DNA 트랜잭션을 이미지화하기 위해 미세유체 유동 세포는 높은 공간 및 시간 분해능으로 단일 분자 수준에서 제어된 측정을 허용하므로 중요한 역할을 합니다(그림 2). 미세유체라는 용어는 5-500 μm10,11 범위의 치수를 갖는 하나 이상의 채널에서 유체를 조작하는 능력을 의미한다. 스트림이라는 용어는 채널 내의 실제 유체를 의미하고 채널은 유체 스트림 또는 스트림이 이동하는 물리적 채널을 나타냅니다. 단일 채널 플로우 셀 설계에는 반응이 관찰되는 일반적인 물리적 채널이 있으며 일반적으로 하나의 유체 스트림만 존재합니다. 따라서 이러한 설계를 단일 스트림 유동 셀이라고 합니다. 대조적으로, 다중 스트림 유동 세포는 2개 이상의 진입 채널이 단일의 공통된 물리적 채널로 수렴하는 미세유체 장치로서 정의된다(도 2A). 공통 채널 내에서 개별 채널에서 시작된 유체 스트림은 서로 평행하게 흐르고 확산으로 인해 발생하는 최소한의 혼합만으로 분리된 상태로 유지됩니다(그림 2B). 대부분의 실험 설정에서 단일 펌프는 동일한 속도로 유체를 각 채널로 밀어 넣습니다. 대조적으로, 경계 조향이 사용되는 경우, 3 개 이상의 독립적으로 제어되는 펌프가 채널을 통해 유체를 밀어냅니다. 그러나 각 펌프는 다른 속도로 작동하지만 공통 채널의 순 유량은 일정합니다12. 이를 통해 펌프 속도를 변경하는 것만으로 메인 채널 구성 요소를 신속하게 교환 할 수 있습니다.
층류 외에도 또 다른 중요한 요소는 층류 유체 흐름 내의 포물선 속도 프로파일입니다. 가장 높은 유속은 하천 중앙에서 발생하고 가장 느린 유속은 표면 옆에서 발생합니다(그림 2C)13. 이 프로파일은 형광 DNA의 정확한 시각화와 정확한 단일 분자 분석을 위해 스트림에 고정된 비드에 부착된 DNA 분자를 완전히 늘리기 위해 고려되어야 합니다. 여기서 DNA는 B형으로 늘어나고 0pN의 힘으로 제자리에 고정됩니다. 이를 위해 광학 트랩 위치의 초점은 하단 커버슬립 표면에서 10-20μm 위치에 있어야 합니다(그림 2D).
