オリエンタルショウジョウバエ、バ クトセラ・ドルサリス は、グアバ、マンゴー、ユージニア属、スリナムチェリー、柑橘類、ビワ、パパイヤなど、150種以上の果物作物に被害を与える国際的な害虫種です1。広東省(中国)だけでも2億元以上の被害が見込まれています。成体の雌は、熟した果実または熟した果実の皮の下に卵を挿入し、果実の腐敗と脱落を引き起こし、果実の品質と作物の全体的な収量を低下させます2。成虫のショウジョウバエは飛行能力が高く、幼虫が果実の皮に穴を開けるため、害虫との直接接触を必要とする殺虫剤は野外ではあまり機能しません。さらに、殺虫剤の広範な使用により、さまざまな農薬に対する B.dorsalis の耐性が高まり、これらの有害な害虫の防除がさらに困難になっています3。したがって、効果的で環境に優しい害虫管理戦略の開発が切実に必要とされています。
近年、分子生物学的ツールやハイスループットシーケンシング技術の開発により、様々な害虫の重要な遺伝子(分子標的)の機能を対象としたRNAiなどの環境にやさしい害虫管理戦略の開発が試みられています。有害生物の生存と繁殖に重要な遺伝子は、機能遺伝子研究を通じて同定することができ、さらに、特異的に標的を絞った環境に優しい害虫管理ツールの改善のための潜在的な分子標的として役立つ可能性があります4。このような戦略を東洋のショウジョウバエ防除に適応させるためには、機能遺伝子研究のための効果的な方法が必要です。
CRISPR/Cas(クラスター化された規則的に間隔を空けた短い回文反復/CRISPR関連)エンドヌクレアーゼシステムは、細菌や古細菌で最初に発見され、バクテリオファージの感染によって導入されるような外来細胞内DNAの認識と分解に関与する適応メカニズムであることが判明しました5。II型CRISPRシステムでは、Cas9エンドヌクレアーゼは小さな関連RNA(crRNAおよびtracrRNA)によって誘導され、不法侵入DNAを切断し6,7,8、現在までに遺伝子編集に最も広く使用されているツールの1つになっています9,10,11,12。CRISPR/Cas9システムには、遺伝子サイレンシングの高効率や低コストなどのいくつかの利点があるため、ネッタイシマカ13,14、イナマカ渡り15、ボンビクスモリ16など、さまざまな昆虫種の遺伝子編集にすでに適用されています。B. dorsalisでは、CRISPR/Cas917,18,19を用いて、体色、翅二型、性決定に関する遺伝子のノックアウトに成功しています。しかし、この昆虫におけるCRISPR/Cas9の適用のための詳細な手順は不完全なままです。さらに、B. dorsalis遺伝子編集のために研究者によって提供されたいくつかのステップも多様であり、標準化が必要です。例えば、Cas9の形態は、公開された参考文献17,18,19では異なっていた。
