脂質二重層コンパートメントは、密閉された有機反応および膜ベースのプロセスを研究するためのモデル合成細胞として、または薬物送達用途におけるキャリアモジュールとして使用される1,2。精製された成分を用いたボトムアップ生物学は、タンパク質や脂質などの生体分子間の特性や相互作用を探求するために最小限の実験システムを必要とする3,4。しかし、この分野の進歩に伴い、生体細胞の条件をよりよく模倣するより複雑な実験システムの必要性が高まっています。GUVsへのカプセル化は、変形可能で選択的に透過性の脂質二重層および閉じ込められた反応空間を提供することによって、これらの細胞様特性のいくつかを提供することができる実用的なアプローチである。特に、細胞骨格系のインビトロ再構成は、合成細胞のモデルとして、膜区画5への封入から利益を得ることができる。多くの細胞骨格タンパク質は細胞膜と結合し、相互作用する。ほとんどの細胞骨格集合体は細胞全体にまたがる構造を形成するので、それらの形状は細胞サイズの閉じ込め6によって自然に決定される。
