操纵纳米颗粒的一种有效方法是使用由铁微粒形成的微流体磁阱，铁微粒包装在微流体结构3中。因此，当外部磁铁接近时，在磁化多孔介质内，磁力和磁通力之间会产生复杂的相互作用。作用在纳米颗粒上的磁力足够强，可以捕获它们并抵抗流动阻力3，4，7。这种方法需要微细加工技术，达到几微米量级的分辨率，以产生保留微粒的微度量结构。

在微铣削中常用的所有材料中，热塑性塑料由于其许多有利的性能和与生物应用的兼容性而仍然是首选10，14。热塑性塑料是微流体系统的一种有吸引力的基材，因为它们在开发低成本、一次性分析系统方面具有显著优势9.此外，这些材料非常适合大批量制造工艺，使其适合商业化和大规模生产。由于这些原因，自微流体10早期以来，PMMA等热塑性塑料一直被认为是可靠和坚固的材料。已经描述了不同的协议来制造热塑性塑料中的闭合通道，例如溶剂键合15，热键合16和紫外线（UV）/臭氧表面处理粘合17。

我们的协议描述了一种高分辨率微铣削方法，用于制造丙烯酸微流体装置，用于分析物浓度的定量免疫检测，与金标准ELISA技术相当。亮点是制造一种简单且低成本的热塑性微流体装置，无需洁净室，这样可以缩短检测时间，并以定量方式实现良好的检测限。从表面磨削开始。用 800 微米立铣刀钻头切割 9 x 25 毫米矩形的 1.3 毫米厚的 PMMA。用双面胶带小心地将这些矩形之一连接到压电平台上。连接 Z 传感器并将其放置在 PMMA 矩形的表面上。选择检测引脚并将其移到传感器表面上。在不接触传感器的情况下手动降低引脚。激活 Z 零感应模式。以 14， 500 rpm 的速度旋转 200 微米立铣刀钻头。慢慢将其降低到 z 轴上的原点坐标。然后将 z 轴重置为原点下方 30 微米。将此坐标设置为新的 Z 原点。单击微铣机软件中的切割按钮以激活切割面板。单击“添加”按钮，然后选择带有先前创建的用于研磨丙烯酸表面的代码的 TXT 文件。点击 输出量 按钮开始该过程。对于五微米限制的铣削，首先，将立铣刀钻头的旋转速度设置为11， 000 rpm。然后，使用压电平台的接口将平台升高6.5微米。沿 y 轴将立铣刀钻头移动 500 微米。使用控制接口将压电平台返回到其在z轴上的初始值。接下来，通过从设计软件打开先前创建的设计文件来执行微通道铣削。单击“打印”按钮，访问属性菜单，然后单击与包含要加工设计的图层相对应的颜色窗口。在工具面板中设置制造参数。对于孔的铣削，请切换到 800 微米立铣刀钻头。通过单击相应的颜色窗口并选择相应的制造参数来激活 1.2 毫米直径孔的设计层。在矩形的对角上加工两个额外的孔，以便在新平台上以倒置方式对齐丙烯酸。翻转亚克力，用双面胶带将其粘在带有机加工支柱的适配器上。从设计软件中打开包含对面孔设计的文件。铣削剩余一半的试剂入口和出口孔，直径为1.5毫米，深度为0.7毫米。用异丙醇清洁两个丙烯酸矩形片，并用蒸馏水冲洗。将丙烯酸浸入超声波浴中10分钟。干燥两张亚克力板，并用双面胶带将它们粘在玻璃培养皿盖的内部。然后将玻璃培养皿的底部放入较大的玻璃培养皿中。将一毫升氯仿倒入培养皿的底部，并迅速将盖子与丙烯酸片一起放置。立即将蒸馏水添加到较大的培养皿的底部，直至培养皿盖的水平。让丙烯酸暴露于氯仿气体一分钟。然后倾斜培养皿以打破水封，并立即揭开培养皿。为了粘合，将两种丙烯酸树脂与氯仿暴露的侧面面对面对齐并形成三明治。将丙烯酸树脂放入压榨机中两次，每次两分钟，改变丙烯酸的对齐方式。然后用即时干燥液体粘合剂将两到三厘米的软管连接到设备的每个孔上。使用注射器用蒸馏水填充通道。将设备浸入超声波浴中10分钟。然后清空设备通道内的水，并使用注射器引入5%BSA溶液。在5%BSA中制备直径为7.5微米的铁微粒悬浮液。将芯片和微粒悬浮液与封闭溶液在室温下孵育至少一小时。接下来，用注射器针头通过侧通道出口软管将微粒插入芯片中。垂直放置芯片，然后以 90 度的两步旋转芯片，使微粒在 5 微米限制处瞄准并压实。用热量密封丙烯酸装置的所有软管。切断入口软管，直到只剩下几毫米。用洗涤缓冲液填充分配针，然后将其插入切割软管中。让溶液滴落，然后将针头连接到设备。从侧向通道切断出口软管，然后连接到注射泵。接下来，对主通道出口软管重复相同的过程。然后将芯片连接到磁铁上。对于免疫检测，保持洗涤缓冲液以每小时50微升的速度流动10分钟。用微量移液管从分配针上除去剩余的洗涤缓冲液，并加入50微升纳米颗粒悬浮液。以每小时100微升的流速流动纳米颗粒悬浮液七分钟。然后将流速更改为每小时 50 微升，并继续流动 15 分钟。更换分配针并以相同的速率流动洗涤缓冲液10分钟。用微量移液管从分配针上除去剩余的洗涤缓冲液，并加入100微升荧光底物。调整底物输入、荧光测量和洗涤步骤的流速和时间参数。以每小时 50 微升的速度激活荧光底物的输入流六分钟。在基板流动停止前15秒，打开显微镜的荧光。在基板停止前10秒使用显微镜相机的软件开始图像捕获，曝光时间为1， 000毫秒。在基材清洗停止后立即单击所需流速参数的“开始”按钮。以每秒一帧的速度执行六分钟的成像。在选定的测量流量停止后，立即单击洗涤流的开始按钮。采用溶菌酶偶联纳米颗粒和马萝卜过氧化物酶偶联二抗进行免疫测定。比较陷阱前后的区域，观察到不同浓度的一抗荧光强度增加，表明底物荧光的变化与一抗浓度成正比。对于给定浓度的一抗，将荧光强度绘制为荧光底物不同流速下的时间函数。HRP酶对底物的转化能力与流速成反比，流速为每小时1微升时获得最大强度。对于不同一抗浓度的不同流速，免疫反应前后的荧光差异曲线显示，对于1，000纳克/毫升的浓度，荧光对于所有评估的流速都饱和。使用每种流速下获得的荧光强度相对于一抗浓度的最大值制备校准曲线。每小时一微升的高变异性和高荧光水平表明，该速率不利于反应底物的流动，并且倾向于在陷阱之后积累。应特别注意微通道的密封步骤，因为暴露于氯仿对温度非常敏感。为了获得可重复的结果，温度必须始终相同。我们的系统有助于了解微粒的致密度和尺寸、纳米粒径、抗原、检测抗体和底物在基于纳米颗粒的微流控免疫分析中检测限的决定因素。
