극저온 전자 현미경 (cryo-EM)은 그들의 자연 상태 1,2,3,4에서 단백질의 3 차원 (3D) 구조를 해결하기 위해 개발되었습니다. 이 기술은 유리체 얼음의 얇은 층 (10-100nm)에 단백질을 고정시키고 샘플을 액체 질소 온도에서 유지하면서 투과 전자 현미경 (TEM)을 사용하여 무작위로 배향 된 단백질의 돌출 이미지를 획득하는 것을 포함합니다. 수천 내지 수백만 개의 프로젝션 이미지들이 획득되어 계산 알고리즘(5,6)에 의해 단백질의 3D 구조를 재구성하는데 사용된다. cryo-EM을 통한 성공적인 분석을 위해 블롯팅 조건, 습도 및 온도를 제어하는 장비를 급락 동결시킴으로써 냉동 시료 준비가 자동화되었습니다. 샘플 용액은 구멍이 많은 탄소 멤브레인이있는 TEM 그리드에 로딩되고, 연속적으로 블롯팅되어 과량의 용액을 제거한 다음 액체 에탄으로 급락 동결시켜 얇은 유리체 얼음 1,5,6을 생성합니다. cryo-EM의 발전과 시료 준비7의 자동화로 인해 cryo-EM은 세포막 8,9,10에서 바이러스용 엔벨로프 단백질 및 이온 채널 단백질을 포함한 단백질의 구조를 해결하는 데 점점 더 많이 사용되고 있습니다. 병원성 바이러스 입자의 외피 단백질의 구조는 바이러스 감염 병리를 이해하고 COVID-19 전염병을 일으킨 SARS-CoV-211과 같은 진단 시스템 및 백신을 개발하는 데 중요합니다. 더욱이, cryo-EM 기술은 최근 배터리(12,13,14) 및 촉매 시스템(14,15)에 사용되는 빔 감응성 물질을 이미징하고 용액 상태(16)에서 무기 물질의 구조를 분석하는 것과 같은 재료 과학에 적용되고 있다.
cryo-EM 및 관련 기술의 눈에 띄는 발전에도 불구하고 냉동 샘플 준비에는 한계가 존재하여 고처리량 3D 구조 분석을 방해합니다. 최적의 두께로 유리체 얼음 필름을 준비하는 것은 원자 분해능을 가진 생물학적 물질의 3D 구조를 얻는 데 특히 중요합니다. 얼음은 얼음에 의해 흩어져있는 전자의 배경 잡음을 최소화하고 전자 빔 경로1,17을 따라 생체 분자의 겹침을 금지 할만큼 충분히 얇아야합니다. 그러나, 얼음이 너무 얇으면, 단백질 분자가 바람직한 방향으로 정렬되거나 변성(18,19,20)될 수 있다. 따라서 유리체 얼음의 두께는 관심있는 물질의 크기에 따라 최적화되어야합니다. 또한, 일반적으로 준비된 TEM 그리드 상에서 얼음 및 단백질 무결성의 샘플 준비 및 수동 스크리닝을 위해 광범위한 노력이 필요합니다. 이 프로세스는 매우 시간이 많이 걸리므로 고처리량 3D 구조 분석의 효율성을 저해합니다. 따라서 cryo-EM 샘플 준비의 신뢰성과 재현성의 향상은 재료 과학뿐만 아니라 구조 생물학 및 상업용 약물 발견에서 cryo-EM의 활용도를 향상시킬 것입니다.
여기에서는 제어된 얼음 두께(21)를 갖는 고처리량 cryo-EM을 위해 설계된 산화그래핀(GO) 윈도우를 갖는 마이크로 패턴화된 칩을 만들기 위한 미세 제조 공정을 소개한다. 마이크로 패턴화된 칩은 이미징 목적에 따라 칩의 구조와 치수를 조작할 수 있는 미세전자기계 시스템(MEMS) 기술을 사용하여 제작되었습니다. GO 윈도우를 갖는 마이크로패터닝된 칩은 샘플 용액으로 충전될 수 있는 마이크로웰 구조를 가지며, 마이크로웰의 깊이는 유리체 얼음의 두께를 제어하도록 조절될 수 있다. 생체 분자에 대한 GO의 강한 친화력은 시각화를위한 생체 분자의 농도를 향상시켜 구조 분석의 효율성을 향상시킵니다. 또한, 마이크로 패턴화된 칩은 그리드(19)에 높은 기계적 안정성을 제공하는 Si 프레임으로 구성되어, 샘플 준비 절차 및 cryo-EM 이미징 동안 칩을 취급하는 데 이상적입니다. 따라서 MEMS 기술로 제작된 GO 윈도우가 있는 마이크로 패턴 칩은 cryo-EM 샘플 준비의 신뢰성과 재현성을 제공하여 cryo-EM을 기반으로 효율적이고 높은 처리량의 구조 분석을 가능하게 합니다.
