En fullstendig forståelse av intestinale parasitt-vert-interaksjoner og immunresponser mot helminthinfeksjon krever identifisering og analyse av de forskjellige cellepopulasjonene og effektormolekylene som er nøkkelen til induksjon av vevsremodellering og ormutvisning. Jordoverførte helminthinfeksjoner representerer et stort problem i utviklingsland over hele verden. Inntil nylig var imidlertid en protokoll som tillot analyse av sjeldne cellepopulasjoner tilstede i tynntarmen, hovedorganet som er berørt av denne infe...

Hakeorm er tarmparasitter som infiserer omtrent 700 millioner mennesker over hele verden, hovedsakelig i tropiske områder i underutviklede land. Høyintensitetsinfeksjoner med Ancylostoma duodenale og Necator americanus, de vanligste hookworm-parasittene hos mennesker, forårsaker anemi og proteinmangel som kan resultere i forsinket vekst og mental utvikling1. N. americanus og gnagerparasitten Nippostrongylus brasiliensis induserer en prototypisk type 2 immunrespons i verten og deler likheter i livssyklusen. Derfor er infeksjon av mus med N. brasiliensis den mest brukte modellen for humane hookworm-infeksjoner. Trinn 3 (L3) N. brasiliensis infeksiøse larver beveger seg fra huden til lungene i løpet av de første timene etter infeksjon. En gang i lungen blir de L4 og migrerer opp luftrøret for å bli svelget, passere gjennom magen og nå tarmen for å bli voksne (L5) innen 4-5 dager. I tarmen legger L5-ormer egg som skilles ut i avføringen for å starte parasittens livssyklus2.
Immunresponsen indusert av N. brasiliensis er preget av en økning i flere type 2 cytokiner, inkludert IL-4, IL-5, IL-9, IL-10 og IL-13, sammen med eosinofili, basofili, beger- og mastcellehyperplasi og økt IgG1- og IgE-produksjon. De fleste studiene som forsøker å identifisere og definere immunresponsene fremkalt ved N. brasiliensis-infeksjon, er sentrert på rollen som IL-4 eller IL-13 i denne modellen3. Imidlertid hadde identifiseringen og karakteriseringen av IL-9-uttrykkende celler og funksjonen til dette cytokinet i stor grad blitt oversett, inntil Licona-Limón et al. publiserte den første studien som demonstrerte en kritisk rolle for IL-9 i immunresponsen mot N. brasiliensis. Ved hjelp av reportermus beskrev denne studien T-celler (for det meste T-hjelper 9) og type 2 medfødte lymfoide celler (ILC2s) som de viktigste cellulære undergruppene som uttrykker IL-9 ved infeksjon4.
Isolering og karakterisering av immunceller fra helminthinfiserte lunger er mulig, og er omfattende rapportert 3,4. På grunn av den iboende vevsremodelleringen og slimproduksjonen viste det seg imidlertid å være en teknisk utfordring å gjøre det i den infiserte tarmen, inntil den nylige publiseringen av Ferrer-Font et al.5. Gruppen skisserte en protokoll for å isolere og analysere enkeltcellede suspensjoner av immunundergrupper fra Heligmosomoides polygyrus-infiserte murine tarmer. Basert på det har vi nå standardisert en protokoll for isolering og cytometrisk analyse av IL-9-uttrykkende lymfoide celler fra N. brasiliensis-infisert tarm. I tillegg har vi etablert IL-9 kinetikk fra forskjellige cellulære kilder og anatomiske steder gjennom hele infeksjonen.
Karakterisering av de forskjellige cellepopulasjonene som er involvert i denne infeksjonen er avgjørende for en bredere forståelse av immunresponsen mot parasitten og dens interaksjon med verten. Denne omfattende protokollen gir en klar rute for å isolere og analysere IL-9-produserende celler fra ønskede organer i sykdomsstadier av interesse, noe som muliggjør en kraftig forbedring av kunnskapen om rollen til disse cellene i N. brasiliensis-infeksjon og parasittinfeksjoner generelt.

IL-9 er et pleiotropisk cytokin assosiert med ulike prosesser, inkludert antitumorimmunitet, induksjon av allergiske patologier og immunresponsen mot helminthinfeksjoner, der den spiller en viktig rolle i utvisningen av parasitten. I en murinmodell av Nippostrongylus brasiliensis-infeksjon produseres IL-9 hovedsakelig av CD4+ T-lymfocytter og medfødte lymfoide celler som finnes i lunge, tynntarm og drenerende lymfeknuter. Mus ble subkutant injisert med 200 L3 stadium N. brasiliensis larver, eller med PBS for narrekontroll. Antall larver som ble brukt i denne protokollen ble justert for å isolere levedyktige celler fra lungene, lymfoidvevet og tynntarmen, i motsetning til tidligere rapporter hvor høyere belastninger av ormer ble brukt til å oppdage celler i lymfoide vev og lunger bare4. This protocol allows for the isolation and characterization of lymphoid cells expressing IL-9 in N.brasiliensis non-lymphoid infected tissues including the infected cut which recently was technically invisible.This protocol also provides an alternative to analyze populations of cells from peripheral tissues previously unappreciated in infection due to the intrinsic difficulties of isolating cells from these anatomical sites.This method allows the study of difficult-to-isolate populations from N.brasiliensis infected cut and could be applied to other helminthic infections, broadening our understanding of hookworm infections in humans.Demonstrating the procedure will be Ofelia Mu
