このプロトコルは、mRNA、rRNA、および長鎖ノンコーディングRNA(lncRNA)を含む細胞質および核長鎖RNAの単離に最適化されており、利用されるRNA精製カラムのサイズ選択性により、目的の他のRNAを単離するために変更することができます。以前は、mRNAやlncRNAなどの長鎖RNAの機能は、細胞内でのそれぞれの局在に大きく依存していました4,5。したがって、核から細胞内ドメインへの輸出の研究は、RNAまたは他の細胞成分の輸出が細胞に与えることができる役割を理解することに向けられています。lncRNAは、その翻訳とその後の効果が他の形態のRNA6との近接性と相互作用に大きく依存するため、この代表的な例として機能します。さらに、核領域と細胞質領域間の細胞要素の交換は、さまざまな癌治療に対する耐性メカニズムに関連しています7。細胞内コンパートメントの単離により、核輸送阻害剤の開発が可能になり、さまざまな治療法に対する耐性メカニズムの影響が減少しました8。
核RNAおよび細胞質RNAを分離した後、目的のRNAを精製するステップが実行される。RNA精製キットは実験室で一般的に見られ、長いRNAを精製および分離するために機能するため、このプロトコルの目的を十分に果たします。RNA精製では、1.8を超える260:280の比率を生成することが、RNAシーケンシングまたは高レベルの純度と単離を必要とするその他の同様の手順のサンプルの品質を確保するために重要です。不規則な260:280の値はフェノール汚染を示し、分離性が不十分で不正確な結果をもたらします9。
RNA精製が完了し、260:280の値が許容範囲を超えていることが確認されたら、qPCRを利用して核画分と細胞質画分の単離結果を検証しました。その際、関心領域に特異的なプライマーを使用して、核および細胞質分画レベルを実証しました。このプロトコルでは、MALAT1およびTUG1がそれぞれ核マーカーおよび細胞質マーカーとして使用された10。これらを組み合わせることで、MALAT1による高レベルの核分画の実証が可能になりますが、細胞質分画は低いと予想されます。逆に、TUG1を使用した場合、細胞質分画レベルは核分画レベルよりも高くなると予想されます。
このプロトコルを利用することで、細胞内での作用位置に基づいてRNAを単離することが可能であった。この実験では、多くの材料への広範なアクセスと溶解バッファーおよび密度ベースの遠心分離技術のみの利用により、他のRNAタイプおよび他の細胞成分への適用性が広まっています。これは、分離しなければ区別できない場所固有の発現イベントに光を当てることによって重要な情報を提供することができます。
