Dit protocol maakt de spontane zelforganisatie van microtubuli tactoïden mogelijk met behulp van een anti-parallelle cross-linker, MAP65. Dit systeem kan ons helpen de organisatie van microtubuli en biologische systemen zoals mitotische spindels te begrijpen. Het belangrijkste voordeel van deze techniek is dat het een minimalistisch systeem is dat spindelachtige vormen voor microtubuli kan recreëren die zeer reproduceerbaar en toegankelijk is voor veel laboratoria.
Deze methode is niet alleen van toepassing op cellulaire systemen, maar kan ook dienen als een model voor meso-schaal vloeibare kristalstudies. Om tubuline te bereiden, haalt u eerst een aliquot ongelabelde tubuline met één milligram gelyofiliseerde tubuline uit de vriezer van min 80 graden Celsius en houdt u deze op ijs. Voeg vervolgens 200 microliter koude PEM-80 toe.
Om al het lyofiel op te lossen, houdt u de buis 10 minuten op ijs. Haal vervolgens een tube rhodamine-gelabelde tubuline met 20 microgram gelyofiliseerd tubulinepoeder uit de vriezer van min 80 graden Celsius en bewaar het op ijs. Voeg vervolgens vier microliter koude PEM-80 toe en houd de buis gedurende 10 minuten op ijs om al het lyofiel op te lossen.
Zodra de gelyofiliseerde tubulinen zijn opgelost, voegt u 100 microliter van de geresuspendeerde ongelabelde tubuline-oplossing toe aan de vier microliter rhodamine-gelabelde tubuline-oplossing. Meng vervolgens de oplossingen door zes tot zeven keer heel langzaam te leidingen. Om de resterende 100 microliter ongelabelde tubuline-oplossing op te slaan, plaatst u eerst de buis in vloeibare stikstof om de oplossing te bevriezen en houdt u de buis vervolgens op min 80 graden Celsius voor toekomstig gebruik.
Verdeel vervolgens de tubulinemix in zeven nieuwe buizen door 15 microliter in elk toe te voegen. Vries de buizen in vloeibare stikstof zoals eerder aangetoond en bewaar ze bij min 80 graden Celsius voor toekomstig gebruik. Om stroomkamers samen te stellen voor het uitvoeren van experimenten, reinigt u eerst een glazen glijbaan met dubbel gedestilleerd water en droogt u deze met een pluisvrij laboratoriumdoekje.
Maak vervolgens de dia eerst schoon met ethanol, gevolgd door dubbel gedestilleerd water. Om een stroompad te maken, snijdt u een 40 tot 50 millimeter lang stuk dubbelzijdige tape. Splits het vervolgens tussen om twee dunnere stroken tape te maken en plaats de twee stroken op de dia vijf tot acht millimeter uit elkaar.
Plaats vervolgens verzande afdekslips bovenop het stroompad. Sluit vervolgens de dia af en deksel gleed de dubbelzijdige tapestrips af door zachtjes op het tapegebied te drukken met de achterkant van een pen. Als de afdichting goed is gemaakt, moet de tape van doorschijnend naar helder worden.
Om de extra tape aan de randen te verwijderen, snijdt u de tape met een scheermesje, waardoor u slechts één millimeter van de ingang van de stroomkamer overblijft. Label vervolgens de kamer met de juiste experimentele parameters Voor het uitvoeren van de tactoïde experimenten, ontdooi eerst alle benodigde reagentia op ijs en bewaar ze op het ijs tijdens het werk. Bedek vervolgens de stroomkamer met 20 microliter 5% niet-ionische blokcopolymeer oppervlakteactieve stof opgelost in PEM-80 met kleine druppels aan beide uiteinden van de kamer om de vorming van luchtbellen binnenin te voorkomen.
Houd vervolgens de stroomkamer in een vochtige kamer gemaakt van petrischaal met een nat, pluisvrij laboratoriumdoekje gedurende ten minste vijf tot zeven minuten. Meng vervolgens PEM-80, GMPPCPP, Pluronic F-127, dithiothreitol, glucose, polyethyleenglycol, het eerder gemaakte tubulinemengsel en MAP65 met GFP MAP65 voor visualisatie in een steriele buis door vijf tot zes keer te pipetteren, waardoor de buis op ijs blijft. Voeg vervolgens een microliter van een voorgemengde oplossing van glucoseoxidase en catalase toe aan de buis van het tubuline-MAP-mengsel en meng opnieuw door zeven tot acht keer te leidingen.
Verdeel het totale volume van de oplossing in twee porties voor gebruik in afzonderlijke kamers. Ondertussen moet de vloeistof die eerder aan de stroomkamer is toegevoegd, worden verwijderd door capillaire werking door een pluisvrij laboratoriumdoekje aan het andere uiteinde van de kamer te gebruiken, samen met de toevoeging van het tubuline-MAP-mengsel aan de stroomkamer. Zodra het monster volledig in de kamer is, sluit u de twee uiteinden van de kamer af met epoxy van vijf minuten en houdt u het ongeveer 30 minuten op 37 graden Celsius om te nucleeren en microtubule-tactoïden te laten groeien.
Voor het in beeld brengen van de tactoïden door fluorescentiemicroscopie, gebruikt u objectieven met een numeriek diafragma van 1,2 of meer in een vergroting van 60X of hoger om voldoende licht in fluorescentie te verzamelen. Neem de beelden op met een gratis metaaloxide halfgeleider of een camera van een opgeladen apparaat. Om het monster te behouden, bewaart u het in een omgevingskamer van 37 graden Celsius.
Als alternatief kunnen andere podiumverwarmers, waaronder heteluchttrapverwarmers en objectieve temperatuurgecontroleerde kragen met circulerend warm water, voor dit doel worden gebruikt. Aangezien geschikte excitatiebronnen essentieel zijn voor de juiste fluorescentie voor rhodamine tubuline, gebruikt u een laser van 561 nanometer met ten minste één milliwatt vermogen bij het monster voor beelden van goede kwaliteit. Verander voor GFP MAP65 echter de excitatiebron in een laser van 488 nanometer.
Als u breedveldeposeloscentiemicroscopie gebruikt, gebruik dan een rhodaminefilterkubus met een excitatie van 540 12,5 nanometer, dichroïsch van 545 nanometer 12,5 nanometer afsnijding en emissie van 575 nanometer long-pass. Gebruik voor GFP MAP een GFP-filterkubus met excitatie van 480 15 nanometer, dichroïsch van 505 nanometer 15 nanometer afgesneden en emissie van 515 nanometer long-pass. Nadat u ervoor hebt gezorgd dat het verlichtingsvermogen en de belichtingstijden zodanig zijn dat de intensiteitsschaal voor de camera niet verzadigd is, neemt u ten minste 10 afbeeldingen van verschillende gebieden om meer dan 100 tactoïden in zowel het rode als het groene kanaal in beeld te brengen en slaat u ze op als 16-bits TIFF-afbeeldingen voor analyse.
Met behulp van dit protocol kunnen microtubule tactoïden waarvan de vorming binnen 30 minuten wordt voltooid, direct onder de microscoop worden gevisualiseerd. De tactoïden zijn zichtbaar met zowel een 561-nanometer laser in het tubulinekanaal als een 488-nanometer laser in het MAP65-kanaal. De beelden overlappen elkaar ook perfect.
Bij deze methode konden ook de lengte en breedte van de tactoïden worden gemeten. Het intensiteitsprofiel van een tactoïde bleek te variëren met de breedte. Bovendien werd de immobiele aard van de microtubule tactoïden aangetoond door fluorescentieherstel na fotobleking of FRAP-experimenten, die geen herstel van fluorescentie na het fotobleken lieten zien.
Aan de andere kant onthulden FRAP-experimenten een mobiel karakter van MAP65, waarvoor een geleidelijk herstel en fluorescentie kon worden waargenomen na het bleken van foto's. Dit fluorescentieherstel door MAP65 kan worden aangepast aan een toenemend exponentieel verval om de amplitude en de tijdschaal van herstel te vinden. Het tactoïde experimentgedeelte moet binnen 10 tot 12 minuten worden voltooid, omdat de tubuline snel slecht kan worden op ijs, wat schadelijk kan zijn voor de nucleatie van tactoïden.
Toekomstig werk met verschillende microtubule cross-linking eiwitten en cross-linking motoreiwitten, enzymen die microtubuli kunnen verplaatsen, zal nieuwe informatie over de zelforganisatie van de mitotische spindel blijven blootleggen.
