慢性神経因性疼痛は、糖尿病、感染症、神経圧迫、自己免疫疾患など、体性感覚系の病変や疾患に起因します3。神経因性疼痛は、末梢および中枢の両方の感作メカニズムに依存しており、神経の病変に由来します。この痛みは、触覚および熱誘発痛覚過敏および異痛症の両方、進行中の痛み、および患肢の温度の変化によって特徴付けることができます4。メカニズムをよりよく理解し、新しい治療法を進歩させるために、神経因性疼痛の症状と原因を模倣するためにげっ歯類でいくつかのモデルが開発されました5。例えば、神経因性疼痛は、化学療法剤注射、脊髄神経結紮術(SNL)、坐骨神経の慢性狭窄損傷(CCI)、pSNL、温存神経損傷、坐骨神経切断、および坐骨神経三分割6で誘発することができる。特に、坐骨神経の結紮は、機械的および熱的過敏症、または複合性局所疼痛症候群(CRPS)に特徴的な患肢の温度変化など、ヒトで観察される神経因性疼痛の複数の特徴を再現します7。したがって、このモデルは、慢性神経因性疼痛を誘発するCRPSまたはその他の神経損傷の影響の研究に適しています。このモデルは、1990年にセルツァーによって最初に開発され8、新規鎮痛化合物を調査したり、慢性疼痛の認知効果を評価したりするための疼痛研究で広く使用されています9,10,11,12,13。このモデルは高い再現性を示し、部分結紮は末梢刺激に対する行動応答を保存する6。
現在使用されているモデルの多くには、pSNLでは観察されない欠点があります。CCIモデルは、収縮器のぴったり性に応じて各動物間の損傷の変動性がはるかに高く、解剖術は後足の指を変化させ、モデルを行動分析に適さないものにします6。SNLモデルは、はるかに複雑で長い手術であり、高度な技術スキルを必要とするだけでなく、重度の運動障害のリスクも高くなります3。これらの欠点は、pSNL モデルでは見られません。再現性の容易さ、手術期間の短さ、および術後に見られる運動障害のリスクの低下により、このモデルは末梢神経障害性疼痛の研究に価値があります8,14。それにもかかわらず、部分結紮手順自体は実験者間でばらつきがあり、その結果、結紮された神経線維の数の一貫性が低下します。したがって、手術の詳細を提示することは、研究間の再現性を高めるために重要です。
