Este método adquiere rebanadas vivas del tronco cerebral auditivo del pollo que abarca un gradiente más grande de región de frecuencia en la misma rebanada. Estas rodajas son adecuadas para experimentos electrofisiológicos y anatómicos con un eje tonotópico más grande. Esta metodología ayudará a comprender el desarrollo de microcircuitos en el tronco cerebral auditivo.
Mientras que la anatomía es específica del pollo, otras especies de aves como las codornices y los pájaros cantores tienen una anatomía similar. Para comenzar, esterilice los huevos con etanol al 70% e incubarlos a 38 grados centígrados y 50% de humedad. A continuación, establezca la tasa de burbujeo para el ACSF tal que el pH sea de 7.2 a 7.4 con una osmolalidad entre 300 y 310 miliosmolares por litro.
Luego mezcle cinco gramos de agarosa en 100 mililitros de dACSF. Disuelva la agarosa completamente usando un baño de agua o microondas hasta que la agarosa comience a burbujear. Vierta la agarosa derretida en una placa de Petri hasta un grosor de cinco milímetros y manténgala a temperatura ambiente para que se fije.
Después de cuajar, selle la placa de Petri y guárdela a cuatro grados centígrados. Corte la agarosa en bloques de cubículos para usarla en el momento de la disección. Primero, limpie el área de disección con etanol al 70%.
Pegue el bloque de agarosa de soporte o en ángulo en la bandeja de vibratomo. Coloque el huevo bajo una luz brillante y ubique el espacio lleno de aire en el lado más grande o más redondo del huevo, luego rompa la cáscara sobre el espacio lleno de aire y exponga el saco de membrana. Haga una incisión suave en el saco para exponer el pico.
Con un bisturí, tire suavemente del cuello y la cabeza fuera del huevo. Después de la decapitación, limpie el cabezal con dACSF refrigerado. Mantenga la cabeza firme en dACSF frío y haga una incisión rostrocaudal.
Comience la incisión detrás y entre los ojos y siga la longitud del cuello cosechado. Separe la piel para exponer el cráneo. Corte el cráneo detrás del ojo en dirección media a lateral.
Repita para ambos hemisferios. Corte la porción rostral del cráneo colocando la cuchilla detrás de los ojos y haciendo un corte rápido. Luego sumerja la cabeza en un plato de dACSF frío.
Usando un par de tijeras pequeñas, haga incisiones de línea media a lateral en la región caudal del cráneo para separar el cerebro sin causar daño tisular. Exponga suavemente el tronco encefálico y el cerebelo. Retraiga el área dorsal de todo el cráneo.
Retire el tronco encefálico y expóngalo con un cepillado suave con un pincel fino. Use fórceps curvos para limpiar el tronco encefálico de la conexión de tejidos y vasos sanguíneos. Separe el tronco encefálico del cerebelo cortando los pedúnculos y retirando los vasos sanguíneos con cuidado.
Recorte el tronco encefálico de vasos sanguíneos adicionales. Primero, coloque la hoja de vibratomo a lo largo del eje horizontal. Para cortes coronales, pegue el tronco encefálico en una bandeja de corte por el lado rostral, manteniendo el eje rostrocaudal vertical.
Para cortes sagitales, mantenga el eje medial lateral vertical. Para rodajas horizontales, pegue el lado ventral manteniendo el eje ventral dorsal vertical. Para lograr el plano horizontal sagital angular agudo, pegue el lado ventral del tronco encefálico de tal manera que el eje dorsal ventral sea vertical en el bloque de agarosa de la superficie de hipotenusa.
Pegue la superficie opuesta del bloque de agarosa frente a la bandeja de corte y mantenga el eje rostrocaudal paralelo al borde de la cuchilla. Como alternativa al corte de vibratomo, sumerja el tronco encefálico aislado en una agarosa de bajo punto de fusión al 4% a 40 grados centígrados en una placa de Petri de 35 por 10 milímetros. Después de que la agarosa se solidifique, corte el bloque de agarosa del cubículo con una cuchilla de afeitar afilada.
Pegue el bloque de agarosa en su lado rostral manteniendo vertical el eje rostrocaudal del tronco encefálico. Tome cortes coronales hasta que se vea la región NM. Retire el bloque de agarosa del pegamento con una cuchilla afilada.
Con una aguja fina, coloque suavemente 0,5 microlitros de colorante azul de toluidina en el NM para detectar los núcleos. Vuelva a montar este bloque en la bandeja de corte para rodajas sagitales u horizontales. Identificar los núcleos con respecto a la región teñida y seccionar el tronco encefálico.
Después de seccionar, coloque las rodajas recolectadas secuencialmente de 200 a 300 micras en una cámara cortada que contenga ACSF para equilibrarse durante una hora a temperatura ambiente. Las secciones coronales de tejido del tronco encefálico de un embrión de pollo E19 a 21 representan las regiones de isofrecuencia relativa de los núcleos auditivos del tronco encefálico desde la región auditiva de menor a mayor frecuencia característica. Las fibras nerviosas auditivas aferentes y la bifurcación de los axones NM fueron visibles.
También se observó Nucleus angularis. En las secciones rostrales, el núcleo ovárico superior se encuentra a lo largo de la región lateral ventral del corte coronal. En las secciones sagitales, se identificaron NM y núcleo laminaris o NL donde las fibras nerviosas auditivas entraron en el grupo de neuronas.
El núcleo ovárico superior se identificó en la región rostrolateral. Se observaron regiones auditivas de baja y alta frecuencia característica de NM y NL a lo largo del eje rostrocaudal. El corte más medial mostró los mechones axonales ipsilateral y contralateral y el punto final de la región auditiva.
En los cortes horizontales, NM y NL se identificaron hacia la línea media, y las neuronas se extendieron a lo largo del eje medial lateral. Los cortes del tejido del tronco encefálico en un ángulo agudo desde un plano horizontal mostraron los núcleos auditivos del tronco encefálico a través de una gran extensión diagonal. Las imágenes ampliadas mostraron el eje tonotópico de los núcleos auditivos a lo largo del eje rostromedial a caudolateral.
Asegúrese de que todo el proceso se realice en dACSF refrigerado con hielo burbujeado continuamente con oxígeno de carbohidratos. Este protocolo se utiliza para investigadores que investigan las propiedades anatómicas y biofísicas del desarrollo de microcircuitos del tronco encefálico, específicamente la relación entre los principales núcleos auditivos.
