vaskemiddel etter at eksperimentet er fullført. Bruk 70% etanol, tørk av det nedre og støtkammeret og lufttørk over natten.Ved å gjenta prosessen ovenfor etter 24 timer, utfør en ny prøveversjon. I en lignende rekkefølge, test fluene. Etter å ha utført en passiv unngåelsesanalyse i Drosophila melanogaster mannlige fluer, har resultatene vist at ventetiden øker, antall støt avtar, noe som viser at fluer lærte assosiasjon mellom det øvre rommet og et elektrisk støt.Frekvensfordeling avslørte at de fleste fluer kommer inn i det øvre rommet og mottar ett til tre støt i den første studien. I etterfølgende eksperimenter ble prøvevarigheten redusert til ett minutt fordi frekvensfordelingen for ventetider tydelig viste at hvis flyet ikke kommer inn i det øvre rommet innen 60 sekunder i den første studien, går den vanligvis ikke inn i det hele tatt.De passive unngåelsesanalyseeksperimentene ble gjentatt i Drosophila simulans mannlige og kvinnelige fluer. Resultatene har vist at fluene var effektive til å lære den passive unngåelsesatferden som tydelig fra endringene i frekvensfordelinger for ventetider og sjokk hos mannlige og kvinnelige fluer. Sammenligning av ventetider og antall sjokk mellom mann og kvinne viste ingen statistisk signifikante forskjeller.Imidlertid fikk kvinnelige fluer litt flere støt i hver studie. Analyse av grooming kamper i forsøk to og tre avslørte at den totale varigheten av grooming reduseres betydelig i kvinnelige fluer fra prøve en til prøve tre. Dette antyder at fluer opplever atferdsstress som paralleller gnager angstlignende oppførsel.Sammenligning av effekten av det vestlige kostholdet og flyøvelsen på passiv unngåelsesatferd viser at det vestlige kostholdet reduserer ventetiden og øker antall sjokk som indikerer at det vestlige kostholdet svekker aversiv assosiativ læring i fluer. Omvendt reduserte flyøvelsen den negative effekten av det vestlige kostholdet. Når du prøver denne prosedyren, husk å ikke stresse fluer.Stressede fluer ville enten komme inn i det øvre rommet for raskt eller vil ikke komme inn i det hele tatt. Alle eksperimenter skal utføres under de samme miljøforholdene og samtidig på dagen. Denne teknikken gir forskere en ny enkel analyse som kan brukes til å dissekere iboende mekanismer for læring og minnekonsolidering i fluer.
