植物細胞壁の機械的性質は、細胞の形状およびその成長能力を決定する。例えば、花粉管の成長先端は、同じ管1の非成長部分よりも柔らかい。シロイヌナズナ分裂組織上の原基形成は、将来の原基2,3の部位における細胞壁剛性の局所的な減少によって先行される。シロイヌナズナの胚軸の細胞壁は、主成長軸に平行で成長が速く、この軸に垂直で成長が遅い細胞壁よりも柔らかくなっています4,5。トウモロコシの根では、分裂から伸長への細胞の移行は、根のすべての組織における弾性率の低下を伴っていた。弾性率は伸長帯では低いままであり、後期伸長帯6で増加した。
様々な方法が利用可能であるにもかかわらず、毎年得られる細胞壁生物学に関する生化学的および遺伝学的情報の大規模な配列は、細胞壁の機械的特性と比較されることはめったにありません。例えば、細胞壁関連遺伝子上の変異体は、しばしば成長および発達を変化させるが4,7,8、バイオメカニクスの観点から記述されることはめったにない。その理由の1つは、細胞レベルおよび細胞内レベルでの測定を行うことの難しさです。原子間力顕微鏡(AFM)は現在、そのような分析の主要なアプローチです9。
近年、植物細胞壁のバイオメカニクスに関するAFMベースの研究が数多く行われています。シロイヌナズナ2,3,4,5,10,11およびタマネギ12の外側組織、ならびに培養細胞13,14,15の細胞壁の機械的性質が調べられている。しかしながら、植物の表在細胞は、内部組織のそれらとは異なる機械的性質を有する細胞壁を有し得る6。さらに、植物細胞は膨圧によって加圧され、植物細胞をより硬くします。turgor圧の影響を取り除くために、研究者はプラスモライザー溶液2,3,4,5,10,11を使用するか、得られた値をturgorと細胞壁の寄与に分解する必要があります12。第1のアプローチは、試料の脱水をもたらし、細胞壁16の厚さおよび特性を変化させるが、第2のアプローチは、追加の測定および複雑な数学を必要とし、比較的単純な形状の細胞にのみ適用される12。内部組織の細胞壁特性は、凍結切片17または樹脂8を含浸させた植物材料の切片で評価することができる。ただし、どちらの方法でもサンプルの脱水や含浸が必要であり、必然的に特性が変化します。単離または培養された細胞の特性は、植物全体の生理機能に関連付けることは困難です。植物細胞の培養と単離の両方が、それらの細胞壁の機械的性質に影響を与える可能性があります。
ここで紹介する方法は、前述のアプローチを補完するものです。それを使用して、あらゆる組織の一次細胞壁および植物発生のあらゆる段階で調べることができる。切片作製およびAFM観察は、サンプルの脱水を回避する液体中で行った。細胞が切断されるにつれて、turgorの問題は解決されました。プロトコルはトウモロコシとライ麦の根での作業について説明していますが、他のサンプルは、ビブラトームの切片化に適しているかどうかを調べることができます。
ここで説明するAFM研究は、力-体積技術を使用して実行されました。このメソッドには、異なる機器が異なる名前を使用します。ただし、基本原則は同じです。試料の力−体積マップは、カンチレバー(または試料)の正弦波または三角形の運動によって得られ、カンチレバーのたわみ18を記録しながら、分析された各点で特定の荷重力を達成する。結果は、サーフェスの地形図と力-距離曲線の配列を組み合わせたものです。各曲線は、特定のポイントでの変形、剛性、ヤング率、接着力、およびエネルギー散逸を計算するために使用されます。同様のデータは、接触モード19でスキャンした後のポイントバイポイントフォース分光法によって得ることができるが、それはより時間がかかる。

植物細胞の成長は、それらの細胞壁の機械的性質によって制御される。したがって、植物のさまざまな臓器や組織でこれらの特性を知ることが重要です。ここに提示された方法は、若い植物の内部組織における非固定および非脱水細胞壁の生体力学的特徴付けを可能にする。ビブラトーム切片作成用の溶液とサンプルの調製から始めます。ペトリ皿の底に4ミリメートルの溶かした3%アガロースの層を注ぎ、サンプルへの熱損傷を防ぐために少し冷まします。長さ約5ミリメートルの植物器官の3つまたは4つの部分をアガロースの上に水平に置きます。約30〜60秒後、最初のアガロース層の上に薄い半固体フィルムが現れます。次に、慎重に2番目の層を上に注ぎます。アガロースが完全に固まったら、検体を含むブロックを切り取ります。ブロックを六角形の切頭ピラミッドに成形して、さらなる断面作成中の安定性を確保します。ビブラトームでサンプル切片作成を開始する前に、シアノアクリレート接着剤でブロックをビブラトームステージに接着します。ピラミッドの角の1つがビブラトームの刃に面するようにステージをビブラトームに配置し、ビブラトームバスに水を注ぎます。断面の厚さ、ブレード速度、振動周波数などの切断パラメータを設定し、サンプルを切断します。細いブラシを使用して、セクションを水浴からスライドガラスに移動し、セクションに水滴を置き、乾燥を防ぎます。光学顕微鏡で切片の品質を確認した後、断面平面に垂直な細胞壁を有する適切な切片を選択する。次に、ピペットを使用してペトリ皿キャップの底に1%溶融アガロースの1ミリリットルの層を注ぐことにより、原子間力顕微鏡測定用の切片を固定化します。アガロースが固まったら、ろ紙を端に近づけて、セクションから余分な水分を取り除きます。ブラシを使用して、スライドからペトリ皿キャップの中央にセクションを慎重に移します。次に、20マイクロリットルのピペットを使用して切片の周りに1%アガロースを慎重に添加し、原子間力顕微鏡用の水またはその他の溶液を固定化した切片のあるペトリ皿キャップに注ぎます。光学顕微鏡を使用して原子間力顕微鏡カンチレバーの下でサンプルをガイドします。アプローチボタンをクリックしてからランディングボタンをクリックして、1ナノアンペアの設定値で接触モードでサンプルにアプローチします。[スキャン]ボタンをクリックしてから、[領域]ボタンをクリックします。スキャンする領域サイズを 50 マイクロメートル x 50 マイクロメートルから選択します。[プローブの移動]ボタンをクリックし、スキャナーをその上に移動し、スキャナーの突出の程度に基づいて最高点を見つけて、スキャン領域全体を確認します。[アプローチ] タブを開きます。次に、[削除]ボタンをクリックして、サンプルから撤回します。最高点をターゲットとして使用し、[ランディング]ボタンをクリックしてサンプルに再度アプローチします。次に、[プローブの移動]ボタンをクリックし、スキャナーをその上に移動して、表面をもう一度確認します。スキャンレートを0.5ヘルツに設定し、スキャンサイズを50マイクロメートル×50マイクロメートル、スキャンポイントを64×64に設定します。[実行]ボタンをクリックしてスキャンし、サンプルの表面を確認し、アガロースによる汚染の可能性があります。[オン]ボタンをクリックした後、プログラムのメインウィンドウのドロップダウンメニューでHDPlusモードを選択し、メインプログラムウィンドウで設定値を0.1ナノアンペアに設定します。HDウィンドウのメインタブで、調査対象のサンプルに適したスキャンパラメータを設定します。次に、HDウィンドウの[ノイズ]タブを開き、カンチレバーの共振周波数を入力します。HDウィンドウの[クォント]タブを開き、IOS、カンチレバー剛性、先端半径、角度を入力します。先端形状に応じて計算に使用される接触モデルを選択します。この後、HDウィンドウの[スキャン]タブを開き、信号と信号が記録される方向を選択します。[フォースボリューム]ボックスにチェックマークを付けてすべてのフォースカーブの記録を取得し、メインプログラムウィンドウの上部にある[オフ]ボタンをクリックしてフィードバックループをオンにしますメインHDウィンドウのフェーズコアボタンをクリックして、光学システムの感度を修正します。メインHDウィンドウの対時間タブには、DFL信号対時間の機能がリアルタイムで表示されます。ベースライン水準の決定に使用するこの関数の部品を選択し、さらなる計算のために接触モデルを適合させます。次に、プログラムのメインウィンドウでスキャンポイントの値を256 x 256に設定します。次に、スキャンレートを0.2ヘルツに設定し、[実行]ボタンをクリックしてサンプルをスキャンします。スキャンが停止したら、メインウィンドウの上部にある[オン]ボタンをクリックして、フィードバックループをオフにします。ドロップダウンメニューで[コンタクトモード]を選択し、[アプローチ]タブを開き、[削除]ボタンをクリックしてサンプルから撤回します。[データ]ボタンをクリックして分析ソフトウェアを開き、出力を保存します。保存したファイルを解析ソフトで開きます。HDフォースボリュームフレームを選択します。Ctrlキーを押しながら、同じスキャン方向で取得したビジュアルフレームを1つ選択します。[外部マップの読み込み]ボタンをクリックして、セル壁がどこにあるかを確認します。メインタブでInvOptSensとカンチレバー剛性の値を確認します。[追加]タブを開き、チップパラメータと接触モデルを確認します。ビジュアルフレーム上のセル壁上のさまざまな点をクリックし、モデルによって適切に記述された曲線のみを選択します。弾性率マップの白い領域は、スキャナーがZ方向の限界に達したことによるヤング率の誤った過大評価に対応しています。この画像は、満足のいく力曲線をさらに選択するための外部マップとして使用するのは便利ではありません。ただし、右側に示されているDFL信号マップは、ここではより適しています。異なる計測器は、DFL信号を偏向信号またはエラー信号と呼ぶ場合があります。サンプルの底に到達しようとしている間に完全に拡張されたスキャナーは、誤った測定やスキャンの中断につながる可能性があります。アガロースの存在は、接触モードで最初のスキャンを取得しながら、細胞壁といくつかの細胞底部がそのようなスキャンで見えるようにチェックすることができます。ただし、不正確な固定化の場合、表面がアガロースで覆われ、サンプルのトポグラフィーがマスクされる可能性があります。同じ細胞壁の異なる点で記録された4つの異なる力曲線のうち、0.1で記録された曲線はベースラインを示さず、カンチレバーの先端が細胞壁から分離されていないことを意味します。0.3で記録された曲線は、細胞壁の曲がりを示す接近部分の肩を示しています。ポイント2と4は、同様の弾性率値を持つ満足のいく力曲線を示しています。サンプルの準備と検査には練習が必要ですが、習得することはできます。このプロトコルの重要な部分は、結果の曲線をフィルタリングすることです。自動的に計算されたモジュライに頼らないでください。同じ手法を使用して、接着力またはエネルギー散逸を測定できます。また、一部の材料のビスコースまたは粘弾性挙動の説明にも重要です。
