放射性標識抗体(rAb)は、医学において様々な用途を有する。その大半は腫瘍学においてイメージングおよび治療薬として利用されているが、リウマチ関連炎症、心臓病学、および神経学のためのイメージング用途がある1。イメージングrAbsは、病変を検出するために高感度を有し、治療のための患者の選択を支援する可能性を秘めている2,3,4,5。それらはまた、それぞれの抗原に対する特異性のために治療に使用される。セラノスティックスとして知られる戦略では、同じrAbが画像化と治療の両方に使用されます6。

この方法は、その抗原に対する放射性標識抗体の結合親和性を決定することができる。また、結合の特異性も測定します。この簡単な方法の主な利点は、非結合親抗体からの干渉なしに放射性標識抗体のみの結合親和性の特異的特性評価である。この技術は、放射性標識抗体がその標的に結合する能力を定量的に検証する。これは、核イメージングやさまざまな疾患における核治療への応用にとって重要です。手順を実演するのは、私の研究室の大学院生アソシエイトであるErika Belitzkyです。まず、191ミリグラムの炭酸水素ナトリウムと23.9ミリグラムの炭酸ナトリウムを50ミリリットルの円錐管に加えて固定化バッファーを調製します。次に、40ミリリットルの18メガオームの水を加え、ボルテックスで溶解します。次に、総容量を50ミリリットルにする前にpHを9.0に調整します。次に、250ミリリットルのボトルに200ミリリットルのPBSおよび100マイクロリットルのTween 20を加えて、200ミリリットルの洗浄バッファーを準備する。また、50ミリグラムのBSAおよび25マイクロリットルのTween 20〜50ミリリットルのPBSを加えて50ミリリットルの結合緩衝液を調製し、ボルテックスによって混合する。50ミリリットルのPBSに1.5グラムのBSAを加えてブロッキングバッファーを準備し、ボルテックスを穏やかに混合する。抗原を固定化バッファーで希釈する。次に、壊れやすい96ウェルの平底プレートを取り、8×3アレイの24ウェルの各ウェルの底に100マイクロリットルの抗原を加えます。プレートをシーリングテープで覆い、摂氏4度で一晩インキュベートします。翌日、洗浄バッファーでプレートを3回洗浄する。流し台でプレートを勢いよくひっくり返して液体を処分し、ペーパータオルの山の上でプレートをタップして余分な液体を取り除きます。次いで、抗原を含むウェルに、多チャンネルピペットを用いてウェルあたり300マイクロリットルの洗浄緩衝液を添加する。1ウェルあたり300マイクロリットルのブロッキングバッファーを、96ウェルプレートの24抗原コーティングウェルおよび24ウェルの空ウェルの両方に加える。次いで、プレートを周囲温度で1時間インキュベートし、プレートの各ウェルを300マイクロリットルの洗浄緩衝液で3回洗浄した。放射性標識抗体の3倍の段階希釈を、プレート上のA〜Hとして指定された8列の結合緩衝液中で作る。まず、第1の濃度の1.2ミリリットル溶液を作るのに必要な放射性標識抗体のストックの体積を計算する。次に、B〜Hで標識した微量遠心チューブに800マイクロリットルの結合緩衝液を加え、Aとラベル付けした微量遠心チューブに必要な量の結合緩衝液を加えます。その後、ミニマイクロ遠心分離機を使用してスピンダウンし、チューブの底にあるすべての液体を収集します。次に、2つのAからチューブBに400マイクロリットルの液体を加え、ボルテックス処理後、ミニマイクロ遠心分離機を使用してスピンダウンします。同様に、溶液をチューブBからチューブCへ、CからDへ、チューブHまで、抗原のみで固定化した3つのウェルおよびBSAのみでブロックされた3つのウェルにウェル当たり100マイクロリットルの各希釈液を加える。次に、A標準物質をH標準物質に標識した微量遠心チューブに各希釈液100マイクロリットルを加え、これらのチューブを放射性標識抗体標準として保存し、ガンマカウンターでアッセイします。プレートを摂氏37度で1時間、穏やかな揺れでインキュベートします。原稿に記載されているように、各ウェルの微量遠心チューブにラベルを付けます。真空アスピレーターを用いて96ウェルプレートのウェルから放射性標識抗体を吸引する。次に、マルチチャンネルピペットを使用して、300マイクロリットルの洗浄バッファーを各ウェルに加えます。洗浄バッファーを吸引し、さらに4回洗浄を繰り返します。ウェルを適切な微量遠心チューブに分解します。H1、H2、H3、H3とマークされた抗原をA1、A2、A3に、次にBSAのみをH4、H5、H6とマークしたチューブ内の放射能をガンマカウンターを使用してA4、A5、A6にカウントします。放射性標識アミバンタマブは、EGFRおよびcMETタンパク質に特異的な結合を示した。同様に結合親和性は、EGFRおよびcMETタンパク質に結合したシングルアーム放射性標識ファブによっても保持された。飽和結合プロットは、放射標識抗体の濃度が低すぎると、線形曲線によって示されるように飽和が達成されないことを示した。放射標識抗体の濃度が高すぎると、水平プラトーが示すように、明確な対数成長のない飽和がもたらされました。また、高濃度の放射性標識抗体による高い非特異的結合が見られた。この手順は、各特異的放射性標識抗体の濃度範囲を最適化するために複数回の試行を要し得る。否定的な結果からのデータを使用して、将来の実験の設計に役立てます。結合親和性がわかれば、放射性標識抗体の特性評価の一環として、他のインビトロアッセイのベンチマークとして使用できます。
