記載されたプロトコルは、骨髄造血幹および前駆細胞ならびに骨髄間質細胞の表現型分析を可能にする。さまざまな環境でこれらの集団への摂動を研究するために使用できます。他の前述の方法と比較して、この技術は、造血細胞、特に2つの機能的骨髄領域、内膜および中枢骨髄、ならびに骨髄間質細胞の表現型分析を可能にする。まず、腹を上にして安楽死させた動物をペトリ皿に置き、70%エタノールをスプレーします。はさみを使用して腹部の上に切り込みを入れ、皮膚を足首まで引き離します。片方の足をつかみ、その底を切ってハサミを使って動物から切り離します。次に、足首を切って脚から足を外し、氷冷PBS 2%FBSに脚を置きます。次に、ピンセットを使用して腰の骨をつかみ、その後ろを切り、ハサミを使用して動物から分離します。股関節の骨を氷冷PBS 2%FBSに入れます。脚と腰の骨をペトリ皿に入れた後、滅菌メスを使用して骨をきれいにします。次に、メスで膝を切って脛骨と大腿骨を分離し、きれいな骨を氷冷PBS 2%FBSに入れます。大腿骨または脛骨を氷冷PBS 2%FBSで乳鉢に入れ、乳棒を使用して乳鉢の壁にそっと押し付けて骨を押しつぶします。次に、得られた細胞懸濁液を10ミリリットルのピペットを用いて上下にピペットで固定し、ホモジナイズし、チューブの上部に載置した40マイクロメートルのセルストレーナーを通して50ミリリットルのチューブに移した。さらにPBS 2%FBSで乳鉢をすすぎ、同じ50マイクロメートルのセルストレーナーを通して同じ40ミリリットルのチューブに溶液を移します。50ミリリットルのチューブを500 Gで摂氏4度で5分間遠心分離します。上清を捨てた後、数回上下にピペッティングして室温で5ミリリットルのRBC溶解バッファーに細胞ペレットを再懸濁する。室温で2分間インキュベートした後、15ミリリットルのPBS 2%FBSを添加し、50ミリリットルのチューブを遠心分離します。細胞ペレットが赤みを帯びていないように見える場合は、上清を廃棄し、ペレットを200マイクロリットルのPBS 2%FBSに再懸濁し、細胞懸濁液を96ウェルV底板のウェルに移して染色します。前に示したように骨を粉砕した後、はさみを使用してP1000ピペットの先端を切り取り、それを使用して乳鉢のすべての内容物を50ミリリットルのチューブに移します。次に、50ミリリットルのチューブを500 Gで摂氏4度で5分間遠心分離します。上清を捨てた後、ペレットを3ミリリットルのコラゲナーゼIVおよびこのディスパーゼII溶液に再懸濁する。チューブを摂氏37度で40分間インキュベートします。チューブをPBS 2%FBSで25ミリリットルに補充し、ボルテックスを加えて混合します。次に、内容物を100マイクロメートルのセルストレーナーを通して新しい50ミリリットルのチューブに移します。次に、古い50ミリリットルのチューブに15ミリリットルのPBS 2%FBSを追加し、100マイクロメートルのセルストレーナーを介して溶液を新しい50ミリリットルのチューブに移します。遠心分離後、上清を廃棄し、ピペットで数回上下にピペッティングすることにより、細胞ペレットを5ミリリットルのRBC溶解バッファーに再懸濁します。室温で2分間インキュベートした後、15ミリリットルのPBS 2%FBSを加える。ここでも、チューブを遠心分離し、上清を廃棄した後に細胞ペレットが赤みを帯びていないように見える場合は、ペレットを200マイクロリットルのPBS 2%FBSに再懸濁し、細胞懸濁液を染色のために96ウェルV底板のウェルに移します。次に、プレートを500 Gで摂氏4度で3分間遠心分離します。大腿骨をペトリ皿の上に置き、滅菌メスを使用して骨の端を切ります。次に、100マイクロリットルのPBS 2%FBSを、死虫のない0.5ミリリットルのインスリンシリンジを使用して骨の内側に両側から1回通過させ、1ミリリットルのPBS 2%FBSを含むマイクロ遠心チューブに溶液を回収することにより、骨の中央部を洗い流します。続いて、細胞懸濁液を、4ミリリットルのPBS 2%FBSを含むフラッシュ骨髄とラベル付けされた50ミリリットルのチューブに移す。乳棒を使用して乳鉢の壁にそっと押し付けて、氷冷PBS 2%FBSで乳鉢で骨の端を押しつぶします。細胞懸濁液が均質化されたら、40マイクロメートルの細胞ストレーナーを通して骨髄の破砕物とラベル付けされた50ミリリットルのチューブに移します。最後に、さらにPBS 2%FBSで乳鉢をすすぎ、溶液を同じチューブに移します。造血幹細胞および多能性前駆細胞のフローサイトメトリー解析を、健康な若年成人C57BL6Jマウスの全骨髄において行った。全骨髄中の間質細胞、内皮細胞、間葉系幹細胞の解析では、レプチン受容体の発現に基づいて間葉系幹細胞を容易に同定することができ、CD31およびSCA-1の発現に基づいて内皮細胞を同定することができる。ICAM1に基づく細動脈内皮細胞と類洞内皮細胞の識別を示す全骨髄中の内皮細胞のフローサイトメトリー分析。中枢および内皮骨髄組織における動脈および類洞骨髄内皮細胞の定量化は、動脈骨髄内皮細胞が内膜領域でより豊富であるのに対し、類洞は中枢骨髄領域でより頻繁であることを示しています。紅潮した骨髄を得るとき、PBS 2%FBSを骨の中央部の内側に通すために示された量または回数を超えてはならない。記載された方法を表現型的に分析された細胞集団の機能的アッセイと組み合わせることは、研究された条件下で起こるこれらの集団への潜在的な摂動についてのさらなる貴重な情報を提供するであろう。記載されたプロトコルは、内膜および中枢骨髄における造血細胞の表現型分析を可能にし、研究者がこれらの骨髄位置で特異的に発生する可能性のある造血への摂動を調査することを可能にする。
