만성 신경병증성 통증은 당뇨병, 감염, 신경 압박 또는 자가면역 질환을 포함한 체감각계의 병변 또는 질병으로 인해 발생합니다3. 신경병성 통증은 말초 및 중추 감작 메커니즘에 의존하며 신경의 병변에서 비롯됩니다. 이 통증은 촉각 및 열 유발 통각과민과 이질통, 지속적인 통증, 영향을 받은 사지의 온도 변화 등을 특징으로 할 수 있다4. 기전을 더 잘 이해하고 새로운 치료법을 발전시키기 위해, 설치류에서 신경병증성 통증의 증상과 원인을 모방하는 몇 가지 모델이 개발되었다5. 예를 들어, 신경병증성 통증은 화학요법제 주사, 척추 신경 결찰술 (SNL), 좌골 신경의 만성 수축 손상 (CCI), pSNL, 보존된 신경 손상, 좌골 신경 절제술 및 좌골 신경 삼부로 유도될 수 있다6. 특히, 좌골 신경의 결찰은 기계적 및 열적 과민증 또는 복합 부위 통증 증후군(CRPS)의 특징인 영향을 받은 사지의 온도 변화와 같이 인간에서 관찰되는 신경병성 통증의 여러 특징을 재현합니다7. 따라서, 이 모델은 만성 신경병증성 통증을 유도하는 CRPS 또는 임의의 다른 신경 손상 애정의 연구에 매우 적합하다. 이 모델은 1990 년 Seltzer에 의해 처음 개발되었으며 8, 새로운 진통제 화합물을 조사하거나 만성 통증의 인지 효과를 평가하기 위해 통증 연구에서 널리 사용됩니다9,10,11,12,13. 이 모델은 높은 재현성을 나타내며 부분 결찰은 말초 자극에 대한 행동 반응을 보존합니다6.
현재 사용되고 있는 모델 중 상당수는 pSNL에서 관찰되지 않는 단점을 가지고 있습니다. CCI 모델은 수축기의 아늑함에 따라 각 동물 간의 손상 변동성이 훨씬 더 높으며, 자가절제술은 뒷발을 변경하여 모델을 행동 분석에 부적합하게 만든다6. SNL 모델은 훨씬 더 복잡하고 긴 수술로 고급 기술이 필요할 뿐만 아니라 심각한 운동 장애의 위험이 높습니다3. 이러한 단점은 pSNL 모델에서는 볼 수 없습니다. 재현성의 용이성, 짧은 수술 기간, 수술 후 나타나는 운동 결손의 위험 감소로 인해 이 모델은 말초 신경병성 통증을 연구하는 데 유용합니다 8,14. 그럼에도 불구하고, 부분 결찰 절차 자체는 실험자들 사이에서 가변성을 가질 수 있으며, 그 결과 결찰된 신경 섬유의 수의 일관성이 떨어질 수 있습니다. 따라서 수술의 세부 사항을 제시하는 것은 연구 간의 재현성을 높이는 데 중요합니다.
