脂質は、幅広い生物学的プロセスに関与しており、その構造的多様性に基づいて、脂肪酸、トリアシルグリセロール(TAG)、リン脂質、ステロール脂質、およびスフィンゴ脂質の5つのカテゴリーに大別することができます1。脂質の基本的な機能は、生物学的プロセス(すなわち、TAG)のためのエネルギー源を提供し、細胞膜(すなわち、リン脂質およびコレステロール)を形成することである。しかし、脂質の付加的な役割は、発生および疾患において注目されており、生物医学分野で広く研究されている。例えば、異なる長さの脂肪酸がユニークな治療的役割を有する可能性があることが報告されている。短い脂肪酸鎖は自己免疫疾患に対する防御機構に関与することができ、中長脂肪酸鎖は発作を緩和することができる代謝産物を産生し、長い脂肪酸鎖は代謝障害を治療するために使用できる代謝産物を生成する2。神経系では、グリア由来のコレステロールおよびリン脂質がシナプス形成に不可欠であることが示されている3,4。他のタイプの脂質は、薬物送達系において利用されるスフィンゴ脂質および免疫系を支持するために使用されるサッカロ脂質を含む、医療用途において有望であることが示されている5,6。生物医学分野における脂質の多くの役割と潜在的な治療用途により、リピドミクス(細胞脂質の経路と相互作用の研究)は重要かつますます重要な分野となっています。
リピドミクスは、分析化学を利用してリピドームを大規模に研究します。リピドミクスで利用される主な実験方法は、様々なクロマトグラフィーおよびイオン移動度技術と組み合わせた質量分析(MS)に基づいている7,8。この領域におけるMSの使用は、その高い特異性および感度、獲得の速度、および(1)低および一過性レベルでも生じる脂質および脂質代謝産物を検出する、(2)単一の実験で何百もの異なる脂質化合物を検出する、(3)これまで知られていなかった脂質を同定する、および(4)脂質異性体を区別するためのユニークな能力のために有利である。脱離エレクトロスプレーイオン化(DESI)、MALDI、および二次イオン質量分析(SIMS)を含むMSの発展の中で、MALDI MSIは、組織区画内の脂質の空間分布に関する独自の情報を提供することにより、従来のMSベースのアプローチを補完する強力なイメージング技術として浮上しています9,10。
リピドミクスの典型的なワークフローは、サンプル調製、質量分析技術を用いたデータ収集、およびデータ分析11からなる。サンプル中の脂質および代謝産物の研究は、生物における代謝プロセスの生理学的および病理学的状態を理解するための技術の出現につながった。生物学的相互作用を理解することは重要ですが、脂質および代謝産物の感受性は、色素または他の修飾なしにそれらを画像化および同定することを困難にする。代謝産物レベルまたは分布の変化は、表現型の変化をもたらし得る。メタボロミクスプロファイリングに使用されるツールの1つは、何百もの分子を同時に検出できるラベルフリーの in situ イメージング技術であるMALDI MSIです。MALDIイメージングは、サンプル中の代謝産物と脂質の完全性と空間分布を維持しながら、その可視化を可能にします。脂質プロファイリングの以前の技術では、放射性化学物質を使用して脂質を個別にマッピングしていましたが、MALDIイメージングはこれを放棄し、さまざまな脂質を同時に検出することを可能にします。
脂質代謝および恒常性は、神経系の維持および発達などの細胞生理学において重要な機能を果たしている。神経系脂質代謝の1つの重要な側面は、超低密度リポタンパク質(VLDL)、低密度リポタンパク質(LDL)、および高密度リポタンパク質(HDL)を含む分子キャリアリポタンパク質によって媒介されるニューロンとグリア細胞との間の脂質シャットリングである12。リポタンパク質は、脂質貨物の構造ブロックおよびリポタンパク質受容体のリガンドとして機能するApoBおよびApoDなどのアポリポタンパク質(Apo)を含む。脂質のニューロンとグリアのクロストークには、グリア由来のApoD、ApoE、ApoJなどの複数のプレーヤーと、それらのニューロンLDL受容体(LDLR)が関与します13,14。ショウジョウバエにおいて、ApoBファミリーのメンバーであるアポリポフォリンは、主要な血リンパ脂質担体15である。アポリポホリンは、哺乳類LDLR 15,16のホモログである2つの密接に関連したリポホリン受容体(LpRs)、LpR1およびLpR2を有する。以前の研究では、ヒトApoDのショウジョウバエのホモログであるアストロサイト分泌リポカリングリア・ラザリロ(GLaz)とそのニューロン受容体LpR1が、ニューロン - グリア脂質シャットリングを協調的に媒介し、樹状突起形態形成を調節することが発見された17。したがって、LpR1の喪失はショウジョウバエの脳内の全体的な脂質含量の減少を引き起こすと推測された。MALDI MSIは、この研究で実証されているように、LpR1-/-変異型および野生型ショウジョウバエ脳の小さな組織における脂質含有量をプロファイリングするための適切なツールとなる。
MALDI MSIの人気が高まっているにもかかわらず、この装置の高コストと実験の複雑さは、多くの場合、個々のラボでの実装を妨げます。したがって、ほとんどのMALDI MSI研究は、共有コア施設を使用して実施されています。MALDI MSIの他のアプリケーションと同様に、信頼性の高い結果を達成するためには、リピドミクスの慎重なサンプル調製プロセスが不可欠です。しかし、サンプルスライドの準備は通常、個々の研究所で行われるため、MALDI MSIの取得にばらつきが生じる可能性があります。これに対抗するために、この論文は、例として、正イオンモードの成体ショウジョウバエ脳の大群の脂質分析を用いたMALDI MSI測定前の小さな生物学的サンプルのサンプル調製のための詳細なプロトコルを提供することを目的としている11,17。しかしながら、いくつかのリン脂質クラスおよび大部分の小さな代謝産物は、以前に11に記載された負イオンモードでのMALDIイメージングによって良好に検出される。したがって、これら2つの研究例を用いて、自立型大組織対埋め込み小組織、融解マウント対ウォームスライドマウント、および正イオンモード対マイナスイオンモードの様々な組み合わせの詳細なサンプル調製プロトコルを提供したいと考えています。
