タイムラプス蛍光顕微鏡は、出芽酵母における減数分裂染色体分配のダイナミクスを研究するための貴重なツールです1,2。出芽酵母細胞は、重要な栄養素の飢餓を通じて減数分裂を起こすように誘導することができます3。減数分裂の間、細胞は1ラウンドの染色体分配を受け、続いて2つの分裂を経て、胞子にパッケージ化された4つの減数分裂産物を作成します(図1)。個々の細胞は減数分裂の各段階で視覚化でき、固定細胞イメージングでは簡単に見逃すことができる空間的および時間的データを生成します。このプロトコルは、タイムラプス蛍光顕微鏡法と、以前に確立された2つの方法、誘導性NDT80システム(NDT80-in)およびアンカーアウェイ技術を組み合わせて、異なる減数分裂段階における特定のタンパク質の機能を研究する方法を示しています。
NDT80-inシステムは、中減数分裂転写因子NDT80 4,5の誘導発現に依存する減数分裂細胞周期同期のための強力なツールです。NDT80発現は、前期I出口6,7に必要である。NDT80-inシステムでは、NDT80は、エストロゲン受容体(Gal4-ER)に融合したGal4転写因子を発現する細胞において、GAL1-10プロモーターの制御下にあります4,5。Gal4-ERはβ-エストラジオールに結合した場合にのみ核に入るため、NDT80-in細胞はβ-エストラジオールの非存在下で前期Iで停止し、前期Iの細胞の同期を可能にします(図1)。β-エストラジオールの付加は、Gal4-ER転写因子の核への転座を促進し、そこでGAL1-10に結合してNDT80の発現を促進し、減数分裂への同期的な侵入をもたらします。タイムラプス顕微鏡は同期なしで実行できますが、同期を使用する利点は、細胞が減数分裂の特定の段階にある間に阻害剤または薬物を追加できることです。
アンカーアウェイ技術は、ラパマイシン8を添加することでタンパク質を核から枯渇させることができる誘導可能なシステムである。この技術は、酵母細胞が核膜が破壊されない閉鎖有糸分裂および減数分裂を経験するため、出芽酵母の細胞分裂中の核タンパク質の研究に理想的です。さらに、この技術は、減数分裂全体を通して複数の機能を有するタンパク質にとって非常に有用である。欠失、変異対立遺伝子、減数分裂ヌル対立遺伝子とは異なり、特定の段階で核から標的タンパク質を除去しても、初期段階での標的タンパク質活性が損なわれないため、結果をより正確に解釈できます。アンカーアウェイシステムは、リボソームの成熟時に発生する核と細胞質の間のリボソームサブユニットの往復を利用します8。核から標的タンパク質を枯渇させるために、リボソームサブユニットRpl13AがFKBP12でタグ付けされている株において、標的タンパク質はFKBP12-ラパマイシン結合ドメイン(FRB)でタグ付けされる。ラパマイシンがなければ、FRBとFKBP12は相互作用せず、FRBタグ付きタンパク質は核内に残る。ラパマイシンを加えると、ラパマイシンはFKBP12およびFRBと安定な複合体を形成し、複合体はRpl13Aとの相互作用により核からシャトルされます(図1)。ラパマイシン添加時の細胞死を防ぐために、細胞はTOR1遺伝子のtor1-1変異を保持している。さらに、これらの細胞には、S. cerevisiae FKBP12タンパク質のヌル対立遺伝子であるfpr1Δが含まれており、内因性Fpr1がFRBとラパマイシンの結合に関してRpl13A-FKBP12を凌駕するのを防ぎます。アンカーアウェイバックグラウンド変異であるtor1-1およびfpr1Δは、減数分裂のタイミングや染色体分配に影響を与えません2。
この技術の有用性を実証するために、動原体タンパク質Ctf19は減数分裂を通して異なる時点で枯渇した。Ctf19は、有糸分裂には欠かせませんが、減数分裂における適切な染色体分配に必要な動原体の成分です9、10、11、12、13。減数分裂では、動原体は前期Iで脱落し、Ctf19は動原体再組み立てに重要です9,14。このプロトコルでは、NDT80-inシステムと細胞を同期させ、アンカーアウェイ技術を使用して、前期Iからの放出の前後で、および減数分裂後のI染色体分配後に標的タンパク質Ctf19を核から枯渇させました(図1)。このプロトコルは、減数分裂および有糸分裂の任意の段階で目的の他のタンパク質を枯渇させるように適合させることができる。
