Den vellykkede integrasjonen av mikrobielle midler for biologisk kontroll av viktige landbruksinsekt i et agroecosystem avhenger av både suksess og enkel masseproduksjon av entomopathogenene som det første trinnet under laboratorieforhold. Masseproduksjonen av EPF er viktig for den store applikasjonen og tilgjengeligheten av EPF-produkter for IPM-programmer ved hjelp av biologisk kontroll 9,10,11,12,13.<sup class="x...

Entomopathogene sopp (EPF) har fått betydning som avlingsbeskyttelsesmidler i den biologiske kontrollen av viktige landbruksinsekt 1,2. Entomopathogenene, som forekommer naturlig i jord, forårsaker epizootikk i befolkningen i ulike skadedyrsarter3. Arten av EPF er vertsspesifikk og utgjør relativt få risikoer når det gjelder å angripe ikke-målrettede arter, og de er ikke-giftige for miljøet4. EPF har en unik mekanisme for å invadere verten, samt for å forplante og vedvare i deres umiddelbare miljø1. De angriper verten hovedsakelig gjennom aseksuelle sporer som fester seg til og trenger inn i verts cuticle for å invadere og spre seg i verts hemocoel. Verten dør til slutt på grunn av uttømming av hemolymfe næringsstoffer eller som følge av toksemi forårsaket av giftige metabolitter frigjort av soppen. Etter døden, under ideelle miljøforhold, kommer soppen ut på den ytre overflaten (overt mykose) av vertskadaveren 5,6.
Økende bekymringer for de negative effektene av kjemiske rester på menneskers helse, miljøforurensning og utvikling av skadedyrsresistens har ført til den globale drivkraften for å redusere innganger av kjemisk baserte insektmidler og for å finne alternative, nye og bærekraftige strategier for avlingsbeskyttelse og skadedyrsbekjempelse 6,7,8 . Dette har gitt muligheter til å utvikle mikrobielle baserte insektmidler til bruk i Integrert Pest Management (IPM) programmer, som er mer økologisk gunstige strategier enn konvensjonell kjemisk kontroll 3,8.
For å utvikle et vellykket mikrobielt kontrollmiddel for et landbruksskade, må en passende organisme først isoleres, karakteriseres, identifiseres og dens patogenitet for målskade bekreftet. En enkel, kostnadseffektiv metode for storskala produksjon av mikrobielle midler er imidlertid nødvendig for å produsere et levedyktig produkt for bruk i biologiske kontrollprogrammer 9,10,11,12,13. Masseproduksjon av betydelige mengder entomopathogener av god kvalitet avhenger av mikrobiell belastning, miljø, mål, formulering, markedet, applikasjonsstrategien og ønsket sluttprodukt 14,15,16. EPF kan masseproduseres ved hjelp av flytende substratgjæring for å produsere blastosporer eller den faste substratgjæringsprosessen for å produsere luftkonidia 6,17,18. Imidlertid påvirker masseproduksjons- og formuleringsprosessen av entomopathogener direkte virulens, kostnaden, holdbarheten og felteffekten av sluttproduktet. For vellykket bruk i IPM må produksjonsprosessen til entomopathogenene være enkel å kjøre, kreve minimal arbeidskraft, produsere en høy avkastning konsentrasjon av virulente, levedyktige og vedvarende propagules, og være lav i kostnad 4,13,14,16.
Å forstå ernæringskravene til entomopathogener er viktig for massedyrking med alle dyrkingsmetoder 4,12. Næringskomponentene i produksjonsmediet har en betydelig innvirkning på egenskapene til de resulterende propagulene, inkludert biokontrolleffekt, utbytte, uttørkingstoleranse og utholdenhet 8,19,20,21. Optimalisering av produksjonsprosedyrer er utformet for å adressere slike faktorer22. For EPF er de viktigste kravene til god vekst, sporulering og masseproduksjon av soppfusi tilstrekkelig fuktighet, optimal veksttemperatur, pH, gassutveksling av CO2 og O2, og ernæring, inkludert gode fosfor-, karbohydrat-, karbon- og nitrogenkilder18.
Jaronski og Jackson18 beskriver den faste substratgjæringsmetoden som den mest effektive og nærmeste tilnærmingsmetoden til den naturlige prosessen for EPF-produksjon i forhold til den flytende substratgjæringsmetoden fordi, under naturlige forhold, bæres soppkonidiumet på faste oppreiste strukturer, som overflaten av insektkadaver. Landbruksprodukter og biprodukter som inneholder stivelse brukes for det meste til masseproduksjon av hypokreolske sopp, da soppene lett dekomponerer stivelse gjennom sekresjon av høyt konsentrerte hydrolytiske enzymer fra hyfaltspissene, for å trenge inn i det faste stoffet, og for å få tilgang til næringsstoffene som er tilstede i stoffet 11,17,18,23 . Kornproduktene stiller også krav til sunn biomasseproduksjon fordi substratene kan absorbere ytterligere næringsstoffer fra ethvert flytende medium 16,18,24 når de er hydrert og sterilisert.
Tidligere forsøkte flere studier å massekulturere EPF-arter som Beauveria bassiana (Bals.) Vuil., Cordyceps fumosorosea (Wize) Kelper B. Shrestha &Spatafora, Verticillium lecanii (Zimm.) Viegas og noen av Metarhizium anisopliae (Metschn.) Sorokin artskompleks isolerer på ulike substrater 16,23,24. Slike masseproduserte og kommersielt utviklede isolasjoner inkluderer Green Muscle® (stamme IMI 330189), utviklet fra M. anisopliae var Metarhizium acridum (Driver &Milner) J.F. Bisch, Rehner &Humber, Metarhizium 69 (Meta 69 stamme ICIPE69) og Real Metarhizium 69 (L9281), utviklet fra M. anisopliae, og Bredbånd® (stamme PPRI 5339) og Eco-Bb®, utviklet fra B. bassiana25,26 . Det er imidlertid gjort begrensede forsøk på massekulturen Metarhizium robertsii J.F. Bisch., S.A. Rehner &Humber og Metarhizium pinghaense Chen &Guo. Disse to isolasjonene ble valgt ut i en tidligere studie som den mest effektive for kontroll av melkebollen, Pseudococcus viburni Signoret (Hemiptera: Pseudococcidae)27. mass production of entomopathogenic fungi, metarhizium robertsii and metarhizium pinghaense, for commercial application against insect pests

Entomopathogene sopp av Metarhizium anisopliae artskomplekset har fått betydning som de biologiske kontrollmidlene til landbruksinsekt. Økningen i skadedyrsmotstand mot kjemiske insektmidler, de økende bekymringene for de negative effektene av insektmidler på menneskers helse, og miljøforurensningen fra plantevernmidler har ført til et globalt pådriv for å finne nye bærekraftige strategier for avlingsbeskyttelse og skadedyrsbekjempelse. Tre landbrukskornprodukter, flakede havre, flaket bygg og ris ble brukt som EPF solid gjæringssubstrater. To inokulasjonsmetoder, konidiale suspensjoner og den flytende soppkulturen av blastosporer ble brukt til å inokulere de faste substratene. Inokulering ved bruk av konidiale suspensjoner ble observert å være relativt mindre effektiv, da økte nivåer av forurensning ble observert på de faste substratene i forhold til ved bruk av blastosporinokuleringsmetoden. Flaked havre ble funnet ikke å være et passende vekstsubstrat for både M. robertsii og M. pinghaense, da ingen tørr conidia ble høstet fra substratet. Flaked bygg ble funnet å favorisere produksjonen av M. robertsii conidia over M. pinghaense, og i gjennomsnitt 1,83 g ± 1,47 g tørr M. robertsii conidia og null gram M. pinghaense conidia ble høstet fra substratet. Riskorn ble funnet å favorisere den konidiale masseproduksjonen av både M. pinghaense og M. robertsii isolerer, med et gjennomsnitt på henholdsvis 8,2 g ± 4,38 g og 6 g ± 2 g høstet fra substratet.

En nedgang i innholdsmassen til kulturene på ris for både M. pinghaense og M. robertsii ble observert over tid under tørkestadiet av soppkulturene, uten at det ble observert noen eller lite endring i massen når kulturene var tørre (figur 5). Mass Production of Entomopathogenic Fungi, Metarhizium robertsii and Metarhizium pinghaense, for Commercial Application Against Insect Pests

Entomopathogene sopp har fått betydning som de biologiske kontrollmidlene til landbruksinsekt. Denne metoden gir ny informasjon om hvordan man masseproduserer infektiv conidia av entomopathogene sopp for å kontrollere viktige i kommersielle agroøkosystemer, gitt den nåværende situasjonen der de fleste insektmidler fases ut på grunn av bekymringer om deres giftige effekt på miljøet og menneskers helse. Teknikken beskriver metoden som brukes for å sikre optimal utbytte av EPF-conidia under masseproduksjon. Metoden er nyttig for integrert skadedyrshåndtering og biologisk kontroll ved å gi innsikt i effektiviteten av masseproduserte conidia for storskala styring av skadelig insektmiddelresistens insekt i agro-økosystemene.Denne prosedyren ble utviklet og demonstrert av Dr.Letodi Luki Mathulwe, Dr.Nomakholwa Faith Stokwe og professor Antoinette Paula Malan. Varm opp en liter destillert vann og slå av varmen før du når kokepunktet. Tilsett nå 30 gram glukose, fire gram kaliumfosfatdibasic, 20 gram gjærekstrakt, 15 milliliter mais bratt brennevin, og kok innholdet i to til tre minutter.Hell totalt 100 milliliter av mediet i ni forskjellige 250 milliliterflasker. Legg en bomullsullplugg på hver kolbe og dekk bomullsullen med aluminiumsfolie som en stopper. Autoklaver mediet i kolber i 55 minutter ved 121 grader Celsius.Senere, la mediet i kolber avkjøles og tilsett 10 milligram per milliliter streptomycin til mediet i hver kolbe. Samle to til tre bakterielle løkker av sopp conidia fra to til tre uker gamle soppkulturplater for begge EPF-isolasjonene. Overfør soppkontrofien til hver 100 milliliter flytende medier i de 250 milliliterflaskene under sterile forhold og forsegle kolbeene.Inkuber væskekulturflaskene ved ca. 25 grader Celsius på en orbital shaker ved 140 RPM i tre dager og stopp inkubasjonen når kulturene viser tegn på høy turbiditet med soppblåsingsvekst. For å oppdage mulig bakteriell forurensning fra kulturene, tegn en 100-mikroliters prøve fra hver flytende kulturflaske etter 24 timers inkubasjon og tallerken på tre SDA-plater per isolat. Inkuber platene i 48 timer ved en kontrollert temperatur på pluss eller minus 25 grader Celsius.Bruk et lite polyvinylkloridavfallsrør for å lage en hals for gjæringsposen i den åpne enden av autoklavposen og bruk autoklavbånd for å feste røret. Lukk røret med en steril bomullsullplugg for å tillate tilstrekkelig gassutveksling under gjæring. Bruk parboiled langkornet hvit ris som et solidt substrat medium for blastospores av både Metarhizium pinghaense og Metarhizium robertsii.For hver gjæringspose, tilsett ett kilo ris og 300 milliliter sterilt destillert vann. Bland forsiktig innholdet i gjæringsposene. Plasser dem i ytre autoklavposer i oppreist stilling og autoklaver ved 121 grader Celsius i 55 minutter.Etter autoklaven, la substratene avkjøles i ca. 45 minutter under sterile forhold. Fjern lukkingen av hver av de flytende kulturflaskene i både Metarhizium pinghaense og Metarhizium robertsii under en laminær strømning og flamme kanten av hver kolbe i 10 sekunder. Hell 100 milliliter flytende kulturer i gjæringsposene ved å fjerne bomullsullpluggene fra nakken.Sett bomullspluggene på igjen og dekk toppen av posens hals med kirurgisk papir festet med et gummibånd. Vri den øverste delen av posen og bland posens innhold ved å riste og manipulere substratet ved massasje. Inkuber posene ved ca 25 grader Celsius ved å flate substratet i posene for å forhindre dannelse av tykke senger.To til tre dager etter inokulering og inkubasjon, når synlig mycelial vekst og bindingen av substratet av soppen hadde skjedd, bryte substratet i gjæringsposene ved å massere posenes innhold. Etter gjæring, overfør de sporulerte kulturene til 26-til-30 kilo brune papirposer og tørk soppkulturene i 10 til 12 dager før bruk i forsøk. For å forbedre strekkfastheten til papirposene, kutt av 1/3 av den øverste delen av posen og line bunnen av posen.Smuldre substratet forsiktig i hver gjæringspose. Klipp av hjørnet av hver pose og overfør sakte hele kulturen til papirposene gjennom plassen som er igjen av det avskårne hjørnet. Merk hver papirpose, brett over den øverste enden av hver pose to ganger, og lukk med stifter for å skape en trekantet teltstruktur.Legg posene på trådtørkingsstativer for å tillate riktig, til og med tørking ved en kontrollert temperatur på ca. 25 grader Celsius og lav luftfuktighet på 30 til 40% Høst soppkontrofier fra kulturene ved hjelp av tre nestede sikter montert på en oppsamlingspanne. Last langsomt inn tørrkulturprøven på ETS mesh nummer 35 og sikt. Sett et lokk på silen for å forhindre frigjøring av sopp conidia i luften.Tilsett 10 til 12 glasskuler til siktene for å hjelpe passasjen av soppkonidiaen gjennom maskeskjermene og unngå oppbevaring av conidia i sikten, noe som kan føre til redusert sporegjenoppretting. Tape og forsegle silleddene ved hjelp av elektrisk tape. Plasser silene på en vibratorisk shaker utstyrt med en klebrig pute for å feste oppsamlingspannen og siktene i 20 til 25 minutter med en bevegelse på 560 til 640 vibrasjoner per minutt.Fjern testsiktene fra oppsamlingspannen, samle conidia, og oppbevar de oppsamlede kondiviene i lufttette og vanngjennomtrengelige glidelåslåsposer for langvarig lagring. Et gjennomsnitt på ca. 1,83 gram tørr Metarhizium robertsii conidia ble høstet fra det flakede byggsubstratet, mens null Metarhizium pinghaense ble høstet fra substratet. Ingen tørr sopp conidia ble høstet fra det flakede havre substratet for noen av Metarhizium-artene.En signifikant forskjell ble observert mellom de to artene av Metarhizium i estimert gjennomsnittlig conidia per gram høstet fra riskornene.
