Om fysiologische /biochemische veranderingen in het materiaal te minimaliseren en bepaalde foto- en thermolabiele prenyllipidepigmenten te beschermen die een rijk bestanddeel van plastoglobulen vormen, is het van cruciaal belang om de isolatie uit te voeren bij 4 °C en beschermd tegen licht. Zoals hierboven aangegeven, worden de eerste stappen uitgevoerd in de koelcel onder een veiligheidslamp met behulp van een groenuitstralende gloeilamp. De volgende stappen die in het laboratorium worden uitgevoerd, zijn onder gedimd...

Het huidige begrip van de samenstelling en functie van plastoglobule is naar voren gekomen door middel van gedetailleerde proteomische en lipidomische studies 1,2,3,4,5. Dergelijke studies zijn enorm geholpen door een snelle en effectieve isolatiemethode die afhankelijk is van hun zeer lage dichtheid voor efficiënte scheiding met behulp van sucrosegradiënten. De eerste methoden voor plastoglobule-isolatie werden bereikt van soorten zoals de beukenboom (Fagus sylvatica), schotse bezem (Sarothamnus scoparius), ui (Allium cepa), spinazie (Spinacia oleracea), viooltje (Viola tricolor), peper (Capsicum annuum) en erwt (Pisum sativum)6,7,8,9,10,11 ,12,13. Een bijgewerkte methode om chloroplast plastoglobules op een efficiëntere en beter renderende manier te isoleren werd later gepresenteerd door Ytterberg et al.3,14. Hoewel we aanvankelijk werden gebruikt voor de studie van de plastoglobulen van Arabidopsis thaliana bladchloroplasten, hebben we deze bijgewerkte methode met succes gebruikt voor het gezonde bladweefsel van andere plantensoorten, zowel eenzaadlobbige als tweezaadlobbige, waaronder maïs (Zea mays), tomaat (Solanum lycopersicum), liefdesgras (Eragrostis nindensis), paars vals brome (Brachypodium distachyon) en wilde tabak (Nicotiana benthamiana ; niet-gepubliceerde resultaten). Bovendien is de isolatiemethode met succes aangepast aan de plastoglobulen van cyanobacteriën, waaronder Synechocystis sp. PCC 6803 en Anabaena sp. PCC 712015, en het uitgedroogde bladweefsel van de opstandingsplant, E. nindensis.
Chloroplast plastoglobulen van gezond bladweefsel zijn fysiek verbonden met de thylakoïde membranen16. Ondanks deze fysieke continuïteit behouden de twee chloroplast-subcompartimenten verschillende lipide- en eiwitsamenstellingen, hoewel de gereguleerde uitwisseling van lipide en eiwit tussen de twee compartimenten is voorgesteld 2,4,17,18,19. In feite is onlangs een interessant hemifusiemodel voorgesteld voor de handel in neutrale lipiden tussen chloroplasten en cytosol19. Vanwege de fysieke continuïteit van plastoglobulen en thylakoïden begint de isolatiemethode met gezond bladweefsel met het verzamelen van een gepelletiseerd ruw thylakoïdepreparaat, dat vervolgens wordt gesoniseerd om de plastoglobulen van de thylakoïden te scheiden, wat in tegenstelling is tot methoden die worden gebruikt voor het isoleren van cytosolische lipidedruppels20 . Ultracentrifugatie op een sucrosekussen drijft vervolgens de plastoglobulen met lage dichtheid omhoog door de sucrose, waardoor ze effectief worden gescheiden van de thylakoïden, kernen en ander materiaal met hoge dichtheid. Plastoglobulen in cyanobacteriën, evenals die van uitgedroogd bladweefsel, bestaan daarentegen blijkbaar in vivo in een vrij zwevende vorm. Vandaar dat hun isolatie direct op een sucrosegradiënt drijft. Dit artikel demonstreert de isolatiemethode van gezond bladweefsel en demonstreert verder twee variaties die kunnen worden gebruikt om plastoglobulen te isoleren uit uitgedroogd bladweefsel of cyanobacteriële culturen, waardoor de fysiologische breedte en evolutionaire context waarin plastoglobules kunnen worden bestudeerd, aanzienlijk wordt uitgebreid.
Geïsoleerde plastoglobulen kunnen vervolgens worden gebruikt voor een willekeurig aantal downstream-analyses om moleculaire kenmerken te onderzoeken. We hebben de geïsoleerde plastoglobulen uit A. thaliana bladweefsel gebruikt voor uitgebreide proteomische en lipidomische analyse onder verschillende omgevingsomstandigheden of genotypen, wat de selectieve modificatie van eiwit- en lipidesamenstelling in aanpassing aan stress 2,4,21,22 aantoont. Daarnaast zijn in vitro kinase-assays uitgevoerd die transfosforyleringsactiviteit geassocieerd met geïsoleerde plastoglobules aantonen22, zijn de oligomere toestanden van eiwitcomponenten onderzocht met behulp van native gel-elektroforese 21 en zijn protease-scheertests uitgevoerd23.
Het belangrijkste voordeel van deze methode is de relatieve snelheid van de procedure. Onze ervaring is dat de onderstaande protocollen binnen ongeveer 4 uur volledig kunnen worden voltooid. Er is een alternatieve methode beschreven om plastoglobulen uit bladweefsel te isoleren, waardoor de gelijktijdige isolatie van andere chloroplast-subcompartimentenmogelijk is 24. Deze alternatieve methode biedt een aantal duidelijke voordelen wanneer kwantitatieve vergelijking met de andere chloroplast subcompartimenten noodzakelijk of gewenst is. Deze alternatieve methode is echter ook vervelender en zal een aanzienlijk lagere opbrengst van geïsoleerde plastoglobulen uit vergelijkbare hoeveelheden bladweefsel opleveren. Wanneer een gerichte studie van plastoglobules het doel is, is de hier geschetste methodologie de optimale keuze. Niettemin kunnen tijdens de monstervoorbereiding totaal blad- en ruwe thylakoïde-aliquots worden verzameld, en het wordt ten zeerste aanbevolen om referentiemonsters te hebben voor latere vergelijking.

plastoglobule lipid droplet isolation from plant leaf tissue and cyanobacteria

Plastoglobule lipidedruppels zijn een dynamisch subcompartiment van plantaardige chloroplasten en cyanobacteriën. Alomtegenwoordig gevonden onder fotosynthetische soorten, wordt aangenomen dat ze een centrale rol spelen bij de aanpassing en remodellering van het thylakoïde membraan onder snel veranderende omgevingsomstandigheden. Het vermogen om plastoglobulen van hoge zuiverheid te isoleren heeft hun studie aanzienlijk vergemakkelijkt door middel van proteomische, lipidomische en andere methodologieën. Met plastoglobulen van hoge zuiverheid en opbrengst is het mogelijk om hun lipide- en eiwitsamenstelling, enzymatische activiteit en eiwittopologie te onderzoeken, naast andere mogelijke moleculaire kenmerken. Dit artikel presenteert een snel en effectief protocol voor de isolatie van plastoglobulen uit chloroplasten van plantenbladweefsel en presenteert methodologische variaties voor de isolatie van plastoglobulen en gerelateerde lipidedruppelstructuren uit maïsbladeren, het uitgedroogde bladweefsel van de opstandingsplant, Eragrostis nindensis, en de cyanobacterium, Synechocystis SP. PCC 6803. Isolatie is afhankelijk van de lage dichtheid van deze lipiderijke deeltjes, wat hun zuivering door sucrosedichtheidsflotatie vergemakkelijkt. Deze methodologie zal waardevol blijken bij de studie van plastoglobulen van diverse soorten.

De isolatie van zeer zuivere plastoglobulen is van cruciaal belang voor hun studie. Een snel en effectief protocol voor hun isolatie dat hun downstream biochemisch onderzoek vergemakkelijkt, wordt hier gepresenteerd. Het belangrijkste voordeel van deze methode is de relatieve snelheid van de procedure en het aanpassingsvermogen aan tal van plantensoorten en omgevingsomstandigheden.Bij het soniceren van thylakoïde resuspensaties moet bijzondere aandacht worden besteed aan voldoende kracht om plastoglobulen uit thylakoïden te verwijderen en bij de daaropvolgende extractie van plastoglobulen uit sacharosegradiënt. Dr. Elsinraju Devadasu, een postdoctoraal onderzoeker in mijn laboratorium, en Febri Susanto, een promovendus in mijn laboratorium, zullen deze procedure demonstreren. Begin met het verwerven van drie weken oude zaailingen van zes gezonde maïs, met een V5-groeistadium en een gewicht van ongeveer 120 gram.Knip alle bladeren aan de basis van de stengel af en dompel ze snel in een ijsbad voordat je ze naar de koelcel transporteert. Werk onder een groene veiligheidslamp, verwijder de maïsbladeren uit het ijsbad en knip ze met een schaar in kleinere stukjes van vijf bij vijf centimeter. Maal voorzichtig en grondig de helft van het geknipte bladweefsel in 350 milliliter van de maalbuffer op een niveau lager dan zeven op een commerciële blender.Start en stop de blender vijf tot zes keer om ervoor te zorgen dat alle bladeren worden gesneden. Filtreer het homogenaat door een laag nylondoek van 25 micron op een grote trechter tot vier centrifugeflessen van 250 milliliter. Nadat u de mengstap met het resterende geknipte bladweefsel hebt herhaald, verdeelt u het filtraat gelijkmatig over de vier flessen.Verwijder een aliquot van het bladfiltraat en leg het apart om te worden bewaard als een representatief totaal bladmonster. Centrifugeer vervolgens het filtraat gedurende zes minuten bij 1, 840 G en vier graden Celsius. Giet het resulterende supernatant af en resuspenseer met zachte wervelende bewegingen van de borstel de pellet in 12 milliliter medium R 0,2, met 0,2 molaire sucrose.Nadat u de pellet in één fles hebt geresuspendeerd, voegt u de suspensie toe aan de volgende fles en herhaalt u de resuspensie om de suspensies in één fles te bundelen. Verdeel de gepoolde suspensie over zes ultracentrifugebuizen van drie milliliter, waarbij een maximaal volume van 2,5 milliliter in elke buis wordt bereikt. Gebruik vervolgens de tip sonicator met een amplitude van 100% om elke buis vier keer gedurende 10 seconden te soniceren.Zorg ervoor dat de sonicatorhoorn ondergedompeld blijft en uit de buurt van het vloeibare oppervlak om schuimvorming te voorkomen. Beweeg de sonicatorhoorn langzaam op en neer in de ophanging tijdens elke ronde. Terwijl u de vier buizen afwisselt, brengt u elke buis na elke ultrasoonapparaat terug naar een ijsemmer om het monster te laten afkoelen.Als u klaar bent, centrifugeert u de gesonificeerde ruwe thylakoïde suspensie bij 150.000 G gedurende 30 minuten bij vier graden Celsius. Scheer het kussenoppervlak af met een spuit en een naald van 22 gauge en oogst het resulterende gele en olieachtige drijvende pad van ruwe plastoglobules uit het sucrosekussen. Herstel ongeveer 500 microliter uit elke buis en deponeer ze in een buis van 2,0 milliliter.Verzamel ruwe thylakoïde aliquots voor en na de ultrasoonapparaat en afgifte van plastoglobulen. Als u klaar bent, gaat u verder met de verwerking van het plantenweefsel of slaat u de ruwe plastoglobulen op bij min 80 graden Celsius en zuivert u ze later. Kweek 50 milliliter synechocystis-soorten PCC 6802-cultuur in BG11-media gedurende 7 tot 10 dagen om de stationaire fase te bereiken.Stel met behulp van een spectrofotometer de celdichtheid in op een optische dichtheid van 2,0 op 750 nanometer. Om de polysacchariden te verwijderen, centrifugeer 50 milliliter cultuur en verwijder het supernatant. Herhaal het wassen van de cellen in 50 milliliter buffer A twee keer.Resuspendeer de gewassen pellet in 25 milliliter buffer A en breek de cellen met behulp van een Franse drukcel op 1, 100 pond vierkante inch. Herhaal het proces drie keer in koude omstandigheden totdat de gelyseerde kleur verandert van groen naar rood, blauw, groen onder wit licht. Verwijder het aliquot van het celhomogenaat en leg het apart om te worden bewaard als een representatief totaalcelmonster.Verdeel het resulterende homogenaat in acht ultracentrifugebuizen van drie milliliter gevuld tot maximaal 2,5 milliliter in elke buis. Leg dit homogenaat over elkaar met 400 microliter medium R, waardoor een stapgradiënt ontstaat. Balanceer de buizen zorgvuldig door indien nodig extra medium R toe te voegen voordat u de buizen centrift.De thylakoïde en andere zwaardere organellen, inclusief polyhydroxyalkanoaatlichamen, vertonen pelletvorming, terwijl plastoglobulen een gele olieachtige pad vormen op of nabij de bovenkant van de sucrosegradiënt. Oogst de resulterende drijvende pad van ruwe plastoglobules met een spuit, zoals eerder aangetoond, en deponeer de oogst in een buis van twee milliliter. Schraap plastoglobulen van de zijkant van de ultracentrifugebuiswand indien nodig met een naaldpunt.Ga verder met de verwerking van cyanobacteriën of bewaar ruwe plastoglobulen bij min 80 graden Celsius en zuiver later. Om de zuivere plastoglobulen te oogsten met behulp van de verwerkingsmethode voor plantenweefsel, bereidt u de sucrosegradiënt die eerder is beschreven met behulp van 500 microliter ruwe plastoglobulen, 400 microliter medium R 0,2 en 400 microliter medium R.Na centrifugatie oogst u het resulterende drijvende pad van pure plastoglobules in een buis van twee milliliter. Na het aliquoteren van de zuivere plastoglobules en het flashvriezen van de aliquots in vloeibare stikstof, bewaar je ze bij min 80 graden Celsius of lyofiliseer je ze tot een droog poeder.Om zuivere plastoglobulen uit cyanobacteriën te oogsten, creëert u een sucrosegradiënt, zoals eerder vermeld, met behulp van 500 microliter van de ruwe plastoglobulen, 750 microliter medium R 0,7 en 750 microliter medium R 0,2. Verzamel na centrifugatie de zuivere plastoglobulen in een buis van 1,5 milliliter en aliquot de zuivere plastoglobulen. Als u klaar bent, bevriest u de zuivere plastoglobules aliquots in vloeibare stikstof en slaat u ze direct op bij min 80 graden Celsius of lyofieliseert u tot een droog poeder.Bij deze methode werd een aanzienlijke hoeveelheid plastoglobule of lipidedruppelmateriaal gezien dat op de bovenste laag van het sucrosekussen dreef. Na daaropvolgende centrifugatie werden gezuiverde plastoglobulen verkregen op of nabij het oppervlak van de sucrosegradiënt. Na de succesvolle isolatie van plastoglobules toonden immunoblots ontwikkeld voor zuiverheidsbeoordeling met behulp van antilichamen tegen arabidopsis thaliana fibrilline 1a en arabidopsis thaliana fotosysteem II subunit D1 aan dat de fibrilline homologen voornamelijk geassocieerd waren met thylakoïden dan de plastoglobulen.Om de efficiënte en geconcentreerde extractie van de zwevende pad te garanderen, plaatst u de naaldopening van de spuit net onder het vloeibare oppervlak van de buffer terwijl u volledig ondergedompeld bent en trekt u vervolgens voorzichtig omhoog. Geïsoleerde plastoglobulemonsters zijn vatbaar voor latere proteomische of lipidomische studies en zijn gebruikt om de veranderingen in de beoogde eiwit-lipidesamenstelling onder verschillende omgevingsomstandigheden aan te tonen. Deze techniek heeft nieuwe studies van de plastoglobule vergemakkelijkt, waaronder de ontdekking van geassocieerde eiwitkinaseactiviteit en de aanwezigheid van oligomere eiwitcomplexen door middel van daaropvolgend biochemisch onderzoek.
