以前にインビトロまたはエキソビボモデルを使用していたが、皮膚細胞成分および皮膚組織の複雑さを欠いている。このex situSCADアッセイは、設定された制限を克服し、負傷した動物以外の瘢痕の発生を研究することを可能にする。SCADアッセイは、皮膚微小環境における線維芽細胞移動および瘢痕形成の可視化を可能にする。これにより、アクチベーターまたはインヒビターのスクリーニングライブラリが瘢痕形成の機構的基礎を理解することができます。創傷修復に関与する基本的なプロセスと分子コースを理解する上で、SCADモデルを用いたフラットライブラリのハイスループットスクリーニングアッセイ。SCADアッセイは、皮膚外植体を使用して行うのが容易である。一貫した瘢痕のためには、出生後0日目または1日目の皮膚を選択することをお勧めします。出生後0日目または新生児1日目の子犬を犠牲にした後、滅菌手術用メスを使用して、骨格筋層まで1.5×1.5センチメートルの全厚背部背部皮膚を慎重に切除する。滅菌湾曲した鉗子を使用して皮膚を剥がし、表在筋膜が根底にあるパンニクルス・カルノーサス筋で無傷であることを確認する。摘出した組織を50~100ミリリットルの冷たいDMEM F-12培地で洗浄し、汚染された血液を除去します。次いで、組織および細胞生存率を維持するためにハンクス平衡塩溶液で組織を洗浄する。DMEM F-12培地を含む10センチメートルのペトリ皿に、表在筋膜を上にして皮膚を逆さまに置きます。次いで、使い捨ての2ミリメートル生検パンチを用いて、全厚の丸い皮膚片を切除してかさぶた組織を生成し、表皮まで下層のパンニクルス・カルノーサス筋で表在筋膜が無傷であることを確認する。フェノールレッドを含まない200マイクロリットルの新鮮なDMEM F-12完全培地を96ウェルプレートの各ウェルに加える。滅菌鉗子を用いて、個々のかさぶた組織を逆さまに移し、96ウェルプレートのウェルに完全に浸漬する。プレートを標準条件下で維持した細胞培養インキュベーターに移す。培養の2日目および4日目に、ウェルに10マイクロリットルを残した培地を除去し、新鮮な予め加温したDMEM F-12完全培地を処理化合物と共に加えて、継続的な細胞および組織の生存率条件を維持する。SCADのライブイメージングのために、電子レンジで加熱することによって、ガラス瓶中のPBS中の2〜3%の低融点アガロース溶液の30ミリリットルを調製する。沸騰後、すぐにボトルを移し、液体アガロース溶液を摂氏40度の水浴中で冷却する。次に、筋膜または瘢痕を上向きにしてSCAD組織を35ミリメートル皿の中心に移す。その後、1000マイクロリットルのピペットチップを使用して、摂氏40度の液体アガロースを組織にゆっくりと注ぐことにより、室温でSCADを埋め込む。アガロースは2分以内に重合する。重合後、2ミリリットルの予備加温DMEM F−12完全培地を加える。次に、原稿で説明した適切なインキュベーションシステムを備えた共焦点顕微鏡または多光子顕微鏡を使用して、0日目から1日目までのタイムラプス画像を取得します。組織採取のために、培地を滅菌PBSで交換することによって、関連する時点で組織を洗浄する。滅菌鉗子を使用して、各SCADを500マイクロリットルの2%パラホルムアルデヒドを含む1.5ミリリットルのマイクロ遠心チューブに移し、組織を摂氏4度で一晩固定した。翌日、PBSで組織を3回洗浄してから、本文原稿に記載されているように2次元または3次元免疫蛍光染色に進む。0日目と5日目のSCADの代表的な全マウント明視野画像がここに示されています。マッソンによる垂直組織切片のトリクローム染色は、0日目および5日目に組織瘢痕化、組織収縮、および瘢痕コアにおける細胞外マトリックスの蓄積のシグネチャを明らかにする。0日目および5日目におけるSCADの免疫蛍光染色が示されている。初期段階および後期段階の瘢痕から、浸潤陽性線維芽細胞は緑色で示され、浸潤陰性線維芽細胞は赤色で示される。三次元画像化のために、組織をアガロース溶液に包埋し、PBSGTをトッピングした。代表的な画像は、5日目のSCADの瘢痕部位におけるN-カドヘリンタンパク質の排他的な局在化を実証する。インキュベーションチャンバーを備えた3次元タイムラプスイメージングセットアップは、最初の12時間または瘢痕発生の初期段階にわたる線維芽細胞群の進行の初期事象を示した。瘢痕発生にわたる個々の線維芽細胞の単一細胞追跡細胞軌道のグラフ表示をここに提示する。筋膜が上を向くように、SCAD組織をウェルプレートの内側に逆さまに配置することが重要です。これを怠ると、移行パターンに避けられないばらつきが生じます。この手順は、化学的モジュレーター、中和抗体、またはウイルス法を用いた治療または培養のための真皮層を調べることを可能にする。これは、多様な医療現場にわたる病理学的線維化応答の評価に役立ちます。我々は、コネキシン-43およびN-カドヘリンを、創傷時の筋膜動員に関与する重要な分子として発現させた。SCAD方法論は、瘢痕化および線維症を抑制するための新規治療戦略の同定を可能にする。
