身体活動の欠如または廃用は、筋萎縮および骨量減少1および全身のコンディショニング解除2などの自発運動エフェクターの劣化をもたらす。さらに、最近、不活動が筋骨格成分の構造的側面だけでなく、運動の質的側面にも影響を与えることが注目されています。例えば、模擬微小重力環境にさらされたラットの四肢位置は、介入終了後1ヶ月でも無傷の動物のそれとは異なっていた3,4。それにもかかわらず、非活動によって引き起こされる運動障害についてはほとんど報告されていません。また、劣化の包括的な運動特性は完全には決定されていません。
現在のプロトコルは、後肢の除荷を受けたラットの廃用によって誘発される歩行運動障害を参照することにより、運動変化を視覚化するための運動学的評価の適用を実証し、議論しています。
模擬微小重力環境後の歩行における四肢の過伸展は、ヒト5と動物4,6,7,8の両方で観察されることが示されています。そこで普遍性のために、本研究では一般的なパラメータである膝関節と足首関節の角度と、立脚期の中間点(中脚)における中足指節関節と股関節の垂直距離(股関節の高さにほぼ相当)に着目した。さらに、ビデオ運動学的評価の潜在的な応用が議論で示唆されています。
一連の運動学的分析は、神経制御の機能的側面を評価するための効果的な尺度であり得る。しかし、モーション解析は、フットプリント観察またはキャプチャされたビデオ9,10の単純な測定から複数のカメラシステム11,12まで開発されているが、普遍的な方法およびパラメータはまだ確立されていない。本研究の方法は、この関節運動解析に包括的なパラメータを提供することを目的としています。
前研究13では、神経病変モデルラットの歩行変化を網羅的な映像解析を用いて図解することを試みた。ただし、一般に、モーション解析の潜在的な結果は、解析フレームワークで提供される所定の変数に制限されることがよくあります。このため、本研究では、広く適用可能なユーザー定義パラメータを組み込む方法をさらに詳しく説明しました。ビデオ解析を使用した運動学的評価は、適切なパラメータが実装されている場合、さらに役立つ可能性があります。

廃用が神経系に影響を及ぼし、関節の動きが変化することはよく知られています。しかし、どのアウトカムがこれらの特性を適切に示すかはまだ不明である。本研究では,映像キャプチャからの3次元(3D)再構成を利用した動作解析手法について述べる.この技術を用いて、模擬微小重力環境にさらされたげっ歯類において、後肢を尾で降ろすことによって、不使用誘発の歩行性能の変化が観察されました。2週間の荷降ろしの後、ラットはトレッドミルの上を歩き、歩行運動を4台の電荷結合素子(CCD)カメラでキャプチャしました。3Dモーションプロファイルを再構築し、画像処理ソフトウェアを使用して対照被験者のモーションプロファイルと比較しました。再構築されたアウトカム指標は、歪んだ歩行運動の明確な側面をうまく描写しました:膝と足首の関節の過伸展と立脚期中の股関節のより高い位置。モーション解析は、いくつかの理由で役立ちます。まず、主観的な観察(特定のタスクでの合格/不合格など)の代わりに定量的な行動評価が可能になります。次に、基本的なデータセットを取得したら、特定のニーズに合わせて複数のパラメーターを抽出できます。より広範な適用のハードルにもかかわらず、労働集約度およびコストを含むこの方法の欠点は、包括的な測定および実験手順を決定することによって軽減され得る。
