Ce protocole teste l’efficacité de la désinfection du lavage à l’eau chaude. Il peut aider à évaluer si une laveuse et une sécheuse conviennent au nettoyage des vêtements contaminés par un virus. Le principal avantage de cette technique de blanchiment est qu’elle peut être reproduite dans d’autres laboratoires à plus petite échelle en utilisant des procédures qui ont suivi le processus d’assurance qualité.
Ahmed Abdel-Hady, microbiologiste, Mariela Monge et Jonathan Sawyer, chercheurs scientifiques, et d’autres personnes de notre laboratoire, feront la démonstration de la procédure. Pour commencer, préparez la gélose au soja tryptique modifiée en pesant et en mélangeant les ingrédients dans de l’eau désionisée comme décrit dans le texte. Ensuite, ajoutez un millilitre de Phi6 aliquote concentré non dilué à une gélose molle et 100 microlitres d’une culture en phase logarithmique de pseudomonas syringae.
Ensuite, versez de la gélose molle sur la surface d’une plaque de gélose de soja tryptique modifiée solidifiée d’un diamètre de 100 millimètres. Évitez les bulles ou les déversements en faisant tourner doucement les plaques et en répartissant uniformément la gélose molle sur la surface de gélose solide. Après une nuit d’incubation à température ambiante, gratter délicatement le contenu des trois plaques avec un épandeur de cellules stériles dans un tube chronique stérile de 50 millilitres contenant 15 millilitres de tampon SM.
Tourbillonner les tubes au maximum pendant une à deux minutes pour casser la gélose puis les centrifuger pendant 15 minutes à 7 000 fois G.Ensuite, recueillir le surnageant et le filtrer à travers un filtre à seringue de 0,2 micron. Conserver le surnageant filtré sous forme d’aliquotes d’un millilitre dans des cryoviales à 80 degrés Celsius jusqu’à une utilisation ultérieure. Effectuer une évaluation visuelle préalable au test en mesurant et en enregistrant la longueur et la largeur des équipements de protection individuelle ou des EPI non lavés à divers endroits.
Pour l’inoculation, préparer une solution d’extrait de bœuf à 10% en dissolvant un gramme d’extrait de bœuf dans un volume total de 10 millilitres de 1X PBS. Filtrer stériliser tout le volume de la solution à l’aide d’un filtre à seringue de 0,2 micron. Ensuite, à l’intérieur de l’enceinte de biosécurité, inoculer les coupons d’essai et de contrôle positif avec environ 10 à septièmes unités formant des plaques par échantillon en pipetant une quantité de 10 microlitres de la solution sur l’EPI sous forme de gouttelettes multiples et en étalant la gouttelette à l’aide de l’extrémité de la pipette.
Réchauffer la solution de lavage à 50 degrés Celsius à l’aide d’une plaque chauffante et d’une barre d’agitation. Ensuite, mesurez et enregistrez le pH et la température de la solution de lavage et collectez 10 millilitres de solution pour le contrôle de la stérilité. Ensuite, versez 3,25 litres de solution de lavage dans une laveuse stérilisée.
Placez les articles d’EPI, un couvre-visage inoculé et quatre masques à film non contaminés sans coupons dans la laveuse. Après avoir lavé les EPI pendant 18 minutes, égoutter la laveuse et rincer trois fois avec cinq litres d’eau du robinet à température ambiante pour enlever la mousse. Ajoutez 3,25 litres d’eau du robinet stérilisée à température ambiante dans la laveuse et effectuez un cycle de rinçage de neuf minutes.
Ensuite, déplacez les EPI dans le côté d’essorage de la laveuse pour les faire tourner pendant cinq minutes. Sécher les EPI pendant 80 minutes à haute température de 93 degrés Celsius dans la sécheuse. Déplacez ensuite les EPI de la sécheuse vers l’espace de travail stérile.
Retirez de manière aseptique les coupons des articles inoculés et placez-les dans des tubes coniques. Ensuite, extrayez les coupons dans 10 millilitres de bouillon neutralisant à 10% Dey-Engley en tourbillonnant pendant deux minutes au réglage maximum de l’équipement. Pour plaquer les extraits à l’aide d’une méthode conventionnelle de recouvrement de gélose à toit souple, ajouter des aliquotes de l’échantillon d’essai dans le tube de gélose souple contenant six millilitres de gélose molle et 100 microlitres de seringues P en phase logarithmique.
Ensuite, versez la gélose molle sur la surface d’une plaque de gélose de soja tryptique modifiée solidifiée et répartissez uniformément la gélose molle sur la surface solide en faisant tourbillonner la plaque. Après une nuit d’incubation à température ambiante, énumérez manuellement les unités de plate-forme sur chaque plaque. Les tests systématiques en laboratoire pour évaluer l’efficacité du blanchiment de la désinfection virale à partir d’EPI ou de vêtements de taille normale ont montré que la numération de l’unité de plate-forme ou de la plaque PFU et le volume d’échantillon extrait permettaient de calculer le nombre d’unités de plate-forme par coupon de test.
Les résultats de la récupération virale représentative ont été obtenus pour un test de couvre-visage. Les tests de lavage ont fourni des informations sur les propriétés des matériaux telles que l’étirement ou le retrait des vêtements et les données de contrôle de la qualité du protocole. L’efficacité du lavage à l’eau chaude pour désinfecter les couvre-visages, le denim et les matériaux d’EPI de gommage de Phi6 a été démontrée dans ce graphique où les étoiles indiquent la désinfection complète.
Il est important d’utiliser des techniques d’asepsie tout au long de cette procédure pour prévenir la contamination croisée. De plus, les temps de séchage de l’inoculum pour les coupons sont uniques pour chaque matériau et sont établis avant les tests. D’autres méthodes complémentaires incluent la microscopie et les tests de pénétration de liquide pour explorer la dégradation des matériaux ou d’autres évaluations de taille garantissant que la protection de la barrière convient toujours au porteur.
Cette technique a servi de base à l’intensification de l’efficacité du blanchiment et des tests de matériaux pour les vêtements et les EPI à grande échelle. Il peut également aider à établir des procédures de blanchiment efficaces lors d’un autre événement de contagion ou d’une pandémie.
