Ces protocoles servent de lignes directrices de meilleures pratiques pour la génération de poudre osseuse à utiliser dans l’extraction d’ADN en aval à travers une gamme d’éléments squelettiques. Il existe quelques anciens protocoles d’échantillonnage d’ADN publiés pour des endroits autres que la dentine et la pyramide pétreuse. Aujourd’hui, nous allons montrer les méthodes d’échantillonnage détaillées pour trois alternatives.
Ces méthodes montrent également un grand potentiel d’utilisation avec des restes médico-légaux dégradés, mais aussi comme base de référence pour le développement de techniques similaires avec des spécimens non humains. Effectuez tous les prélèvements dans une salle blanche dédiée, sous une hotte PCR équipée de lumière UV ou une armoire de biosécurité. Coupez le flux d’air et étalez du papier d’aluminium stérile sur le dessus de la paillasse pour attraper toute poudre ou fragment d’os égaré.
Placez une feuille de papier de pesée dans un plateau de pesée stérile. Tenez la molaire décontaminée par l’émail avec la racine vers le bas au-dessus d’un plateau de pesée, à l’aide d’une pince à main. Équipez une perceuse dentaire d’une roue de coupe circulaire à bords diamantés.
Lorsque la perceuse est réglée à une vitesse moyenne, touchez légèrement le bord de la perceuse à la racine à un angle d’environ 20 degrés. Grattez l’échantillon vers le bas dans le plateau pour recueillir le matériau jaune le plus externe de la racine. Arrêtez la collecte une fois que le matériau plus léger de la dentine devient visible.
Transférer la poudre dans un tube LoBind Safe-Lock de deux millilitres et enregistrer cette masse de poudre recueillie à l’aide d’une balance fermée avec une précision d’au moins 0,01 milligramme. Transférer la poudre du papier de pesage dans un tube LoBind Safe-Lock de deux millilitres pour l’extraction. Conservez ce tube à 20 degrés Celsius.
Assurez-vous de récupérer tous les fragments d’os et autant de poudre que possible. Changez la feuille entre chaque échantillonnage, en jetant la feuille usagée dans un sac autoclavable pour risque biologique. Décontaminez la zone de travail et étalez du papier d’aluminium stérile sur le dessus de paillasse à chaque fois avant de passer à l’os suivant.
Placez une petite feuille de papier de pesée dans un plateau de pesée standard. Retirez et jetez la couche la plus externe de l’os cortical de l’arc vertébral supérieur à l’aide d’une perceuse dentaire. Ensuite, fixez les vertèbres dans une pince à main, avec le processus épineux vers l’extérieur et l’aspect supérieur vers le bas.
Tout en tenant la vertèbre, percez vers le haut au centre de l’encoche en forme de V, formée par la fusion de l’apophyse épineuse avec les lamelles à basse vitesse et à couple élevé. Cesser de forer lorsqu’il y a une baisse notable de la résistance. Changez légèrement la position de forage et répétez jusqu’à ce que 50 à 100 milligrammes de poudre d’os soient recueillis.
Transférez la poudre d’os du papier de pesée dans un tube LoBind de deux millilitres pour l’extraction et conservez-la à 20 degrés Celsius. Assurez-vous que tous les fragments d’os et la plus grande quantité possible de poudre sont récupérés avant de jeter une feuille d’aluminium. Décontaminer la zone de travail et préparer le prélèvement d’échantillons, comme décrit précédemment.
Placez une petite feuille de papier de pesée dans un plateau de pesée standard. Tenez le dôme du talus vers le haut et la surface médiale vers le collecteur, au-dessus du plateau de pesée. Grattez l’os cortical à une profondeur d’environ un millimètre du col du talus à l’aide d’une perceuse dentaire avec un taillant de faible calibre réglé à basse vitesse et à couple élevé.
Changez la position de forage et répétez cette opération jusqu’à ce qu’environ 50 à 100 milligrammes de poudre d’os soient recueillis. Transférer cette poudre d’os du papier de pesage dans un tube LoBind de deux millilitres pour l’extraction. Conservez la poudre à 20 degrés Celsius indéfiniment.
Conservez le reste de l’os et l’excès de poudre dans un environnement stérile sec et à température contrôlée à 25 degrés Celsius. Jeter tous les déchets dans des sacs ou des récipients pour animaux à risque biologique en autoclave. Stériliser ou décontaminer tout l’équipement réutilisable et l’exposer aux UV entre chaque échantillonnage.
Le Pars petrosa a produit un ADN endogène plus élevé que les 23 autres sites d’échantillonnage anatomique étudiés. Les sept autres emplacements d’échantillonnage anatomique présentés dans ce protocole ont produit les rendements les plus élevés suivants. Les emplacements alternatifs ont systématiquement produit des rendements d’ADN adéquats pour les analyses génétiques standard des populations.
Les taux de duplication dans les bibliothèques provenant de tous les sites d’échantillonnage anatomique étaient faibles, ce qui indique une grande complexité. Les dents, ainsi que les vertèbres thoraciques ont montré une récupération moléculaire adéquate. L’un des aspects les plus difficiles de l’échantillonnage osseux est le processus de forage, comme la façon de tenir une perceuse, où forer et quand arrêter le forage.
