O dispositivo de avaliação da dor orofacial, ou OPAD, é uma ferramenta valiosa para a pesquisa pré-clínica da dor, pois usa um paradigma de conflito de recompensa operante. Isso engloba nocicepção, aprendizagem e motivação, que são importantes na experiência da dor humana e na melhoria da validade translacional. Uma vantagem significativa do OPAD sobre os ensaios tradicionais baseados em reflexos é que ele é totalmente automatizado, o que elimina a necessidade de restringir os animais, prevenindo os efeitos do estresse relacionados e também reduz o viés do experimentador.
Demonstrando a cirurgia CCI do nervo infraorbitário estará Darrice Montgomery, um técnico de laboratório do meu laboratório. Comece com a configuração do dispositivo de avaliação da dor orofacial, ou OPAD, colocando as bandejas de gotejamento de leite, gaiolas de plexiglass e grades de piso de metal. Fixe a fiação às gaiolas e coloque o porta-garrafas no poste de metal na parte de trás do dispositivo.
Em seguida, coloque as garrafas de leite de recompensa no porta-garrafas de uma forma que o bico possa ser alcançado pelo animal. Aperte o botão esquerdo do suporte para fixar o frasco no lugar. Em seguida, ligue o painel frontal e ligue as gaiolas.
Para configurar o protocolo, abra o software, digite a senha e clique em Log Me On ou pressione Enter. Clique em New Empty Experiment e no menu Protocol (Entrar). Em Aparelho, clique em Protocolo sem nome e selecione o modo que esse protocolo usará.
Em Modos Específicos do Equipamento, selecione Modo de Gaiola Mecânica OPAD e nomeie o protocolo. Depois de garantir que todas as gaiolas estejam ligadas, adicione as gaiolas OPAD clicando em Aparelho e Adicionar Item na parte superior do painel de protocolo, seguido de selecionar Nova Gaiola OPAD e Adicionar Todas as Gaiolas OPAD Conectadas. Para adicionar os estágios de teste do experimento, vá para a guia Teste para selecionar os estágios e as guias Primeiro estágio e nomeie o estágio.
Adicione 10 minutos como a duração do teste. Adicione mais estágios clicando em Adicionar Guia Item, seguido por Novo Palco. Em seguida, atribua os grupos clicando em Informações Adicionais e Grupos de Tratamento.
Marque Grupos de Tratamento Usados, em seguida, selecione a opção O usuário atribuirá manualmente os animais a seus grupos. Atribua identificações de animais clicando no menu Protocolo. Em Informações Adicionais, clique em Animal ID e marque a opção Usar Minhas IDs para Referir-se a Animais.
Defina o experimento digitando o título do experimento antes de clicar em Exibir Tratamentos e digitar os nomes do tratamento. Adicione animais e atribua os tratamentos e IDs de animais clicando em Exibir Animais e Adicionar Animais. Insira o número de animais a serem testados, seguido de OK.
Quando a lista de animais aparecer, adicione a identificação do animal e o tratamento para cada rato. Salve o protocolo clicando no menu Protocolo seguido de Salvar Protocolo. Digite o nome do arquivo e a senha do software e clique em Salvar.
Em seguida, salve o arquivo de experimento clicando no arquivo e, em seguida, salve, digitando a senha do software e clicando na guia Salvar. Depois de ligar as gaiolas, procure a luz verde na gaiola indicando que a gaiola está pronta para testar. Abra o arquivo de experimento salvo com um duplo clique.
Digite a senha, clique em Conectar-me ou pressione Enter. No lado esquerdo do ecrã, anote o número de animais e a gaiola correspondente, o estágio em funcionamento nesse dia e o estado do ensaio. Observe a identificação do animal de teste na tela da gaiola.
Coloque o rato na gaiola e pressione o botão na gaiola correspondente. Observe que a luz verde se transforma em uma luz laranja quando o teste é iniciado. Certifique-se de que o animal possa acessar a garrafa de recompensa enquanto faz contato suficiente com os picos mecânicos.
No lado direito da tela, observe o gráfico de cada animal mostrando o número de lambidas e contatos. Um som de aviso é ouvido quando a sessão de teste termina. Coloque o rato anestesiado em uma bancada cirúrgica e contenha-o.
Mantenha a temperatura do corpo a 37 graus Celsius usando uma almofada de aquecimento. Aplique pomada oftálmica nos olhos para evitar que eles sequem. Use a lâmina de bisturi 15 para fazer uma pequena incisão entre a gengiva dorsal e o lábio.
Remova o tecido mole usando a lâmina do bisturi e revele o ramo dos nervos infraorbitais, ou ION. Com a ajuda de uma agulha de seringa curvada contundente, coloque duas ligaduras intestinais cromáticas 5-O ao redor do ION e, em seguida, feche a ferida usando adesivo de tecido. As tentativas de contato e os dados de lambida de um único rato operado por CCI ION e Sham durante a sessão de teste padrão de 10 minutos no início do estudo, e duas semanas, quatro semanas e seis semanas após a cirurgia são apresentados aqui.
O modelo revelou que durante os períodos não nocivos os ratos têm longas sessões de bebida. Após a lesão, o animal não pode manter contato facial com as barras cravadas por um longo período, resultando em uma diminuição do número de lambidas. No grupo Sham, não foram observadas mudanças significativas para o período de consumo.
O número médio de lambidas entre os ratos operados por CCI ION e Sham no início do estudo não mostrou diferenças significativas. No entanto, o CCI ION reduziu significativamente o número de lambeduras até quatro semanas após a cirurgia e aumentou a latência para a primeira lambida durante a semana pós-cirurgia e uma semana após a lesão nervosa, indicando hiperalgesia mecânica. Não houve mudança significativa em ratos operados por Sham.
Ao usar o OPAD, otimize a distância da garrafa de leite e a largura dos picos. É importante manter isso consistente para reduzir erros durante a coleta de dados.
