SECは、サイズに基づいて流体の成分を分離する方法です。この技術では、樹脂のカラム(社内で製造されたもの、または市販品で購入されたもの)を使用してサンプルを分画します。サンプル中の小さな粒子は樹脂内のビーズの間に閉じ込められますが、大きな粒子は樹脂をより自由に通過できるため、プロセスの早い段階で溶出します。EVは多くの細胞外タンパク質やタンパク質凝集体よりもサイズが大きいため、EVは細胞外タンパク質よりも速くカラムを通過し、早い画分で溶出します14。
