De succesvolle integratie van microbiële agentia voor de biologische bestrijding van belangrijke landbouwinsectenplagen in een agro-ecosysteem hangt af van zowel succes als gemak van massaproductie van de entomopathogenen als eerste stap onder laboratoriumomstandigheden. De massaproductie van EPF is belangrijk voor de grootschalige toepassing en beschikbaarheid van EPF-producten voor IPM-programma's met behulp van biologische bestrijding 9,10,11,12,13.</sup...

Entomopathogene schimmels (EPF) hebben aan belang gewonnen als gewasbeschermingsmiddelen in de biologische bestrijding van belangrijke landbouwinsectenplagen 1,2. De entomopathogenen, die van nature in de bodem voorkomen, veroorzaken epizoötie in de populaties van verschillende plaagsoorten3. De soorten EPF zijn gastheerspecifiek en vormen relatief weinig risico's in termen van het aanvallen van niet-doelsoorten, en ze zijn niet giftig voor het milieu4. EPF heeft een uniek mechanisme voor het binnendringen van hun gastheer, evenals voor het verspreiden en volharden in hun directe omgeving1. Ze vallen de gastheer voornamelijk aan via aseksuele sporen die zich hechten aan en doordringen in de nagelriem van de gastheer om binnen te dringen en zich te vermenigvuldigen in de gastheerhemocoel. De gastheer sterft uiteindelijk als gevolg van uitputting van de hemolymfe voedingsstoffen of als gevolg van de toxemie veroorzaakt door de toxische metabolieten die door de schimmel worden vrijgegeven. Na de dood, onder ideale omgevingsomstandigheden, komt de schimmel tevoorschijn op het buitenoppervlak (openlijke mycose) van het gastheerkadaver 5,6.
Groeiende bezorgdheid over de negatieve effecten van chemische residuen op de menselijke gezondheid, milieuvervuiling en de ontwikkeling van resistentie tegen plagen hebben geleid tot de wereldwijde drang om de input van insecticiden op chemische basis te verminderen en alternatieve, nieuwe en duurzame strategieën voor gewasbescherming en ongediertebestrijding te vinden 6,7,8 . Dit heeft mogelijkheden geboden om microbiële insecticiden te ontwikkelen voor gebruik in Integrated Pest Management (IPM) -programma's, die ecologisch gunstiger strategieën zijn dan conventionele chemische bestrijding 3,8.
Om een succesvol microbieel bestrijdingsmiddel voor een landbouwplaag te ontwikkelen, moet eerst een geschikt organisme worden geïsoleerd, gekarakteriseerd, geïdentificeerd en de pathogeniciteit ervan voor het doelorganisme worden bevestigd. Een eenvoudige, kosteneffectieve methode voor grootschalige productie van het microbiële agens is echter vereist om een levensvatbaar product te produceren voor gebruik in biologische bestrijdingsprogramma's 9,10,11,12,13. Massaproductie van aanzienlijke hoeveelheden entomopathogenen van goede kwaliteit is afhankelijk van de microbiële stam, het milieu, de doelplaag, de formulering, de markt, de toepassingsstrategie en het gewenste eindproduct 14,15,16. EPF kan in massa worden geproduceerd met behulp van vloeibare substraatfermentatie om blastospores te produceren of het fermentatieproces van vaste substraten om luchtconidia 6,17,18 te produceren. Het massaproductie- en formuleringsproces van entomopathogenen heeft echter een directe invloed op de virulentie, de kosten, de houdbaarheid en de veldeffectiviteit van het eindproduct. Voor succesvol gebruik in IPM moet het productieproces van de entomopathogenen gemakkelijk te uitvoeren zijn, minimale arbeid vereisen, een hoge opbrengstconcentratie van virulente, levensvatbare en persistente propagules produceren en goedkoop zijn 4,13,14,16.
Het begrijpen van de voedingsbehoeften van entomopathogenen is belangrijk voor massateelt met alle kweekmethoden 4,12. De voedingscomponenten van het productiemedium hebben een aanzienlijke invloed op de eigenschappen van de resulterende propagules, waaronder biocontrole-werkzaamheid, opbrengst, uitdrogingstolerantie en persistentie 8,19,20,21. De optimalisatie van productieprocedures is ontworpen om dergelijke factoren aan te pakken22. Voor EPF zijn de belangrijkste vereisten voor goede groei, sporulatie en massaproductie van schimmelconidia voldoende vocht, optimale groeitemperatuur, pH, gasuitwisseling van CO2 en O2 en voeding, waaronder goede fosfor-, koolhydraat-, koolstof- en stikstofbronnen18.
Jaronski en Jackson18 beschrijven de vaste substraatfermentatiemethode als de meest efficiënte en de dichtstbijzijnde benaderingsmethode voor het natuurlijke proces voor EPF-productie ten opzichte van de vloeibare substraatfermentatiemethode, omdat onder natuurlijke omstandigheden het schimmelconidium wordt gedragen op vaste rechtopstaande structuren, zoals het oppervlak van insectenkadavers. Landbouwproducten en bijproducten die zetmeel bevatten, worden meestal gebruikt voor de massaproductie van hypocrealean schimmels, omdat de schimmels zetmeel gemakkelijk ontbinden door afscheiding van sterk geconcentreerde hydrolytische enzymen uit hun hyphale uiteinden, om de vaste stof binnen te dringen en om toegang te krijgen tot de voedingsstoffen die aanwezig zijn in de stof 11,17,18,23 . De graanproducten bieden ook de vereisten voor een gezonde biomassaproductie, omdat, wanneer ze worden gehydrateerd en gesteriliseerd, de substraten verdere voedingsstoffen uit elk vloeibaar medium kunnen opnemen 16,18,24.
Eerder probeerden verschillende studies EPF-soorten zoals Beauveria bassiana (Bals. Vuil., Cordyceps fumosorosea (Wize) Kelper B. Shrestha & Spatafora, Verticillium lecanii (Zimm.) Viegas en enkele van de Metarhizium anisopliae (Metschn.) Sorokin soortencomplex isoleert op verschillende substraten 16,23,24. Dergelijke in massa geproduceerde en commercieel ontwikkelde isolaten omvatten Green Muscle® (stam IMI 330189), ontwikkeld uit M. anisopliae var Metarhizium acridum (Driver & Milner) J.F. Bisch, Rehner & Humber, Metarhizium 69 (Meta 69 stam ICIPE69), en Real Metarhizium 69 (L9281), ontwikkeld uit M. anisopliae, en Breedband® (stam PPRI 5339) en Eco-Bb®, ontwikkeld uit B. bassiana25.26 . Er zijn echter beperkte pogingen gedaan om Metarhizium robertsii J.F. Bisch., S.A. Rehner & Humber en Metarhizium pinghaense Chen & Guo te masseren. Deze twee isolaten werden in een eerdere studie geselecteerd als de meest effectieve voor de bestrijding van de wolluis, Pseudococcus viburni Signoret (Hemiptera: Pseudococcidae)27. Daarom was de huidige studie gericht op het formuleren en massaal produceren van een voldoende aantal veerkrachtige infectieuze propagules van de lokale isolaten van M. robertsii en M. pinghaense voor commerciële toepassing tegen insectenplagen. mass production of entomopathogenic fungi, metarhizium robertsii and metarhizium pinghaense, for commercial application against insect pests

Entomopathogene schimmels van het Metarhizium anisopliae soortencomplex hebben aan belang gewonnen als de biologische bestrijders van landbouwinsectenplagen. De toename van de resistentie tegen plagen tegen chemische insecticiden, de groeiende bezorgdheid over de negatieve effecten van insecticiden op de menselijke gezondheid en de milieuvervuiling door pesticiden hebben geleid tot een wereldwijde drive om nieuwe duurzame strategieën voor gewasbescherming en ongediertebestrijding te vinden. Eerder zijn pogingen gedaan om dergelijke entomopathogene schimmels (EPF) -soorten zoals Beauveria bassiana massaal te kweken. Er zijn echter slechts beperkte pogingen ondernomen om Metarhizium robertsii en M. pinghaense massaal te kweken voor gebruik tegen insectenplagen. Deze studie was gericht op het massaal produceren van een voldoende aantal veerkrachtige infectieuze propagules van Zuid-Afrikaanse isolaten van M. robertsii en M. pinghaense voor commerciële toepassing. Drie landbouwproducten, haver in vlokken, gerst in vlokken en rijst, werden gebruikt als de EPF vaste gistingssubstraten. Twee inentingsmethoden, conidiale suspensies en de vloeibare schimmelcultuur van blastospores werden gebruikt om de vaste substraten te enten. Inenting met behulp van conidiale suspensies bleek relatief minder effectief te zijn, omdat verhoogde niveaus van verontreiniging werden waargenomen op de vaste substraten ten opzichte van bij gebruik van de blastospore-inentingsmethode. Vlokken haver bleken geen geschikt groeisubstraat te zijn voor zowel M. robertsii als M. pinghaense, omdat er geen droge conidia uit het substraat werden geoogst. Vlokken gerst bleek de productie van M. robertsii conidia te bevoordelen boven die van M. pinghaense, en een gemiddelde van 1,83 g ± 1,47 g droge M. robertsii conidia en nul gram M. pinghaense conidia werd geoogst van het substraat. Rijstkorrels bleken de conidiale massaproductie van zowel M. pinghaense - als M. robertsii-isolaten te bevorderen, met een gemiddelde van respectievelijk 8,2 g ± 4,38 g en 6 g ± 2 g geoogst van het substraat.

Een afname van de inhoudsmassa van de culturen op rijst voor zowel de M. pinghaense als de M. robertsii werd in de loop van de tijd waargenomen tijdens het droogstadium van de schimmelculturen, waarbij geen of weinig verandering werd waargenomen in de massa zodra de culturen droog waren (figuur 5). Het geoogste droge schimmel conidia poeder van zowel de M. pinghaense als de M. robertsii is weergegeven in figuur 6.

Mass Production of Entomopathogenic Fungi, Metarhizium robertsii and Metarhizium pinghaense, for Commercial Application Against Insect Pests

Entomopathogene schimmels hebben aan belang gewonnen als de biologische bestrijders van landbouwinsectenplagen. In deze studie werd de massaproductie van een voldoende aantal veerkrachtige infectieuze propagules van Zuid-Afrikaanse isolaten van zowel Metarhizium robertsii als M. pinghaense voor commerciële toepassing tegen insectenplagen met succes uitgevoerd met behulp van landbouwproducten.

Deze methode biedt nieuwe informatie over hoe massaal infectieuze conidia van entomopathogene schimmels kunnen worden geproduceerd om belangrijke insectenplagen in commerciële agro-ecosystemen te bestrijden, gezien de huidige situatie waarin de meeste insecticiden worden uitgefaseerd vanwege bezorgdheid over hun toxische effect op het milieu en de menselijke gezondheid. De techniek beschrijft de methode die wordt gebruikt om de optimale opbrengst van EPF conidia tijdens massaproductie te garanderen. De methode is nuttig voor geïntegreerde plaagbestrijding en biologische bestrijding door inzicht te geven in de effectiviteit van massaproductie conidia voor grootschalig beheer van schadelijke insecticideresistentie-insectenplagen in de agro-ecosystemen.Deze procedure werd ontwikkeld en gedemonstreerd door Dr.Letodi Luki Mathulwe, Dr.Nomakholwa Faith Stokwe en Professor Antoinette Paula Malan. Verwarm een liter gedestilleerd water en schakel het vuur uit voordat u het kookpunt bereikt. Voeg nu 30 gram glucose, vier gram kaliumfosfaat dibasisch, 20 gram gistextract, 15 milliliter maïs steile vloeistof toe en breng de inhoud gedurende twee tot drie minuten zachtjes aan de kook.Giet in totaal 100 milliliter van het medium in negen verschillende kolven van 250 milliliter. Plaats een wattenplug op elke kolf en bedek de watten met aluminiumfolie als stop. Autoclaaf het medium in de kolven gedurende 55 minuten bij 121 graden Celsius.Laat het medium in de kolven later afkoelen en voeg 10 milligram per milliliter streptomycine toe aan het medium in elke kolf. Verzamel twee tot drie bacteriële lussen van schimmelconidia van twee tot drie weken oude schimmelkweekplaten voor beide EPF-isolaten. Breng de schimmelconidia onder steriele omstandigheden over op elke 100 milliliter vloeibare media in de kolven van 250 milliliter en sluit de kolven af.Incubeer de vloeibare kweekkolven bij ongeveer 25 graden Celsius op een orbitale shaker bij 140 RPM gedurende drie dagen en stop de incubatie zodra de culturen tekenen van hoge troebelheid vertonen met schimmelblastosporegroei. Om eventuele bacteriële besmetting van de culturen te detecteren, trekt u na 24 uur incubatie een monster van 100 microliter uit elke vloeibare kweekkolf en plaat op drie SDA-platen per isolaat. Incubeer de platen gedurende 48 uur bij een gecontroleerde temperatuur van plus of min 25 graden Celsius.Gebruik een kleine polyvinylchloride-afvoerpijp om een hals te maken voor de fermentatiezak aan het open uiteinde van de autoclaafzak en gebruik autoclaaftape om de pijp vast te zetten. Sluit de buis met een steriele wattenplug om voldoende gasuitwisseling tijdens de gisting mogelijk te maken. Gebruik voorgekookte langkorrelige witte rijst als een vast substraatmedium voor de blastospores van zowel Metarhizium pinghaense als Metarhizium robertsii.Voeg voor elke gistingszak een kilogram rijst en 300 milliliter steriel gedestilleerd water toe. Meng voorzichtig de inhoud van de gistingszakken. Plaats ze in buitenste autoclaafzakken rechtop en autoclaaf bij 121 graden Celsius gedurende 55 minuten.Laat na de autoclavering de substraten ongeveer 45 minuten afkoelen onder steriele omstandigheden. Verwijder de sluiting van elk van de vloeibare kweekkolven van zowel Metarhizium pinghaense als Metarhizium robertsii onder een laminaire stroom en vlam de rand van elke kolf gedurende 10 seconden. Giet 100 milliliter vloeibare culturen in de gistingszakken door de wattenpluggen uit hun nek te verwijderen.Plaats de katoenen pluggen terug en bedek de bovenkant van de hals van de tas met chirurgisch papier dat is vastgezet met een elastiekje. Draai het bovenste deel van de zak en meng de inhoud van de zak door het substraat te schudden en te manipuleren door massage. Incubeer de zakken bij ongeveer 25 graden Celsius door het substraat in de zakken plat te maken om de vorming van dikke bedden te voorkomen.Twee tot drie dagen na inenting en incubatie, wanneer zichtbare myceliale groei en de binding van het substraat door de schimmel hadden plaatsgevonden, breekt u het substraat in de fermentatiezakken door de inhoud van de zakken te masseren. Breng na de fermentatie de gesporuleerde culturen over in bruine papieren zakken van 26 tot 30 kilogram en droog de schimmelculturen gedurende 10 tot 12 dagen voordat ze in proeven worden gebruikt. Om de treksterkte van de papieren zakken te verbeteren, snijdt u horizontaal 1/3 van het bovenste deel van de zak af en bekleedt u de onderkant van de zak.Verkruimel het substraat voorzichtig in elke gistingszak. Snijd de hoek van elke zak af en breng langzaam de hele cultuur over naar de papieren zakken door de ruimte die overblijft door de afgesneden hoek. Label elke papieren zak, vouw het bovenste uiteinde van elke zak twee keer en sluit met nietjes om een driehoekige tentstructuur te creëren.Plaats de zakken op draaddroogrekken om goed te kunnen drogen, zelfs drogen bij een gecontroleerde temperatuur van ongeveer 25 graden Celsius en een lage luchtvochtigheid van 30 tot 40% Oogst schimmelconidia uit de culturen met behulp van drie geneste zeven gemonteerd op een verzamelpan. Laad het droge cultuurmonster langzaam op het ETS-gaas nummer 35 en zeef. Plaats een deksel op de zeef om te voorkomen dat schimmel conidia in de lucht vrijkomt.Voeg 10 tot 12 glazen knikkers toe aan de zeven om de doorgang van de schimmelconidia door de gaasschermen te helpen en het vasthouden van de conidia in de zeef te voorkomen, wat kan resulteren in verminderd sporenherstel. Tape en sluit de zeefverbindingen af met elektrische tape. Plaats de zeven op een trilschudder voorzien van een kleverig kussen om de opvangpan en zeven gedurende 20 tot 25 minuten vast te zetten met een beweging van 560 tot 640 trillingen per minuut.Haal de testzeven uit de verzamelpan, verzamel conidia en bewaar de verzamelde conidia in luchtdichte en waterdoordringbare zakken met ritssluiting voor langdurige opslag. Gemiddeld werd ongeveer 1,83 gram droge Metarhizium robertsii conidia geoogst uit het afgevlokken gerstsubstraat, terwijl nul Metarhizium pinghaense uit het substraat werd geoogst. Er werden geen droge schimmelconidia geoogst uit het afgebladderde haversubstraat voor beide Metarhizium-soorten.Een significant verschil werd waargenomen tussen de twee soorten Metarhizium in de geschatte gemiddelde conidia per gram geoogst uit de rijstkorrels. Metarhizium robertsii had een iets hoger gemiddeld aantal conidia per gram dan Metarhizium pinghaense. Er werd geen significant verschil waargenomen tussen de twee soorten Metarhizium in het geschatte aantal conidia dat aanwezig is op het afgewerkte rijstsubstraat.Metarhizium pinghaense had echter iets hogere conidia op het rijstsubstraat dan Metarhizium robertsii Er wordt geen significant verschil geschat in de twee soorten Metarhizium in hun totale conidia-opbrengst verkregen uit de rijstsubstraatculturen. Metarhizium pinghaense produceerde echter een iets hogere conidia-opbrengst dan Metarhizium robertsii, die een lagere conidiale opbrengst produceerde. Het uitdagende deel van deze techniek is de substraatverontreiniging door inenting met een verontreinigd blastospores-entmateriaal.Zorg dus altijd voor steriliteit tijdens het werken. Deze demonstratie helpt de studenten de techniek efficiënt te reproduceren zonder onhandige fouten die kunnen resulteren in het einde van het hele proces.
