그람 음성균(Gram-negative Pseudomonas syringae)은 학문적, 경제적 중요성 때문에 전형적인 식물 병원성 박테리아이다3. P. syringae의 수명주기는 물 순환4과 관련이 있습니다. P. syringae는 기공이나 상처와 같은 자연적인 구멍을 통해 식물의 잎 아포플라스트인 중엽세포(mesophyll cell) 사이의 세포간 공간으로 들어간다5. 일단 아포플라스트에 들어가면, P. syringae는 III형 분비계(T3SS)와 III형-전위 이펙터(T3E)에 의존하여 식물 면역을 억제하고 병원체의 이익을 위해 식물 세포 기능을 조작한다6. T3SS 및 T3E의 발현은 표적 유전자7의 프로모터 영역에서 hrp-box 모티프에 결합하는 대체 시그마 인자인 마스터 조절인자 HrpL에 의존한다.
관심 유전자의 하류에 있는 형광 단백질 유전자에 염색체 위치 전사 융합체를 생성함으로써, 단일 세포 수준8에서 방출되는 형광 수준을 기반으로 유전자 발현을 모니터링할 수 있습니다. 이 방법을 사용하여 hrpL의 발현은 실험실에서 자란 박테리아 배양과 식물 아포플라스트 8,9에서 회수된 박테리아 개체군 내에서 이질적이라는 것이 확인되었습니다. 단일 세포 수준의 유전자 발현 분석은 일반적으로 실험실 배지에서 성장한 박테리아 배양에서 수행되지만, 이러한 분석은 식물 내에서 자라는 박테리아 개체군에 대해서도 수행될 수 있으므로 자연적 맥락에서 하위 개체군 형성에 대한 귀중한 정보를 제공할 수 있습니다. 식물에서 추출한 박테리아 개체군 분석에 대한 잠재적인 한계는 아포플라스트에 주사기 압력 침투에 의한 고전적인 접종 방법에 이어 잎 조직의 침용에 의한 박테리아 추출이 일반적으로 다운스트림 분석을 방해하는 다량의 세포 식물 파편을 생성한다는것입니다 10. 대부분의 세포 파편은 GFP 형광과 겹치는 엽록체의 자가형광 단편으로 구성되어 있어 오해의 소지가 있는 결과를 초래합니다.
본 프로토콜은 2개의 모델 병리학에서 단세포 유전자 발현 이질성을 분석하는 과정을 설명한다: 하나는 P. syringae pv에 의해 형성된다. 토마토 균주는 DC3000 및 애기장대 탈리아나 (Col-0)이고, 다른 하나는 P. syringae pv에 의한 것이다. phaseolicola 균주 1448A 및 콩 식물 (Phaseolus vulgaris 품종 Canadian Wonder). 진공 챔버와 펌프를 이용한 진공 침투를 기반으로 한 접종 방법을 제안하여 잎 전체에 빠르고 손상없이 침투하는 방법을 제안합니다. 또한, 기존 프로토콜을 개선한 것으로, 주사기 내에서 소량의 부피를 사용하여 양압 및 음압 주기를 적용하여 아포성형액 추출을 기반으로 조직 파괴를 크게 줄이는 아포플라스트에서 박테리아 집단을 추출하는 더 부드러운 방법이 사용됩니다.
