Bacteriële motiliteit speelt een cruciale rol bij bacteriële pathogeniciteit, biofilmvorming en medicijnresistentie1. Bacteriële motiliteit is nauw verbonden met pathogeniciteit en is noodzakelijk voor bacteriële kolonisatie tijdens vroege infectie van gastheercellen2. Biofilmvorming is nauw verwant aan bacteriële motiliteit, waarbij bacteriën zich door beweeglijkheid hechten aan het oppervlak van vaste media. Bacteriële motiliteit wordt al lang beschouwd als positief gecorreleerd met biofilmvorming. Een hoge mate van bacteriële resistentie tegen geneesmiddelen als gevolg van biofilm kan leiden tot aanhoudende infecties die een bedreiging vormen voor de menselijke gezondheid 3,4,5. Daarom is het belangrijk om bacteriële beweeglijkheid te detecteren. De bacteriële motiliteitstest wordt voornamelijk gebruikt om de beweeglijkheid van verschillende vormen van bacteriën in de levende toestand te onderzoeken, die indirect de aan- of afwezigheid van flagellen kunnen bepalen en dus een belangrijke rol spelen bij de identificatie van bacteriën.
Er zijn directe en indirecte methoden om bacteriële motiliteitte detecteren 6. Omdat bacteriën met flagella beweeglijkheid vertonen, is het mogelijk om te detecteren of bacteriën indirect beweeglijk zijn door de aan- of afwezigheid van flagella te detecteren. Het is bijvoorbeeld mogelijk om beweeglijkheid indirect te detecteren door elektronenmicroscopie en flagellaire kleuring om aan te geven dat bacteriën beweeglijk zijn. Het is ook mogelijk om te detecteren met directe methoden, zoals suspensieval en semisolid punctiemethoden. 
De semisolid punctiemethode die vaak wordt gebruikt in niet-gegradueerde microbiologische laboratoria om bacteriële motiliteit te detecteren, ent de bacteriën in de punctie in het semisolid agar-medium met 0,4-0,8% agar, afhankelijk van de richting van bacteriële groei. Als de bacteriën langs de punctielijn groeien om zich te verspreiden, verschijnen er wolkachtige (borstelachtige) groeisporen, wat wijst op de aanwezigheid van flagella en dus beweeglijkheid. Als er geen groeisporen van de priklijn zijn, is de bacterie niet gevlagd of beweeglijk. 
Deze methode heeft echter zijn nadelen: de bacteriën zijn kleurloos en transparant, de flagellaire activiteit wordt beïnvloed door de fysiologische kenmerken van de levende bacteriën en andere factoren, en de concentratie van agar en de kleine diameter van de reageerbuis. Bovendien zijn aërobe bacteriën alleen geschikt voor groei op het agaroppervlak, wat de observatie van bacteriële beweeglijkheid beïnvloedt. Om dit experiment te verbeteren, werd daarom 2,3,5-trifenyltetrazoliumchloride (TTC) (kleurloos) aan het medium toegevoegd om een betrouwbaardere en intuïtievere methode voor het bepalen van bacteriële motiliteit vast te stellen dan de huidige directe punctiemethode met behulp van intracellulaire dehydrogenasen om de vorming van een rood product van TTC 7,8,9,10 te katalyseren.

Bacteriële motiliteit is cruciaal voor bacteriële pathogeniciteit, biofilmvorming en medicijnresistentie. Bacteriële beweeglijkheid is cruciaal voor de invasie en/of verspreiding van vele pathogene soorten. Daarom is het belangrijk om bacteriële beweeglijkheid te detecteren. Bacteriële groeiomstandigheden, zoals zuurstof, pH en temperatuur, kunnen de bacteriegroei en de expressie van bacteriële flagellen beïnvloeden. Dit kan leiden tot verminderde beweeglijkheid of zelfs verlies van motiliteit, wat resulteert in de onnauwkeurige evaluatie van bacteriële motiliteit. Op basis van de kleurreactie van 2,3,5-trifenyltetrazoliumchloride (TTC) door intracellulaire dehydrogenasen van levende bacteriën, werd TTC toegevoegd aan traditionele semisolid agar voor bacteriële motiliteitsdetectie. De resultaten toonden aan dat deze TTC semisolid agar-methode voor de detectie van bacteriële motiliteit eenvoudig is, gemakkelijk te bedienen en geen grote en dure instrumenten omvat. De resultaten toonden ook aan dat de hoogste beweeglijkheid werd waargenomen in halfvast medium bereid met 0,3% agar. In vergelijking met het traditionele semisolid medium zijn de resultaten gemakkelijker te evalueren en nauwkeuriger.

Zowel standaardstammen als geïsoleerde stammen werden vergeleken voor motiliteitsdetectie en de resultaten zijn weergegeven in tabel 1. Door de afwezigheid van flagella groeiden Staphylococcus aureus en Klebsiella pneumoniae alleen langs de geënte lijn op zowel traditionele als TTC semisolid media. Detection of bacterial motility is important because motility is crucial for pathogenicity. The protocol described here is a reliable method for the detection of bacterial motility, because it is based on the color reaction of TTC with intracellular dehydrogenases of living bacteria. The TTC semi-solid agar method for detecting bacterial motility is simple, easy to operate and does not involve large and expensive instruments.The method can be used by clinical microbiologists or researchers to investigate pathogenic bacteria. Demonstrating the procedure will be Xiyi Zhuang, a senior experimentalist from my laboratory. To begin, prepare the traditional semi-solid medium by adding 10 grams of Tryptose, 15 grams of sodium chloride and four grams of agar in an adequate volume of distilled water.Adjust the pH to 7.2 and make up the final volume to 1, 000 milliliters. Autoclave the medium at 121 degrees Celsius for 20 minutes and dispense the medium into 10 milliliter test tubes to a height of three centimeters. To prepare the TTC medium, cool the autoclaved traditional medium to 50 degrees Celsius then add five milliliters of sterile 1%TTC solution to 100 milliliters of medium.Mix and dispense the medium into 10 milliliter test tubes to a height of three centimeters. Pick single colonies of bacteria from agar plates and inoculate them into both the semi-solid media by puncture using inoculating needles. Remember to include non-modal strains as negative controls and modal strains as positive controls.To observe the effect of agar concentration on bacterial motility, prepare semi-solid media containing different agar concentrations and inoculate them by puncture. Incubate the tubes at 37 degrees Celsius and observe the results after 24 to 48 hours of incubation. If only the puncture line is red, characterize the bacteria as non-modal.If the red color spreads outward along the puncture line, characterize the bacteria as modal. Bacterial motility was compared for various strains on traditional and TTC semi-solid media and summarized in this table. After culturing for 24 hours on TTC semi-solid medium, Staphylococcus aureus showed growth along the inoculated line.Whereas, Escherichia coli showed growth in all directions around the inoculated line. Highest motility for Escherichia coli was observed in TTC semi-solid medium prepared with 0.3%agar in which the medium turned almost entirely red. In contrast, the area of red diffusion decreased in 0.5%agar and the diffusion was prolonged with increasing agar concentration to 0.8%It is important to adjust the pH to 7.2 and cool the autoclaved medium to 50 degrees Celsius before inoculation using the needles.Bacteria should be inoculated by puncture into the semi-solid medium containing TTC.
