Het is vastgesteld dat virussen de gastheerfactoren en -paden in hun voordeel afstemmen. Op hun beurt weerstaan de gastheercellen dat door verschillende strategieën toe te passen. Een van die strategieën is PANoptosis, die gastheercellen gebruiken als een antivirale strategie tegen virusinfecties. Virussen zoals IAV hebben echter hun eigen strategieën ontwikkeld om PANoptosis tegen te gaan en in hun voordeel te exploiteren 1,3,6</s...

Influenza A-virus (IAV) is het prototypische lid van de Orthomyxoviridae-familie en staat erom bekend wereldwijde epidemieën en onvoorspelbare pandemieën te veroorzaken. IAV veroorzaakt menselijke ademhalingsziekte, influenza, algemeen bekend als "griep". De griep is een acute ziekte die resulteert in de inductie van gastheer pro- en anti-inflammatoire aangeboren immuunresponsen en de dood van epitheelcellen in de menselijke luchtwegen. Beide processen worden beheerst door een fenomeen dat geprogrammeerde celdood1 wordt genoemd. De signalering voor geprogrammeerde celdood wordt geïnduceerd zodra verschillende pathogeenherkenningsreceptoren de binnenkomende virusdeeltjes in gastheercellen detecteren. Dit leidt tot de programmering van de dood van geïnfecteerde cellen en signalering aan de naburige gezonde cellen door drie onderling verbonden paden genaamd pyroptose, apoptose en necroptose - onlangs bedacht als één proces, PANoptosis1.
PANoptosis omvat de proteolytische verwerking van veel gastheer- en virale eiwitten van inductie tot uitvoering. Een dergelijke verwerking van eiwitten wordt voornamelijk geleid door een familie van cysteïneproteasen genaamd caspases 1,2. Er zijn maximaal 18 caspasen (van caspase 1 tot caspase 18) bekend3. De meeste caspasen worden uitgedrukt als pro-caspasen en geactiveerd door hun eigen proteolytische verwerking te ondergaan, hetzij door autokatalyse of andere caspasen4 als reactie op een stimulus zoals een virusinfectie. De PANoptosis van IAV-geïnfecteerde cellen werd beschouwd als een gastheerverdedigingsmechanisme, maar IAV heeft manieren ontwikkeld om het te ontwijken en te exploiteren om de replicatie ervan te vergemakkelijken 1,2,5,6. Een daarvan is om de gastheerfactoren te antagoniseren via caspase-gemedieerde splitsing of afbraak die inherent antiviraal zijn of interfereren met een van de stappen van de IAV-levenscyclus. Hiertoe zijn gastheerfactoren, cortactine, HDAC4 en HDAC6 ontdekt om caspase-gemedieerde splitsing of afbraak te ondergaan in IAV-geïnfecteerde epitheelcellen 7,8,9. De HDAC4 en HDAC6 zijn anti-IAV-factoren 8,10 en cortactine interfereert met IAV-replicatie in een later stadium van infectie, mogelijk tijdens virale assemblage en ontluikende11.
Daarnaast worden ook verschillende caspasen geactiveerd, die op hun beurt meerdere eiwitten splitsen om de ontstekingsreactie van de gastheer tijdens IAV-infectie te activeren 1,2. Bovendien ondergaan nucleoproteïne (NP), ionkanaal M2-eiwit van IAV 12,13,14 en verschillende eiwitten van andere virussen 3,15,16 ook caspase-gemedieerde splitsing tijdens infectie, wat de virale pathogenese beïnvloedt. Daarom is er een voortdurende behoefte om caspase-gemedieerde splitsing of afbraak van gastheer- en virale eiwitten tijdens IAV en andere virusinfecties te bestuderen om de moleculaire basis van virale pathogenese te begrijpen. Hierin worden de methoden gepresenteerd om (1) de splitsing of afbraak van dergelijke eiwitten door caspasen te beoordelen, (2) die caspasen te identificeren en (3) de splitsingsplaatsen te lokaliseren.

Caspases, een familie van cysteïneproteasen, orkestreert geprogrammeerde celdood als reactie op verschillende stimuli, waaronder microbiële infecties. Aanvankelijk beschreven als optredend door apoptose, is nu bekend dat geprogrammeerde celdood drie onderling verbonden paden omvat: pyroptose, apoptose en necroptose, samen bedacht als één proces, PANoptosis. Behandeling met caspase 3-remmer Er is ontdekt dat gastheer cortactine, HDAC4 en HDAC6 polypeptiden degradatie ondergaan als reactie op IAV-infectie in zowel honden (MDCK) als menselijke (A549, NHBE) cellen 7,8,9. Influenza A-virus (IAV) -infectie activeert de caspasen die gastheer en virale eiwitten splitsen, die op hun beurt pro- en antivirale functies hebben. Deze methode zal helpen om te bepalen en te bevestigen of een eiwit in met influenzavirus geïnfecteerde cellen afbraak of splitsing door caspasen ondergaat en de caspase-splitsingsplaatsen te identificeren. Deze methode vereist alleen elementaire moleculaire en cellulaire biologie laboratoriuminstellingen en onderzoeksvaardigheden en vereist geen gespecialiseerde apparatuur of softwaretraining. Deze methode kan worden aangepast om vergelijkbare doelstellingen te bereiken met betrekking tot infectie met andere dierlijke virussen of microben en blootstelling aan externe stimuli, zoals toxines en chemicaliën.Om te beginnen, zaad MDCK- of A549-cellen in een 12-well celkweekplaat. Incubeer de cellen 's nachts bij 37 graden Celsius onder een atmosfeer van 5% koolstofdioxide. Was de celpellet met 250 microliter fosfaatgebufferde zoutoplossing en centrifuge.Verwijder het supernatant en voeg 100 microliter cellysebuffer toe. Meng per vortex. Verwarm de buis op 98 graden Celsius gedurende 10 minuten om de cellen volledig te lyseren.Los vervolgens gelijke hoeveelheden eiwit op uit niet-geïnfecteerde en geïnfecteerde monsters door standaard SDS-polyacrylamidegelelektroforese. Breng na elektroforese de eiwitgel over naar een nitrocellulose- of PVDF-membraan en voer Western blotting uit. Vergelijk de eiwitniveaus in de met mock behandelde en met remmers behandelde geïnfecteerde monsterstroken.Verdun in 100 microliter geschikt medium, zoals Opti-MEM, 100 nanomolaren van controle- of caspase siRNA, of een geschikt volume transfectiereagens. Incubeer gedurende vijf minuten bij kamertemperatuur. Voer Western blotting uit en vergelijk de eiwitniveaus zoals eerder getoond. Zoek de caspase cleavage motieven XXXD en DXXD op het polypeptide.Muteer het P1-asparaginezuur, twee glutaminezuur of alanine door plaatsgerichte mutagenese en bevestig door DNA-sequencing. Verdun in 100 microliter van een geschikt medium twee microgram wild-type of mutant plasma-DNA of het aanbevolen volume van het transfectiereagens en incubeer de buizen gedurende vijf minuten bij kamertemperatuur. Meng 100 microliter plasma-DNA-oplossing met 100 microliter van de transfectiereagensoplossing en incubeer gedurende 20 tot 45 minuten bij kamertemperatuur.Splits in de tussentijd 300,000 MDCK- of A549-cellen en voeg deze toe aan 800 microliter compleet groeimedium. Voeg 200 microliter DNA-transfectiereagenscomplex toe en voeg deze suspensie toe aan een 12-well kweekplaat. Incubeer de plaat bij 37 graden Celsius onder een atmosfeer van 5% koolstofdioxide.Infecteer na 48 uur de cellen met influenza A-virus zonder remmers. Voer Western blotting uit en vergelijk de eiwitniveaus zoals eerder getoond. Met behulp van Western blotting werd de afbraak van gastheercoractine door influenza-geïnduceerde gastheer caspasen geëvalueerd.Behandeling van de met influenzavirus geïnfecteerde cellen met een caspase-3-remmer resulteerde in de redding van cortactine en virale NP-afbraak. Kwantificering van de intensiteit van cortactinebanden en de normalisatie ervan met de overeenkomstige actineband toonde aan dat het niveau van cortactine in met caspase-3-remmer behandelde geïnfecteerde cellen hoger was in vergelijking met onbehandelde geïnfecteerde cellen. In caspase-6 of caspase-7 uitgeputte cellen.Er werd geen significant herstel in cortactine polypeptide niveaus waargenomen, terwijl in caspase-3 uitgeputte cellen, cortactine hersteld van het influenzavirus geïnduceerde afbraak. Kwantificering en vergelijking van polypeptideniveaus toonde aan dat het cortiactinepolypeptideniveau in geïnfecteerde cellen met uitgeputte caspase-3-expressie hoger was dan cellen met normale caspase-3-expressie. De mutatie van asparaginezuur op positie 116 in glutaminezuur maakte het cortactinepolypeptide resistent tegen door influenzavirus geïnduceerde afbraak.Kwantificering en vergelijking van polypeptideniveaus toonde aan dat het niveau van mutant cortactinepolypeptide in geïnfecteerde cellen hoger was dan dat van wild-type cortactinepolypeptide. Het is vereist om tientallen mutanten te genereren en herhaalde Western blotting uit te voeren om de caspase-splitsingsplaats in het polypeptide te identificeren. Een in vitro biochemische assay met gezuiverde caspase en doelpolypeptide kan worden ontwikkeld om te bevestigen dat het eiwit van belang een direct op gas gebaseerd substraat is.Dit protocol zal helpen om de betekenis, de afbraak of splitsing van doeleiwit in het systeem dat wordt bestudeerd, zoals virusinfectie of een ziekte, te begrijpen.
