本質的ラベリング Feバイオアベイラビリティの評価のための植物性食品の固有の標識は、外因性標識における平衡化の精度の問題を排除します。しかしながら、このアプローチは、水耕栽培のための温室スペースを必要とするため、大量の材料を生み出すことができない。水耕栽培は労働集約的であり、大量の高価な安定同位体を必要とし、そしてしばしば収量および種子Fe濃度の点で異なる植物の成長をもたらす。コストがかかるため、内因性標識は、Feの取り込みの根底にあるメカニズムや食品からのFeの取り込みに影響を与える要因を理解することを目的とした小規模な研究にのみ適しています。主食作物の1〜2 kgの生産には、同位体と水耕栽培のアプローチにもよりますが、材料だけで約20,000ドルから30,000ドルの費用がかかります13,14。
同位体標識に関連する課題を考慮して、研究者はin vitroアプローチの開発を目指しました。初期の方法は、生物学的利用能の推定値としてFe溶解度またはFe透析可能性の測定と相まって、シミュレートされた胃および腸の食物を利用しました15。このような研究では、Feは可溶性であり、化合物に強固に結合しているため、交換不可能であり、バイオアベイラビリティの過大評価につながる可能性があるため、Fe透析可能性はバイオアベイラビリティの一貫した尺度ではないことがすぐにわかりました。これらの課題に対処するために、ヒト腸管細胞株を利用する方法論が進化し、それによって生体成分が追加され、Fe取り込み16の測定が可能になりました。ヒト腸細胞であるCaco-2細胞は、ヒト結腸癌に由来し、栄養摂取研究に広く使用されています。この細胞株は、培養において、細胞が小腸のブラシ境界細胞と同様に機能する腸細胞に分化するので有用である。研究によると、Caco-2細胞は、Feの取り込みに影響を与える因子に対して適切なトランスポーターと応答を示します17,18。
Caco-2細胞におけるFe取り込みを測定するために放射性同位元素を利用した最初の研究は、Caco-2細胞のフェリチン形成に基づいてFe取り込みを測定するように改良された。Caco-2細胞フェリチン測定は、サンプルスループットを向上させ、放射性同位元素の取り扱いと外因性Feと内因性Fe19,20の平衡化の問題を打ち消しました。フェリチン形成によるFe取り込みの測定により、研究者は複雑な食事を含む幅広い食品を研究することができました21。したがって、Caco-2細胞のFe取り込みと組み合わせたシミュレートされた(in vitro)消化は、食品からのFe取り込みのより良い生理学的評価を提供しました。このモデルは主にFeバイオアベイラビリティの相対的な違いを決定することに注意することが重要です。多くの有用な細胞株と同様に、Caco-2細胞も応答性にばらつきを示していますが、食品間のFe取り込みに一貫した相対的な違いを維持しています。適切な技術と細部への注意深い注意は、Caco-2細胞における一貫した細胞フェリチン形成応答を改善することができる。
