Descreve-se aqui um conjunto de métodos que podem ser utilizados para induzir lesões por arma branca no tecto óptico, utilizando uma agulha para facilitar a avaliação da proliferação e diferenciação do CGR após lesão cerebral. A facada mediada por agulha é um método simples e eficientemente implementado que pode ser aplicado a muitas amostras experimentais usando um conjunto padrão de ferramentas. O peixe-zebra (Danio rerio) tem maior capacidade de regeneração do sistema nervoso central (SNC) quando comparado a outros mamíferos 1,2,3. Recentemente, para melhor compreender os mecanismos moleculares subjacentes a esse aumento da capacidade regenerativa, análises comparativas da regeneração tecidual utilizando tecnologia de sequenciamento de última geração têm sidorealizadas4,5,6. As estruturas cerebrais do peixe-zebra e dos tetrápodes são bastante diferentes 7,8,9. Isso significa que vários modelos de lesão cerebral usando peixes pequenos com estruturas cerebrais e características biológicas semelhantes foram desenvolvidos para facilitar a investigação dos mecanismos moleculares subjacentes que contribuem para esse aumento da capacidade regenerativa.
Além disso, o medaka (Oryzias latipes) é um animal de laboratório popular com baixa capacidade de regeneração cardíaca e neuronal10,11,12,13 em comparação com o peixe-zebra. Zebrafish e medaka têm estruturas cerebrais e nichos semelhantes para células-tronco neurais adultas (NSCs)14,15,16,17. Em zebrafish e medaka, o tecto óptico inclui dois tipos de NSCs, células-tronco neuroepiteliais e células gliais radiais (RGCs)15,18. Uma lesão por arma branca no tecto óptico de zebrafish adultos foi previamente desenvolvida, e este modelo foi usado para investigar os mecanismos moleculares que regulam a regeneração cerebral nesses animais 19,20,21,22,23. Este modelo de lesão por arma branca induziu neurogênese regenerativa a partir de CGRs 19,24,25. Esta lesão por arma branca no tecto óptico é um método robusto e reprodutível13,19,20,21,22,23,24,25. Quando o mesmo modelo de lesão foi aplicado ao medaka adulto, a baixa capacidade neurogênica dos CGRs no tecto óptico de medaka foi revelada através da análise comparativa da proliferação e diferenciação do CGR após alesão13.
Modelos de lesão por arma branca no tecto óptico também foram desenvolvidos em modelos de mummichog26, mas detalhes da lesão tectômica não foram bem documentados quando comparados com lesão telencálica27. A lesão por arma branca no tecto óptico utilizando zebrafish e medaka permite a investigação das respostas celulares diferenciais e expressão gênica entre espécies com capacidade regenerativa diferencial. Este protocolo descreve como realizar uma lesão por arma branca no tecto óptico usando uma agulha de injeção. Este método pode ser aplicado a pequenos peixes como zebrafish e medaka. Os processos de preparação de amostras para análise histológica e análise de proliferação e diferenciação celular usando imunohistoquímica fluorescente e criossecções são explicados aqui.

stab wound injury model of the adult optic tectum using zebrafish and medaka for the comparative analysis of regenerative capacity

Enquanto o peixe-zebra tem uma capacidade superior de regenerar seu sistema nervoso central (SNC), o medaka tem uma menor capacidade regenerativa do SNC. Um modelo de lesão cerebral foi desenvolvido no tecto óptico adulto de zebrafish e medaka e análises histológicas e moleculares comparativas foram realizadas para elucidar os mecanismos moleculares que regulam a alta capacidade regenerativa deste tecido entre essas espécies de peixes. Aqui é apresentado um modelo de lesão por arma branca para o tecto óptico adulto usando agulha e análises histológicas para proliferação e diferenciação das células-tronco neurais (NSCs). Uma agulha foi inserida manualmente na região central do tecto óptico e, em seguida, os peixes foram perfundidos intracárdicos e seus cérebros dissecados. Esses tecidos foram então criossecados e avaliados por imunomarcação contra os marcadores apropriados de proliferação e diferenciação do NSC. Este modelo de lesão tectum fornece resultados robustos e reprodutíveis tanto em zebrafish quanto em medaka, permitindo comparar as respostas do NSC após a lesão. A lesão por arma branca é um método de lesão mecânica que induz ablação celular inespecífica. Este método facilita a avaliação da proliferação e diferenciação do CGR após lesão cerebral. A lesão por arma branca mediada por agulha é um método simples e eficientemente implementado para induzir lesão cerebral.Este método pode ser aplicado a muitas amostras experimentais usando um conjunto padrão de ferramentas. A capacidade regenerativa do peixe-zebra foi ilustrada usando o modelo de lesão por arma branca. Novos conhecimentos sobre o mecanismo molecular que regula a regeneração neuronal contribuem para a terapia regenerativa no sistema nervoso central.Comece colocando um peixe-zebra adulto anestesiado ou medaka em uma bandeja de isopor. Fixe o peixe na vertical entre duas agulhas de calibre 30 inseridas verticalmente no isopor. Segure o corpo do peixe para evitar que a cabeça do peixe se mova e, em seguida, insira verticalmente uma agulha de calibre 30 através do crânio na região medial de uma das esferas do tecto óptico.Em seguida, transfira os peixes feridos para um tanque de peixes com água fresca da instalação de peixes. Uma vez que os peixes tenham se recuperado totalmente da anestesia, mova o tanque para o sistema de reprodução. Depois de cortar a medula espinhal e remover o crânio do tecto óptico e telencéfalo, cortar os nervos ópticos e dissecar o cérebro.Em seguida, transfira os cérebros para tubos de 1,5 mililitro com um mililitro de paraformaldeído a 4% em PBS, para fixar durante a noite a quatro graus Celsius. Depois de lavar os cérebros fixos três vezes em PBS por cinco minutos cada, transfira os cérebros em tubos de 1,5 mililitro com um mililitro de sacarose de 30% do peso em volume em PBS como um crioprotetor e incube-os durante a noite a quatro graus Celsius. Prepare o composto de incorporação combinando 30% de sacarose com 30 mililitros de temperatura de corte ideal, ou OCT, composto e armazene o composto a quatro graus Celsius.Para resfriar o criomold uma vez, incube um bloco de alumínio durante a noite a 80 graus Celsius negativos e coloque o bloco pré-resfriado em uma caixa de isopor com tampa para evitar o aquecimento. Em seguida, coloque o criomold no bloco de alumínio pré-resfriado na caixa de isopor e deixe o composto OCT congelar e ficar branco.À medida que o composto congela, aplique um pequeno círculo de composto OCT em um disco de amostra antes de montar o criobloco no disco da amostra. Ao colocar o disco da amostra no criostato, congele o composto de OCT. Em seguida, coloque o disco da amostra na cabeça da amostra no criostato.Depois de definir a orientação do criobloco, corte o OCT para remover quaisquer regiões extras. Corte seções seriais de 14 micrômetros de espessura através de todo o tecto óptico usando um criostático e armazene as lâminas a menos 25 graus Celsius. A proliferação de células gliais radiais, ou RGC, no tecido cerebral do peixe-zebra foi avaliada.A maioria dos CGRs estava proliferando com antígeno celular negativo no hemisfério contralateral não lesado. Dois dias após a lesão, a proliferação do RGC foi induzida no lado lesado do tecto medaka. Após a lesão cerebral, a linhagem celular e a diferenciação do RGC foram avaliadas com a marcação com bromodeoxiuridina, ou BrdU.A análise representativa mostra neurônios recém-nascidos sete dias após a lesão no peixe-zebra e medaka lesionados. Uma das etapas críticas nas lesões por arma branca é a inserção manual da agulha. Uma lesão consistente é necessária para criar resultados reprodutíveis.Corantes fluorescentes são eficazes como marcadores de feridas. A extensão da difusão do corante fluorescente indica a localização e a profundidade da lesão por arma branca. Esta técnica é potencialmente adaptada a outras espécies de peixes pequenos, e a adaptação ao medaka permite comparações da capacidade regenerativa.
