타임랩스 형광 현미경은 신진 효모 1,2에서 감수 분열 염색체 분리의 역학을 연구하는 데 유용한 도구입니다. 신진 효모 세포는 주요 영양소의 기아를 통해 감수 분열을 겪도록 유도 될 수 있습니다3. 감수 분열 동안 세포는 한 번의 염색체 분리 후 두 번의 분열을 거쳐 포자로 포장되는 4 개의 감수 분열 생성물을 만듭니다 (그림 1). 감수 분열의 각 단계에서 개별 세포를 시각화 할 수 있으며, 이는 고정 세포 이미징으로 쉽게 놓칠 수있는 공간 및 시간 데이터를 생성합니다. 이 프로토콜은 타임 랩스 형광 현미경을 이전에 확립된 두 가지 방법인 유도성 NDT80 시스템(NDT80-in) 및 앵커 어웨이 기법과 결합하여 별개의 감수 분열 단계에서 특정 단백질의 기능을 연구하는 방법을 보여줍니다.
