Patogenesen af CNS-sygdomme er stadig ikke fuldt ud forstået, hvilket forhindrer udviklingen af nye terapier og lægemidler. Centralnervesystemet (CNS) sygdomme med høj sygelighed, såsom neurodegenerativ sygdom, traumatisk hjerneskade, hypoxi-induceret hjerneskade i stor højde og iskæmisk slagtilfælde, er afgørende årsager til den voksende dødelighed på verdensplan 1,2,3. Realtidsovervågning af cytokiner og proteinændringer i specifikke hjerneområder bidrager til diagnostisk nøjagtighed af CNS-sygdomme og hjernefarmakokinetiske undersøgelser efter medicinering. Traditionel videnskabelig forskning anvender hjernevævshomogenat eller en manuel indsamling af interstitiel hjernevæske fra dyr til påvisning af specifikke stoffer og til farmakokinetiske undersøgelser. Dette har dog nogle mangler, såsom en begrænset stikprøvestørrelse, manglende evne til dynamisk at observere ændringer i indikatorer og ujævn stikprøvekvalitet 4,5,6. Cerebrospinalvæske, en interstitiel væske, beskytter hjernen og rygmarven mod mekanisk skade. Dens sammensætning er forskellig fra serumets på grund af eksistensen af blod-hjerne-barrieren (BBB)7. Direkte analyse af cerebrospinalvæskeprøver er mere befordrende for at afsløre mekanismen for CNS-læsioner og lægemiddelopdagelse. Uundgåeligt har cerebrospinalvæskeprøverne, manuelt opnået direkte fra cisterna magna og cerebrale ventrikler gennem en sprøjte, ulemper ved blodforurening, en tilfældig chance for prøveindsamling, usikkerhed om mængde og næsten ingen mulighed for multipel hentning 8,9. Mere bemærkelsesværdigt kan konventionelle interstitielle hjernevæskeprøveudtagningsmetoder ikke få prøver fra beskadigede hjerneområder, hvilket hindrer udforskningen af patogenesen af CNS-sygdomme i specifikke hjerneområder og effektivitetsevalueringen af målrettede etnomedicinbehandlinger 9,10.
Hjernemikrodialyse er en teknik til prøveudtagning af interstitiel hjernevæske hos vågne dyr11. Mikrodialysesystemet efterligner vaskulær permeabilitet ved hjælp af en sonde implanteret i hjernen. Mikrodialysesonden er bevæbnet med en semipermeabel membran og implanteres i specifikke hjerneområder. Efter perfusion med isotonisk kunstig cerebrospinalvæske (ACSF) kan den dialyserede interstitielle hjernevæske med fordel opsamles med fordelene ved små prøvestørrelser, kontinuerlig prøveudtagning og dynamisk observation12,13. Med hensyn til placering kan hjernemikrodialyseprober selektivt implanteres i hjernestrukturer eller kraniale cisterner af interesse14. En observation af unormale niveauer af et endogent stof i hippocampus ekstracellulær væske (HECF) antyder forekomsten af CNS-sygdomme eller patogenesen af sygdom. Flere undersøgelser har vist, at biomarkørerne for CNS-sygdomme, såsom D-aminosyrer i skizofreni, β-amyloid- og tau-proteiner i Alzheimers sygdom, neurofilament-lyskæder i traumatisk hjerneskade og ubiquitincarboxy-terminal hydrolase L1'er i hypoxisk iskæmi encephalopati, kan analyseres i cerebrospinalvæske15,16,17 . En kemisk analysemetode baseret på hjernens mikrodialyseprøveudtagningsteknik kan bruges til at overvåge dynamiske ændringer af eksogene forbindelser såsom aktive ingredienser i etnomedicin, som diffunderer og distribuerer i specifikke hjerneområder14.

real-time dynamic collection of hippocampal extracellular fluid from conscious rats using a microdialysis system

En række sygdomme i centralnervesystemet (CNS) er forbundet med ændringer i sammensætningen af hippocampus ekstracellulær væske (HECF). Imidlertid har vanskeligheder med at opnå HECF i realtid fra bevidste rotter længe begrænset evalueringen af CNS-sygdomsprogression og effektiviteten af etnomedicinsk terapi. Det er opmuntrende, at en hjernemikrodialyseteknik kan anvendes til kontinuerlig prøveudtagning med fordelene ved dynamisk observation, kvantitativ analyse og en lille prøveudtagningsstørrelse. Dette muliggør overvågning af ændringer i det ekstracellulære væskeindhold for forbindelser fra traditionelle urter og deres metabolitter i hjernen hos levende dyr. Formålet med denne undersøgelse var således nøjagtigt at implantere en cerebrospinalvæskemikrodialysesonde i hippocampusområdet hos Sprague Dawley (SD) rotter med et tredimensionelt hjernestereotaksisk apparat, der afskærer molekylvægte større end 20 kDa. HECF af høj kvalitet blev derefter opnået fra bevidste rotter ved hjælp af et mikrodialyseprøvetagningskontrolsystem med en justerbar prøveudtagningshastighed fra 2,87 nL / min - 2,98 ml / min. Afslutningsvis giver vores protokol en effektiv, hurtig og dynamisk metode til at opnå HECF hos vågne rotter ved hjælp af mikrodialyseteknologi, som giver os ubegrænsede muligheder for yderligere at udforske patogenesen af CNS-relaterede sygdomme og evaluere lægemiddeleffektivitet.

Efter ovennævnte forsøgsprotokol og prøvetagningsparametrene i tabel 1 blev vandlignende, farveløs og gennemsigtig rotte-HECF opnået ved den indstillede prøvetagningshastighed (figur 1K). Det osmotiske tryk af den opnåede rotte HECF var 290-310 mOsm/L, hvilket indirekte kan sikre kvaliteten af prøverne18,19.

Hjernemikrodialyse teknisk opnår dynamisk og kontinuerlig inkvisition af interstitiel hjernevæske i specifikke hjerneområder hos ikke-bedøvede rotter. Efter 60 minutters ligevægt i mikrodialyse HECF-opsamlingssystemet indsamles HECF med jævne mellemrum.Efter prøvetagning afsluttes, tage hjernen mikrodialyse sonde ud af sonde kateteret. Dyp sonden i deioniseret vand og skylning med deioniseret vand i 12 timer for at fjerne strandede saltaflejringer fra rørledningen og sonden. Fjern sonden og læg den i en 0,05% tripsinopløsning ved fire grader Celsius.Hippocampus ekstracellulær væske erhvervet ved hjernens mikrodialyseteknik er vandlignende, farveløs og gennemsigtig ved den indstillede prøveudtagningshastighed. De vigtigste procedurer er at implantere mikrodialysen omhyggeligt i CA1-regionen og indsætte sonden i popkateteret. Mikrodialyseteknikken gør det muligt at hjælpe den dynamiske ændring af sygdomsmarkører i specifikke hjerneområder, stoffet i lægemidlets effektivitet og vævskatabolisme af lægemiddelkomponenter.
