2光子レーザー走査型顕微鏡は、in vivoで無傷の組織内の細胞のダイナミクスを観察するための強力なツールです。この技術の利点の1つは、標本がNIRによって励起されたときに、近赤外光(NIR)が可視光または紫外線よりも組織の浸透が深く、適切な条件下で組織に明らかな光損傷を引き起こさないことです5,6。実際、唾液腺は非常に均質で表在性の組織であり、表面の腺房細胞は腺表面からわずか約30μmしか離れていません7,8。生体内共焦点顕微鏡は、生きたマウス唾液腺における外分泌とアクチン細胞骨格を細胞内分解能で研究できることが示されています8。それにもかかわらず、2光子レーザー走査顕微鏡は、従来の共焦点顕微鏡の利点があるだけでなく、より深い組織や画像をより明確に検出するためにも使用できます。ここでは、傍細胞透過性トレーサーとして頻繁に使用され、サイズが異なるという利点を有する蛍光標識デクストランスを使用して、TJ細孔9の大きさを試験することができる。本研究では、マウスSMGの内皮バリア機能をその場で評価するために、生体内リアルタイム2光子レーザー走査顕微鏡技術を確立します。マウスSMGにおけるin vivo血管透過性検出のための各作業ステップは、現在のプロトコルに記載されている。マウスSMG管結紮モデルにおける内皮バリア機能の検出例である。
