C. elegansの感染に関するプロトコルは、現在の研究に記載されており、ユニークな微胞子虫生物学を明らかにし、感染に対する宿主の応答を解剖するために使用することができる。キチン結合色素Direct Yellow 96(DY96)で染色された固定ワームの顕微鏡観察は、キチン含有微胞子虫胞子の腸全体に感染が広がることを示している。DY96染色はまた、宿主の適応度の読み出しとしてワームの重力(胚を産生する能力)の同時評価のために、キチン含有ワーム胚の視覚化を可能にする。
最近の研究により、N. parisiiに感染したC. elegansは、同じ感染に対して頑丈に耐性のある子孫を産むことが明らかになりました19。この遺伝性免疫は単一世代持続し、より重度に感染した両親からの子孫が微胞子虫に対してより耐性であるため、用量依存的である。興味深いことに、N. parisii-primedの子孫はまた、細菌の腸内病原体緑膿菌に対してより耐性があるが、天然の病原体オルセーウイルス19に対して保護されていない。本研究はまた、免疫プライムされた子孫が微胞子虫による宿主細胞の侵入を制限することを示している。この方法はまた、免疫プライミングされた子孫の収集と、FISHを使用して腸細胞中のN. parisii RNAを検出し、宿主細胞の侵入および胞子の発火をアッセイする方法についても説明しています20。
