폐 혈관 구조는 저압 혈관 시스템으로, 주요 기능은 산소가 제거 된 혈액을 폐의 가스 교환 영역으로 전달하는 것입니다. 폐의 폐동맥은 기관지 나무와 평행 한 가지로 배열되어 궁극적으로 여러 폐포에 걸쳐 연속적이고 마지막으로 정맥과 정맥으로 모이는 광범위한 모세 혈관 네트워크를 형성합니다. 폐동맥의 혈관 색조는 내피와 혈관 평활근 세포 (VSMC) 사이의 상호 작용을 포함하는 여러 요인에 의해 제어됩니다1.

VSMC의 프로스타사이클린 수용체(IP)와 결합하여 아데닐릴 사이클라제(AC) 효소를 자극한 다음 아데노신 삼인산(ATP)을 고리형 아데노신 모노포스페이트(cAMP)로 전환합니다(그림 1)3,4. cAMP는 단백질 키나아제 A (PKA)를 활성화시켜 세포질Ca 2+ 수준을 감소시키고 혈관 이완5을 유발합니다 (그림 1). EDHF 경로는 또한 다양한 내피 매개체 및 전기적 사건을 통해 내피 의존성 혈관 이완에 참여합니다. EDHF 경로의 활성화는 VSMC의 과분극을 유도하여 전압 작동 Ca 2+ 채널 (VOCC)을 폐쇄하고 세포 내Ca 2+ 수준을 감소시키고 혈관 이완을 유도합니다6. 내피 비 의존성 혈관 이완은 세포 내Ca2+ 수준의 감소, 미오신 경쇄 키나아제 (MLCK)의 억제, 미오신 경쇄 포스파타제 (MLCP)의 활성화 및 VSMC의 수축 기계에 대한Ca 2+ 민감도의 감소와 같은 여러 메커니즘을 통해 VSMC에서 직접 발생합니다. 이 연구에서는 다양한 K+ 채널의 개방, VOCC의 봉쇄 및 근형질 세망7에서 Ca2+ 방출 억제로 인한 혈관 이완에 중점을 두어 세포 내Ca2+ 수준을 감소시켜 VSMC 미오신 경쇄 인산화 및 미오신 - 액틴 결합 또는 교차 다리 형성을 각각 감소시키고, 궁극적으로 혈관 이완을 초래합니다.
분리 된 IPA에서 혈관 수축 및 혈관 이완 측정을 평가하는 기술은 설치류에 대해 잘 확립되어 있지만 데이터는 실험 프로토콜에 따라 다릅니다. 본 연구는 신경 신호, 호르몬, 사이토 카인, 혈압 등과 같은 생체 내 혈관 반응을 조절하는 외부 요인이없는 상태에서 만들어진 시험관 내 래트 IPA 제제의 혈관 반응성을 평가하는 데 사용되는 방법을 설명합니다.
우리는 IPA의 혈관 반응성을 연구하기 위한 예로서 식물 추출물을 사용하는 여러 실험 프로토콜을 사용했습니다. 다양한 차단제 (그림 1)를 사용하여 식물 추출물에 의해 유도 된 내피 의존성 및 비 의존성 혈관 이완 메커니즘을 확인했습니다. 그럼에도 불구하고, 다양한 폐 병리의 치료에 사용되는 약물, 추출물 또는 식물 화학 물질에 대한 IPA의 혈관 반응을 평가하기 위해 동일한 프로토콜을 적용 할 수 있습니다.
