Cette technique présente une approche pratique pour estimer plusieurs biomarqueurs fonctionnels de spermatozoïdes humains par cytométrie en flux sans compromettre substantiellement la qualité de l’expérience induite par des temps d’attente prolongés. Cette technique peut être une boîte à outils pratique et fiable pour le diagnostic de l’infertilité et l’évaluation de la fonction des spermatozoïdes à la fois sur le banc et au lit, et compléter l’examen de routine du sperme. Cette procédure est réalisée avec la plate-forme Accuri C6 et elle pourrait être appliquée à d’autres plates-formes commerciales avec des modifications mineures.
Pour commencer, incubez l’échantillon de sperme à 37 degrés Celsius et liquéfiez complètement l’échantillon pendant une heure. Pour compter les spermatozoïdes, mélangez soigneusement l’échantillon de sperme liquéfié, puis ajoutez 10 microlitres de sperme dans une chambre de comptage des spermatozoïdes et scannez la lame dans un analyseur de classe de sperme. Comptez au moins six zones et 400 spermatozoïdes pour estimer la concentration de spermatozoïdes.
Pour la détection de l’apoptose des spermatozoïdes, à l’aide de la coloration à l’annexine-5 FITC PI, ajoutez 10 aux sixièmes spermatozoïdes dans un tube à centrifuger de 1,5 millilitre. Lavez les cellules avec 500 microlitres de PBS froid. Ensuite, centrifugez la suspension des spermatozoïdes et jetez le surnageant avant de remettre les cellules en suspension dans 200 microlitres de tampon de liaison.
Ensuite, ajoutez deux microlitres d’annexine-5 FITC et deux microlitres de PI. Faites vortex doucement les cellules et incubez-les pendant 15 minutes à température ambiante dans l’obscurité avant de les placer dans le cytomètre en flux pour analyse. Pour l’analyse du potentiel membranaire mitochondrial, ou MMP, à l’aide de la sonde JC-1, ajoutez deux fois 10 aux sixièmes spermatozoïdes dans un tube de 1,5 millilitre et centrifugez. Après centrifugation, remettez en suspension la suspension de spermatozoïdes dans une solution de travail JC-1 d’un millilitre.
Faites tourbillonner doucement les cellules et incubez-les pendant 20 minutes à 37 degrés Celsius dans l’obscurité. Encore une fois, centrifugez la suspension, jetez le surnageant et lavez la pastille deux fois avec un millilitre de PBS. Ensuite, suspendez à nouveau les spermatozoïdes colorés dans un millilitre de PBS dans un tube de cytomètre en flux et placez immédiatement le tube dans le cytomètre en flux pour analyse.
Ensuite, utilisez le cyanure de carbonyle m-chlorophénylhydrazone, ou CCCP comme témoin positif. Remettre les spermatozoïdes en suspension dans un millilitre de solution de travail CCCP et les faire tourbillonner doucement avant d’incuber à température ambiante pendant 20 minutes. Après l’incubation, centrifugez la suspension, jetez le surnageant et effectuez les étapes d’analyse de la MMP à l’aide de la sonde JC-1 comme démontré.
Pour le test de la structure de la chromatine des spermatozoïdes, ou SCSA, décongelez l’échantillon de sperme au bain-marie à 37 degrés Celsius et diluez-le avec un tampon TNE à une concentration d’une à deux fois 10 à la sixième cellule par millilitre. Ensuite, ajoutez 200 microlitres de l’échantillon dilué dans un tube à essai de cytomètre en flux et mélangez-le avec 400 microlitres de solution acide. Colorer l’échantillon avec 1,2 millilitre de solution d’acridine orange.
Utilisez un échantillon de référence comme étalon interne pour la configuration et les étalonnages du cytomètre en flux. Diluer l’échantillon avec un tampon TNE à quatre degrés Celsius à une concentration d’une à deux fois 10 à six cellules par millilitre, et effectuer les étapes pour l’ACSS, comme démontré. Ouvrez le logiciel du cytomètre en flux et démarrez le cytomètre.
Pour collecter des données d’échantillon, chargez un échantillon de contrôle sur le cytomètre en flux et cliquez sur Exécuter pour démarrer la collecte de données. Créez des diagrammes à points pour afficher les données. Sélectionnez les paramètres de l’axe XY et affichez linéaire pour spécifier les données.
Ensuite, sélectionnez et appliquez une porte polygonale pour délimiter la population de spermatozoïdes à analyser. Pour l’apoptose des spermatozoïdes, définissez FL1 comme axe X et FL2 comme axe Y. Ensuite, pour isoler les cellules apoptotiques ou nécrotiques vivantes, appuyez et faites glisser la porte du quadrant pour subdiviser les populations de spermatozoïdes en quatre populations spécifiques.
Pour MMP, définissez FL1 comme axe X et FL2 comme axe Y. Ensuite, créez une porte polygonale pour subdiviser les populations de spermatozoïdes en deux populations spécifiques. Pour SCSA, définissez FL4 comme axe X et FL1 comme axe Y.
Dessinez la porte, définissez un angle de 45 degrés pour exclure les signaux de débris cellulaires. Calculez 10 000 spermatozoïdes pour chaque échantillon pour les analyses d’apoptose des spermatozoïdes, MMP et SCSA, définissez une limite de traitement pour indiquer quand arrêter la collecte de données, le taux fluidique, en fonction du nombre de cellules et le seuil pour éliminer les débris et le bruit. Appliquez ces paramètres à tous les échantillons.
Si nécessaire, réglez la compensation des couleurs pour corriger les débordements fluorescents. Pipetez doucement l’échantillon avant de le charger sur le cytomètre en flux. Ensuite, entrez le nom de l’exemple et cliquez sur Exécuter.
Une fois l’exécution terminée, enregistrez les exemples de données en tant que fichier FCS en donnant un nom de fichier pour une analyse plus approfondie. La mesure de l’apoptose des spermatozoïdes à l’aide de la coloration FITC PI à l’annexine-cinq est présentée ici. Cette analyse comprenait un contrôle négatif des spermatozoïdes non colorés pour définir la compensation et les quadrants.
Dans un échantillon présentant une apoptose des spermatozoïdes, les résultats ont été exprimés sous la forme de pourcentages de cellules apoptotiques précoces ou apoptotiques viables ou vivantes, d’apoptose tardive et de cellules nécrotiques. La sous-population de spermatozoïdes avec une MMP élevée et faible est montrée ici. Un échantillon de sperme de bonne qualité a montré une fluorescence orange élevée et une faible fluorescence verte.
À l’inverse, un échantillon de sperme de mauvaise qualité présentait une faible fluorescence orange et une fluorescence verte élevée. Les cytogrammes SCSA d’un homme fertile et infertile sont présentés ici. La population de spermatozoïdes a été identifiée comme normale avec une stabilité élevée de l’ADN, un indice de fragmentation de l’ADN et des débris cellulaires.
L’échantillon de sperme doit être incubé à 37 degrés Celsius et complètement liquéfié pendant une heure maximum.
