ペルフルオロアルキル酸(PFAA)は、表面活性や熱的および化学的安定性などの優れた物理化学的特性により、さまざまな商業製品および工業製品に広く使用されています1,2,3。ペルフルオロオクタンスルホン酸(PFOS)とペルフルオロオクタン酸(PFOA)は、世界中で使用されている4,5,6つの最も重要なPFAAですが、これらの化合物はそれぞれ2009年と2019年の国際ストックホルム条約7,8に記載されています。PFOSとPFOAは、その持続性と広範な使用により、さまざまな環境マトリックスで広く検出されています。世界中のさまざまな河川や湖沼からの地表水中のPFOAとPFOSの濃度は、それぞれ0.15〜52.8 ng / Lと0.09〜29.7 ng / Lです9。地下水または再生水を灌漑に使用し、バイオソリッドを肥料として使用するため、PFOAとPFOSは土壌中に広く存在し、それぞれ0.01〜123μg/ kgから0.003〜162μg / kgの範囲であり10、植物に大量のPFAAを導入し、人間の健康に潜在的なリスクをもたらす可能性があります。農業用土壌および穀物(小麦およびトウモロコシ)中のPFAA(C4-C8)濃度は正の線形相関を示す11。したがって、植物内でのPFAAの蓄積と転座を調査することが不可欠です。

師部は植物から直接採取し、長距離にわたる有機化合物の転座を観察したが、植物苗から師部を取得することは困難である。比較的短期間の曝露中に、植物におけるPFAAの転座を研究するために、簡単で効果的な方法であるスプリットルート技術が導入されました。この方法は、植物における異なる化合物の長距離輸送を開示することもできる。標的PFAAの所定の濃度は、実際の環境での濃度よりも比較的高くなければならず、非添加溶液中の小麦中の標的PFAAを確実にモニタリングします。同様のサイズのTriticum aestivum、または小麦の種子を選択することから始め、8%過酸化水素水で15分間消毒します。消毒した種子を脱イオン水で徹底的にすすいでください。発芽のために室温で5日間暗所で湿ったろ紙の上に種子を置きます。均一なサイズの約9本の発芽苗を選択し、250ミリリットルの1/4強度のHoagland溶液を含むプラスチックビーカーに移します。処理の前に、生育室で22°Cで14時間、27°Cで10時間のサイクルで7日間苗を栽培します。苗栽培のために、それぞれ50ミリリットルの1/4強度のホーグランド溶液を含む2つの遠心分離管AとBを準備します。市販のパーフルオロオクタン酸(PFOA)とペルフルオロオクタンスルホン酸(PFOS)をメタノールに溶解し、原液として滅菌栄養液で希釈し、原液をPFOAとPFOS1リットルあたり100マイクログラムでチューブBに加えます。次に、小麦苗の根全体をピンセットで2等分し、根を同じ苗条に接続し、それぞれチューブAとBに挿入します。2本のチューブをアルミホイルで密封した後、インキュベーター内で22°Cで14時間、27°Cで10時間のサイクルで7日間培養します。7日後、小麦の苗を集め、パーフルオロアキル酸(PFAA)の添加溶液で培養した苗条や根などを非添加溶液でハサミで3つに分けます。植物サンプルを凍結乾燥機でマイナス55°Cで48時間凍結乾燥します。2日後、均質化する。根の重さを量り、サンプルを撃ちます。スパイク溶液サンプルと非スパイク溶液サンプルを収集します。15ミリリットルのポリプロピレンチューブに、2ミリリットルの炭酸ナトリウム緩衝液、1ミリリットルの硫酸水素テトラブチルアンモニウム、5ミリリットルのメチルtert-ブチルエーテルを加え、均質化した根または苗条を加える。終わったら、チューブを250 RPMで20分間振とうした後、室温で2000 gで10分間遠心分離して上清有機相を得た。2回目の抽出後、同様の処理抽出物をプールし、穏やかな窒素流で気化乾固させた後、5ミリリットルのメタノールで再構成し、約30秒間同じ速度を維持しながらボルテックスします。PestiCarbカートリッジを、メタノール中の5ミリリットルの0.1%水酸化アンモニウム、5ミリリットルの水、および5ミリリットルのメタノールで調整します。顔料を除去するためにPestiCarbカートリッジを通して5ミリリットルの抽出メタノール溶液を加える。カートリッジを5ミリリットルのメタノールで溶出し、サンプルチューブに集めます。回収した 10 ミリリットルのメタノール溶液を蒸発させて乾固し、ボルテックスする前に 200 マイクロリットルのメタノールで再構成し、室温で 20 分間 10 、 000 g でサンプルを遠心分離します。極性増強ポリマー抽出カートリッジを5ミリリットルのメタノールと5ミリリットルの水で調整して作動させます。1ミリリットルの添加溶液または50ミリリットルの非添加溶液サンプルをカートリッジを通して加えます。10ミリリットルのメタノールでターゲットPFAAを回避します。穏やかな窒素流で抽出物を蒸発させ、次に分析のために200マイクロリットルのメタノールで蒸発した抽出物を再構成します。分割根実験では、小麦中のPFAAの長距離輸送を調査しました。PFOAおよびPFOSは、添加溶液中にそれぞれ11.94ナノグラム/グラムおよび30.67ナノグラム/g乾燥重量の濃度でコムギ根に取り込まれ、5.01ナノグラム/グラムおよび4.17ナノグラム/グラム乾燥重量の濃度で芽に移されることが観察された。その結果,非添加溶液中で培養したコムギの根からPFOAおよびPFOSが検出されたが,それぞれ乾燥重量0.26ナノグラム/グラムおよび0.64ナノグラムの濃度であった。PFOAとPFOSは、シュートから根への師部を介した長距離輸送を経験する可能性があることが示唆されました。PFOAとPFOSは、それぞれ1リットルあたり17.8ナノグラムと28.5ナノグラム/リットルの濃度の非添加栄養溶液にも見られ、PFOAとPFOSが根のカスパリアンストリップを通過できることを示唆しました。チューブBの添加溶液がチューブAの非添加溶液を汚染しないように注意する必要があります.この方法の視覚的デモンストレーションは、小麦中のPFAAの長距離輸送を明らかにするための簡単な操作を提供できます。
