腸内細菌は、昆虫宿主の複数の生理学的プロセスに影響を与える可能性がある。軸索幼虫の準備と維持は、腸内細菌の機能を研究するための強力なツールです。このプロトコールの主な利点は、植物組織培養が、昆虫由来の表面滅菌卵を食べるアクセニックリーフを後方に無菌葉を得るために使用され、この方法が人工食によって制限されないことである。この方法は便利であり、将来の腸内細菌研究、特によく発達した人工飼料のない非モデル昆虫のために、軸索昆虫を維持することの可視性を高める。成長チャンバー内のP.versicolora個体群を摂氏27度、相対湿度70%に維持し、16時間の明光と8時間の暗光周期で維持する。傾いた濡れた吸収紙で昆虫を穴あきのプラスチック箱に入れ、新鮮なポプラの枝を与えます。吸湿紙にきれいな水をスプレーして水分を維持し、2日ごとに枝を交換します。蛹化後の産卵のために成虫を単離し、より多くの卵を得るために彼らに柔らかい葉を与える。24時間以内に、新しく産まれた卵を集め、湿った吸収紙の上に60時間置いて、軸索幼虫を調製する。MS培地を1ミリリットルあたり1ミリグラムのα−ナフタレン酢酸またはNAAストック溶液で調製する。バイオセーフティフード内で、500ミリリットルのMS培地に50マイクロリットルのNAAストック溶液を加え、よく混ぜるために振る。その後、組織培養容器あたり約50ミリリットルを注ぎ、固化を待つ。メスと鉗子をバイオセーフティフード内のアルコールランプの炎で滅菌します。約1ヶ月の成長でポプラの苗木から頂端芽または側芽で3〜4センチメートルの茎セグメントを切断し、容器ごとに1つまたは2つの茎セグメントを培地に挿入する。これらの茎セグメントを成長チャンバー内で約30日間インキュベートする。そして、これらの無菌の葉を使って、軸索の幼虫に餌を与えます。オートクレーブシャーレ、ペイントブラシ、ろ紙、LB寒天培地を含むシャーレに蒸留水。付着した卵の葉をペトリ皿に入れます。それから慎重に鉗子を使って葉から卵を取り除き、それらを別のペトリ皿に移します。これらの卵を75%エタノールで8分間洗浄する。その後、滅菌水で洗浄を4回繰り返す。卵をLB寒天培地に移して、孵化のために水分を保存します。ペトリ皿を成長室に入れ、卵が24時間以内に孵化するのを待つ。バイオセーフティフードに、ペトリ皿に濡れたろ紙を3枚篩い、無菌ポプラの葉を紙の上に置きます。幼虫を集めて葉の上に置きます。ペトリ皿をパラフィルムで密封し、成長室でインキュベートする。2日ごとに葉を交換してください。従来飼育されているグループでは、葉から湿ったろ紙を入れたペトリ皿に卵を移し、これらの幼虫に無菌ポプラの葉を与えます。無菌で従来飼育されている群から3つの第1、第2、第3齢幼虫をランダムに選択する。実体顕微鏡下で滅菌ハサミと鉗子で3齢幼虫を解剖し、微量遠心チューブに腸を採取する。無傷の第一および第二齢幼虫をチューブに集める。100マイクロリットルのPBSと3つの鋼球を1.5ミリリットルのチューブに加え、ビーズビートホモジナイザーを使用して組織をホモジネートに粉砕する。100マイクロリットルのホモジネートをLB寒天培地および滅菌ガラス拡散棒を用いてプレートに加える。プレートを摂氏37度で24時間インキュベートする。その後、細菌コロニーを観察します。DNA抽出キットを用いて先に得られた組織の全DNAを抽出し、分光光度計でDNA濃度を測定した。PCRでユニバーサル16S rRNAプライマーを用いて細菌の16S rRNA遺伝子を増幅し、本文原稿に記載されているように反応をセットアップする。PCR 産物を、さらに分析が進むまで摂氏 4 度で保管してください。PCR産物を核酸色素と混合し、1X TAEバッファー中の1%アガロースゲル上で電気泳動を使用して分析します。10マイクロリットルのDNAマーカーを基準として使用します。UVトランスイルミネーターでゲルを観察し、1,500塩基対の周りのターゲットフラグメントを探します。いずれの無菌群においても細菌コロニーは観察されなかったが、従来飼育群すべてで観察され、ポプラ葉を組織培養した滅菌卵由来の幼虫には細菌が含まれていないことが示された。この1,500塩基対PCRバンドは、従来飼育された全ての群に出現した。対照的に、無菌群またはネガティブコントロールではバンドは観察されず、軸索幼虫に腸内細菌が存在しなかったことを示唆した。小さな卵を持つ昆虫種の場合、消毒剤の種類と滅菌期間を最適化する必要がありました。さらに、組織培養植物における内生植物は注目に値するべきである。このプロトコルは、無菌昆虫を維持するための新しい方法を提供し、特によく発達した人工飼料のない非モデル昆虫の場合、昆虫 - 腸内細菌相互作用研究を促進するための便利なツールである。
