Den vellykkede integration af mikrobielle agenser til biologisk bekæmpelse af vigtige landbrugsinsektskadegørere i et agroøkosystem afhænger af både succes og let masseproduktion af entomopatogenerne som det første skridt under laboratorieforhold. Masseproduktionen af EPF er vigtig for den store anvendelse og tilgængelighed af EPF-produkter til IPM-programmer, der bruger biologisk kontrol 9,10,11,12,13.<sup cla...

Entomopatogene svampe (EPF) har fået betydning som plantebeskyttelsesmidler i den biologiske bekæmpelse af vigtige landbrugsinsektskadegørere 1,2. Entomopatogenerne, som forekommer naturligt i jorden, forårsager epizootier i populationerne af forskellige skadedyrsarter3. EpF-arterne er værtsspecifikke og udgør relativt få risici med hensyn til at angribe ikke-målarter, og de er ugiftige for miljøet4. EPF har en unik mekanisme til at invadere deres vært samt til at udbrede og fortsætte i deres umiddelbare miljø1. De angriber værten hovedsageligt gennem aseksuelle sporer, der fastgøres til og trænger ind i værtskutiklen for at invadere og sprede sig i værtshæmokølen. Værten dør til sidst på grund af udtømning af hæmolymfenæringsstofferne eller som følge af toksæmi forårsaget af de toksiske metabolitter frigivet af svampen. Efter døden, under ideelle miljøforhold, fremkommer svampen på den ydre overflade (åbenlys mykose) af værtskadaveren 5,6.
Voksende bekymring over de negative virkninger af kemiske rester på menneskers sundhed, miljøforurening og udviklingen af skadedyrsresistens har ført til den globale indsats for at reducere input af kemisk baserede insekticider og finde alternative, nye og bæredygtige strategier for afgrødebeskyttelse og skadedyrsbekæmpelse 6,7,8 . Dette har givet mulighed for at udvikle mikrobielt baserede insekticider til brug i Integrated Pest Management (IPM) programmer, som er mere økologisk gunstige strategier end konventionel kemisk bekæmpelse 3,8.
For at udvikle et vellykket mikrobielt bekæmpelsesmiddel til et landbrugsskadegører skal en egnet organisme først isoleres, karakteriseres, identificeres og dens patogenicitet for målskadegøreren bekræftes. Der kræves imidlertid en nem og omkostningseffektiv metode til storskalaproduktion af det mikrobielle agens for at fremstille et levedygtigt produkt til brug i biologiske bekæmpelsesprogrammer 9,10,11,12,13. Masseproduktion af betydelige mængder entomopatogener af god kvalitet afhænger af den mikrobielle stamme, miljøet, målskadegøreren, formuleringen, markedet, anvendelsesstrategien og det ønskede slutprodukt 14,15,16. EPF kan masseproduceres ved hjælp af flydende substratfermentering til fremstilling af blastosporer eller fermenteringsprocessen for fast substrat til fremstilling af luftkonidier 6,17,18. Masseproduktions- og formuleringsprocessen af entomopatoggener påvirker imidlertid direkte virulensen, omkostningerne, holdbarheden og felteffektiviteten af slutproduktet. For en vellykket anvendelse i IPM skal produktionsprocessen for entomopatogerne være let at køre, kræve minimal arbejdskraft, producere en høj udbyttekoncentration af virulente, levedygtige og vedvarende propaguler og være lav i omkostninger 4,13,14,16.
Forståelse af de ernæringsmæssige behov for entomopatogener er vigtig for massedyrkning med alle dyrkningsmetoder 4,12. De ernæringsmæssige komponenter i produktionsmediet har en betydelig indvirkning på egenskaberne ved de resulterende propaguler, herunder biokontroleffektivitet, udbytte, udtørringstolerance og persistens 8,19,20,21. Optimeringen af produktionsprocedurerne er designet til at imødegå sådanne faktorer22. For EPF er de vigtigste krav til god vækst, sporulation og masseproduktion af svampekonidier tilstrækkelig fugtighed, optimal væksttemperatur, pH, gasudveksling af CO2 ogO2 og ernæring, herunder gode fosfor-, kulhydrat-, kulstof- og nitrogenkilder18.
Jaronski og Jackson18 beskriver fermenteringsmetoden med fast substrat som den mest effektive og tætteste tilnærmelsesmetode til den naturlige proces til EPF-produktion i forhold til fermenteringsmetoden for flydende substrat, fordi svampekonidiumet under naturlige forhold bæres på faste oprejste strukturer, som overfladen af insektkadavere. Landbrugsprodukter og biprodukter, der indeholder stivelse, anvendes for det meste til masseproduktion af hypokrealeanske svampe, da svampene let nedbryder stivelse gennem udskillelse af stærkt koncentrerede hydrolytiske enzymer fra deres bindestregsspidser, for at trænge ind i det faste stof og for at få adgang til de næringsstoffer, der er til stede i stoffet 11,17,18,23 . Kornprodukterne stiller også krav til sund biomasseproduktion, fordi substraterne, når de hydreres og steriliseres, kan optage yderligere næringsstoffer fra ethvert flydende medium 16,18,24.
Tidligere forsøgte flere undersøgelser at massekulturere EPF-arter som Beauveria bassiana (Bals.) Vuil., Cordyceps fumosorosea (Wize) Kelper B. Shrestha & Spatafora, Verticillium lecanii (Zimm.) Viegas og nogle af Metarhizium anisopliae (Metschn.) Sorokin artskompleks isolerer på forskellige substrater 16,23,24. Sådanne masseproducerede og kommercielt udviklede isolater omfatter Green Muscle® (stamme IMI 330189), udviklet fra M. anisopliae var Metarhizium acridum (Driver & Milner) J.F. Bisch, Rehner & Humber, Metarhizium 69 (Meta 69 stamme ICIPE69) og Real Metarhizium 69 (L9281), udviklet fra M. anisopliae, og bredbånd® (stamme PPRI 5339) og Eco-Bb®, udviklet fra B. bassiana25,26 . Der er dog gjort begrænsede forsøg på massekultur Metarhizium robertsii J.F. Bisch., S.A. Rehner & Humber og Metarhizium pinghaense Chen & Guo. Disse to isolater blev udvalgt i en tidligere undersøgelse som de mest effektive til bekæmpelse af mælkebuggen, Pseudococcus viburni Signoret (Hemiptera: Pseudococcidae)27. Derfor havde den nuværende undersøgelse til formål at formulere og masseproducere et tilstrækkeligt antal modstandsdygtige infektiøs propaguler af de lokale isolater af M. robertsii og M. pinghaense til kommerciel anvendelse mod skadedyr. Den faste substratfermenteringsmetode blev brugt til at masseproducere svampekonidierne for begge EPF-isolater. To EPF-podningsmetoder ved anvendelse af konidiale suspensioner og den flydende svampekultur af blastosporer blev anvendt til at inokulere de faste substrater.

mass production of entomopathogenic fungi, metarhizium robertsii and metarhizium pinghaense, for commercial application against insect pests

Entomopatogene svampe i Metarhizium anisopliae artskomplekset har fået betydning som de biologiske bekæmpelsesmidler for landbrugsinsektskadedyr. Stigningen i skadedyrsresistens over for kemiske insekticider, den voksende bekymring over insekticidernes negative virkninger på menneskers sundhed og miljøforureningen fra pesticider har ført til en global indsats for at finde nye bæredygtige strategier for afgrødebeskyttelse og skadedyrsbekæmpelse. Tidligere er forsøg på at massekulturere sådanne entomopatogene svampe (EPF) arter som Beauveria bassiana blevet udført. Der er dog kun gjort begrænsede forsøg på at massedyre Metarhizium robertsii og M. pinghaense til brug mod skadedyr. Denne undersøgelse havde til formål at masseproducere et tilstrækkeligt antal modstandsdygtige infektiøs propaguler af sydafrikanske isolater af M. robertsii og M. pinghaense til kommerciel anvendelse. Tre landbrugskornprodukter, flager havre, flager byg og ris, blev brugt som EPF faste gæringssubstrater. To podningsmetoder, konidiale suspensioner og den flydende svampekultur af blastosporer blev brugt til at inokulere de faste substrater. Inokulation ved hjælp af konidiale suspensioner blev observeret at være relativt mindre effektiv, da der blev observeret øgede niveauer af kontaminering på de faste substrater i forhold til ved anvendelse af blastospore-podningsmetoden. Flaget havre viste sig ikke at være et egnet vækstsubstrat for både M. robertsii og M. pinghaense, da der ikke blev høstet tørre conidier fra substratet. Flaked byg viste sig at favorisere produktionen af M. robertsii conidia frem for M. pinghaense, og et gennemsnit på 1,83 g ± 1,47 g tør M. robertsii conidia og nul gram M. pinghaense conidia blev høstet fra substratet. Riskorn viste sig at favorisere den konidiale masseproduktion af både M. pinghaense og M. robertsii isolater, med et gennemsnit på 8,2 g ± henholdsvis 4,38 g og 6 g ± 2 g høstet fra substratet.

Et fald i indholdsmassen af kulturerne på ris for både M. pinghaense og M. robertsii blev observeret over tid under tørringsfasen af svampekulturerne, hvor der ikke blev observeret nogen eller lidt ændring i massen, når kulturerne var tørre (figur 5). Mass Production of Entomopathogenic Fungi, Metarhizium robertsii and Metarhizium pinghaense, for Commercial Application Against Insect Pests

Entomopatogene svampe har fået betydning som biologiske bekæmpelsesmidler for skadedyr i landbruget. I denne undersøgelse blev masseproduktionen af et tilstrækkeligt antal elastiske infektivende propaguler af sydafrikanske isolater af både Metarhizium robertsii og M. pinghaense til kommerciel anvendelse mod skadedyr med succes udført ved anvendelse af landbrugskornprodukter.

Masseproduktion af entomopatogene svampe, Metarhizium robertsii og Metarhizium pinghaense til kommerciel anvendelse mod skadedyr

Denne metode giver nye oplysninger om, hvordan man masseproducerer infektivende conidier af entomopatogene svampe til bekæmpelse af vigtige insektskadegørere i kommercielle landbrugsøkosystemer i betragtning af den nuværende situation, hvor de fleste insekticider udfases på grund af bekymring for deres toksiske virkning på miljøet og menneskers sundhed. Teknikken beskriver den metode, der anvendes til at sikre det optimale udbytte af EPF-conidier under masseproduktion. Metoden er nyttig til integreret skadedyrsbekæmpelse og biologisk bekæmpelse ved at give indsigt i effektiviteten af masseproducerede conidier til storstilet håndtering af skadelige insektskadedyr med insekticidresistens i agroøkosystemerne.Denne procedure blev udviklet og demonstreret af Dr. Letodi Luki Mathulwe, Dr. Nomakholwa Faith Stokwe og professor Antoinette Paula Malan. Opvarm en liter destilleret vand og sluk for varmen, inden du når kogepunktet. Tilsæt nu 30 gram glucose, fire gram kaliumphosphat dibasisk, 20 gram gærekstrakt, 15 ml majsstejl spiritus, og bring indholdet i en blid kogning i to til tre minutter.Der hældes i alt 100 ml af mediet i ni forskellige kolber på 250 ml. Læg en vatprop på hver kolbe og dæk bomuldsulden med aluminiumsfolie som prop. Autoklaver mediet i kolberne i 55 minutter ved 121 grader Celsius.Senere afkøles mediet i kolberne, og der tilsættes 10 mg streptomycin pr. milliliter streptomycin til mediet i hver kolbe. Saml to til tre bakterielle sløjfer af svampekonidier fra to til tre uger gamle svampekulturplader til begge EPF-isolater. Svampekonidierne overføres til hver 100 ml flydende medie i 250 ml kolberne under sterile forhold, og kolberne forsegles.Inkubere de flydende dyrkningskolber ved ca. 25 grader Celsius på en orbitalryster ved 140 o / min i tre dage og ophøre med inkubationen, når kulturerne viser tegn på høj turbiditet med svampeblastosporevækst. For at påvise eventuel bakteriel kontaminering fra kulturerne udtages en prøve på 100 mikroliter fra hver flydende dyrkningskolbe efter 24 timers inkubation og plade på tre SDA-plader pr. isolat. Inkuber pladerne i 48 timer ved en kontrolleret temperatur på plus eller minus 25 grader Celsius.Brug et lille polyvinylchloridaffaldsrør til at skabe en hals til gæringsposen i den åbne ende af autoklaveposen, og brug autoklavetape til at fastgøre røret. Luk røret med en steril bomuldsuldprop for at muliggøre tilstrækkelig gasudveksling under gæringen. Brug parboiled langkornet hvid ris som et fast substratmedium til blastosporerne af både Metarhizium pinghaense og Metarhizium robertsii.Til hver gæringspose tilsættes et kilo ris og 300 ml sterilt destilleret vand. Bland forsigtigt indholdet af gæringsposerne. Placer dem i ydre autoklaveposer i opretstående stilling og autoklave ved 121 grader Celsius i 55 minutter.Efter autoklaven skal substraterne afkøles i ca. 45 minutter under sterile forhold. Lukningen af hver af de flydende dyrkningskolber af både Metarhizium pinghaense og Metarhizium robertsii fjernes under en laminær strømning, og hver kolbe flammes kant i 10 sekunder. Hæld 100 ml flydende kulturer i gæringsposerne ved at fjerne bomuldsuldpropperne fra halsen.Sæt bomuldspropperne på igen, og dæk toppen af posens hals med kirurgisk papir, der er fastgjort med et gummibånd. Drej den øverste del af posen og bland posens indhold ved at ryste og manipulere underlaget ved massage. Inkuber poserne ved ca. 25 grader Celsius ved at flade substratet i poserne for at forhindre dannelse af tykke senge.To til tre dage efter podning og inkubation, når synlig mycelial vækst og svampens binding af substratet havde fundet sted, brydes substratet i fermenteringsposerne ved at massere posernes indhold. Efter gæring overføres de sporulerede kulturer til 26 til 30 kg brune papirposer og tørres svampekulturerne i 10 til 12 dage, før de anvendes i forsøg. For at forbedre papirposernes trækstyrke skal du vandret afskære 1/3 af den øverste del af posen og beklæde bunden af posen.Smuldr forsigtigt substratet i hver gæringspose. Skær hjørnet af hver pose af og overfør langsomt hele kulturen til papirposerne gennem det rum, der efterlades af det afskårne hjørne. Mærk hver papirpose, fold over den øverste ende af hver pose to gange, og luk med hæfteklammer for at skabe en trekantet teltstruktur.Læg poserne på trådtørrestativer for at muliggøre korrekt, jævn tørring ved en kontrolleret temperatur på ca. 25 grader Celsius og lav luftfugtighed på 30 til 40%Høst svampekonidier fra kulturerne ved hjælp af tre indlejrede sigter monteret på en opsamlingspande. Læg langsomt tørkulturprøven på ETS-maskenummer 35 og sigten. Læg et låg på sigten for at forhindre frigivelse af svampekonidier i luften.Tilsæt 10 til 12 glaskugler til sigterne for at hjælpe med at passere svampekonidierne gennem maskeskærmene og undgå tilbageholdelse af conidierne i sigten, hvilket kan resultere i reduceret sporegenvinding. Tape og forsegl sigtesamlingerne ved hjælp af elektrisk tape. Anbring sigterne på en vibrationsryster udstyret med en klæbrig pude for at fastgøre opsamlingspanden og sigterne i 20 til 25 minutter med en bevægelse på 560 til 640 vibrationer pr. Minut.Fjern testsigterne fra opsamlingspanden, saml conidier, og opbevar de opsamlede conidier i lufttætte og vanduigennemtrængelige lynlåslåseposer til langtidsopbevaring. I gennemsnit blev ca. 1,83 gram tør Metarhizium robertsii conidia høstet fra det flagede bygsubstrat, mens nul Metarhizium pinghaense blev høstet fra substratet. Der blev ikke høstet tørre svampekonidier fra det flagede havresubstrat for nogen af Metarhizium-arterne.En signifikant forskel blev observeret mellem de to arter af Metarhizium i den estimerede gennemsnitlige conidia pr. Gram høstet fra riskornene. Metarhizium robertsii havde lidt højere gennemsnitligt conidiatal pr. gram end Metarhizium pinghaense.
