極低温電子顕微鏡(cryo-EM)は、タンパク質の3次元(3D)構造を天然状態に分解するために開発されました1、2、3、4。この技術は、ガラス質氷の薄層(10〜100nm)にタンパク質を固定し、透過型電子顕微鏡(TEM)を使用してランダムに配向したタンパク質の投影画像を取得し、サンプルを液体窒素温度に維持することを含む。数千から数百万の投影画像が取得され、計算アルゴリズムによってタンパク質の3D構造を再構築するために使用される5,6。クライオEMによる分析を成功させるために、ブロッティング条件、湿度、温度を制御する装置をプランジフリーズすることにより、クライオサンプル調製が自動化されています。試料溶液を穴状の炭素膜を有するTEMグリッド上にロードし、連続的にブロットして余分な溶液を除去し、次いで液体エタンでプランジ凍結して薄いガラス質氷を生成する1,5,6。クライオEMの進歩とサンプル調製7の自動化に伴い、クライオEMは、ウイルスのエンベロープタンパク質や細胞膜のイオンチャネルタンパク質などのタンパク質の構造を解決するためにますます使用されています8、9、10。病原性ウイルス粒子のエンベロープタンパク質の構造は、ウイルス感染の病態を理解し、COVID-19パンデミックを引き起こしたSARS-CoV-2 11などの診断システムとワクチンを開発するために重要です。また、クライオEM技術は、電池12,13,14や触媒系14,15に用いられる光感応性材料のイメージングや、溶液状態の無機材料の構造解析16など、近年材料科学にも応用されている。
クライオEMおよび関連技術の著しい発展にもかかわらず、クライオサンプル調製には限界があり、ハイスループット3D構造解析の妨げとなっています。最適な厚さの硝子体氷膜を作製することは、原子分解能を有する生物材料の3D構造を得るために特に重要である。氷は、氷によって散乱された電子からのバックグラウンドノイズを最小限に抑え、電子ビーム経路1,17に沿った生体分子の重なりを禁止するのに十分なほど薄くなければならない。しかし、氷が薄すぎると、タンパク質分子が好ましい向きで整列したり、変性したりする可能性があります18、19、20。したがって、ガラス質氷の厚さは、目的の材料のサイズに応じて最適化する必要があります。さらに、サンプル調製と、調製されたTEMグリッド上の氷およびタンパク質の完全性の手動スクリーニングには、通常、広範な努力が必要です。このプロセスは非常に時間がかかり、ハイスループットな3D構造解析の効率を妨げます。したがって、クライオEMサンプル調製の信頼性と再現性の向上は、構造生物学および商業創薬、ならびに材料科学におけるクライオEMの利用を強化するであろう。
ここでは、氷の厚さを制御したハイスループットクライオEM用に設計された酸化グラフェン(GO)ウィンドウを備えたマイクロパターンチップを製造するための微細加工プロセスを紹介します21。マイクロパターン化されたチップは、イメージング目的に応じてチップの構造と寸法を操作できるマイクロエレクトロメカニカルシステム(MEMS)技術を使用して製造されました。GOウィンドウを備えたマイクロパターンチップは、サンプル溶液で充填できるマイクロウェル構造を有し、マイクロウェルの深さを調整してガラス質氷の厚さを制御することができる。生体分子に対するGOの強い親和性により、可視化のための生体分子の濃度が向上し、構造解析の効率が向上します。さらに、マイクロパターン化されたチップはSiフレームで構成されており、グリッド19に高い機械的安定性を提供し、サンプル調製手順およびクライオEMイメージング中のチップの取り扱いに最適です。したがって、MEMS技術によって作製されたGOウィンドウを備えたマイクロパターンチップは、クライオEMサンプル調製の信頼性と再現性を提供し、クライオEMに基づく効率的でハイスループットな構造解析を可能にします。

極低温電子顕微鏡(cryo-EM)を用いた生体分子の効率的かつハイスループットな構造解析における主な制限は、ナノスケールで氷厚を制御したクライオEMサンプルを調製することの難しさです。シリコン(Si)ベースのチップは、厚さ制御された窒化シリコン(SixNy)膜上にパターン化された酸化グラフェン(GO)窓を有するマイクロホールの規則的な配列を有し、マイクロエレクトロメカニカルシステム(MEMS)技術を適用して開発された。UVフォトリソグラフィー、化学蒸着、薄膜のウェットエッチングおよびドライエッチング、および2Dナノシート材料のドロップキャスティングは、GO窓を有するマイクロパターンチップの量産に使用された。マイクロホールの深さは、クライオEM分析用の試料のサイズに応じて、必要に応じて氷の厚さを制御するために調整されます。生体分子に対するGOの良好な親和性は、クライオEMサンプル調製中に目的の生体分子をマイクロホール内に集中させます。GOウィンドウを備えたマイクロパターンチップは、さまざまな生体分子や無機ナノ材料のハイスループットクライオEMイメージングを可能にします。

このプロトコルは、クライオEMを用いて異なるサイズのタンパク質やナノ材料をうまく観察するために、ナノスケールで画像サイズの厚さを調整するために重要です。Memの製造技術は、マイクロチップの大量生産を可能にします。また、マイクロパターンウェルの深さと設計の選択も可能です。実験目的に応じて。この技術は、市販の創薬に利用される生体分子のハイスループット3D構造解析の効率向上に貢献する可能性があり、シリコンニトロ膜のリスク傾向のある薄いウェーハやマイクロチップは扱いにくい場合があります。ウェーハを曲げたり、硝酸シリコンウィンドウに垂直な力を加えたりしないことが重要です。窒化シリコン蒸着シリコンウェーハをヘキサメチルジシラザン溶液で覆い、スピンコーダー上で3000 RPMで30秒間スピンコーティングすることにより、フォトレジストまたはPRを開始します。コーティングされたウェーハを摂氏95度でホットプレートで30秒間焼き、水面を疎水性にし、PRで良好なコーティング性能を確保します。次に、ウェーハにポジティブPRをスピンコートし、摂氏100度で90秒間焼きます。スピンコートPRの厚さは500ナノメートルです。PRコーティングされたウェーハをアライナーを使用してクロムマスクを通して紫外線に5秒間さらします。現像液を用いて1分間PRを現像し、ウェーハをイオン交換水に浸して2回すすぎます。その後、水面に窒素ガスを吹き付けてPRパターンウェーハを乾燥させます。PRのパターン化に従います。ラボで構築された反応性イオンエッチャーを無線周波数電力50ワットで使用し、六フッ化硫黄ガスを毎分3標準立方センチメートルで使用します。露出した窒化ケイ素を毎秒6オングストロームの速度でエッチングする。窒化ケイ素パターンウェーハをアセトンに室温で30分間浸漬することにより、PRを排除します。続いて、ウェーハをイオン交換水で2回リンスし、ウェーハを窒素ガスで乾燥させた。露出したsiをエッチングするには、新たに調製した水酸化カリウム溶液に窒化シリコンパターンウェーハを浸漬する。自立型窒化ケイ素窓まで連続的に攪拌しながら、パターン化されたウェーハの反対側に観察することができる。エッチングされたウェーハを脱イオン水浴に数回浸して洗浄します。次に、ウェーハを空気中で乾燥させます。エッチング残渣をなくすには、チップアレイの境界をピンセットで軽く押して、マイクロパターニングするチップのアレイを取得します。次に、チップアレイを新しく調製した水酸化カリウム溶液に30秒間浸漬した後、2回すすぎ、チップに窒素ガスを吹き付け、空気中で1時間乾燥させます。固体支持体については、前に示したように、スピンコーティングを施したブランクの525マイクロメートルシリコンウェーハを準備します。ウェーハを焼成する前にシリコンウェーハ上にチッパー光線を取り付け、前述の手順に従って、マイクロパターンシリコンウェーハを得た。パターンチップセットを1つのメチル2つの純粋なリタノール溶液に摂氏60度で一晩浸漬することによりPRを排除します。翌日、チップセットを脱イオン水で2回すすぎます。パターンチップセットを窒素乾燥後、反応性イオンエッチャーで150ワットの無線周波数電力で1分間、毎分100標準立方センチメートルの酸素ガスを使用する酸素プラズマプロセスでPR残留物を除去します。その後、マイクロパターンチップを水酸化カリウム溶液に30秒間浸して、PR残留物を完全に除去します。次に、チップセットをすすぎ、完全に乾かします。1ミリリットルあたり2ミリグラムの酸化グラフェンまたは溶液を脱イオン水で10倍に希釈し、希釈溶液を10分間超音波処理してシートの凝集体を破壊する。その後、希釈液を室温で30秒間Gの300倍で遠心分離する。1500万アンペアのグローディスチャージャーを使用して、マイクロパターンチップのシリコンエッチング側を1分間グロー放電し、チップの表面を正電荷でレンダリングします。完了したら、3マイクロリットルの溶液をマイクロパターンチップのグロー放電側にドロップします。1分後、チップ上の余分な溶液をろ紙で拭き取ります。転写したチップをパラフィンフィルム上の脱イオン水滴で洗浄し、余分なチップを濾紙で吸い取ります。ドロップキャスト手順を転送側で2回、反対側で1回繰り返します。移したチップを室温で一晩乾燥させます。フォトリソグラフィ手順の間、マイクロパターンチップの設計は、クロムマスクの異なる設計を使用して操作された。自立型窒化ケイ素膜の数と寸法が制御されました。作製されたマイクロパターンチップは、最大25, 000個の吊り下げ穴を有することができることが観察された。窓のラーメンスペクトルは、 さらに、多重配向六方晶屈折パターンは、窓が多層からなることを示し、窓付きマイクロホールの構造と深さを走査型電子顕微鏡と原子間力顕微鏡で調べました。窓付きマイクロホールのウェル型構造を画像に観察し、窓付きマイクロパターンチップの設計の可能性を確認しました。マイクロパターンチップの助けを借りて、いくつかの生物学的標本と無機ナノ粒子をクライオ電子顕微鏡で画像化しました。パターンの大きさやデザインに応じて、強度を利用した純粋な膜厚やマイクロパターニングの現像時間などの条件を最適化することが重要です。FIBやリソグラフェンなどのナノパターンを適用することにより、いくつかのマイクロメートルパターンを生成することができ、他の分析技術で使用されるこのマイクロデバイスのアプリケーションを拡大する可能性があります。
