다공성 매질에서 생물막의 성장은 모공을 막아 소위 생물 막힘을 유발할 수 있습니다. 생물막 발달은 다공성 매질의 두 기둥을 분리하는 거리로 정의되는 유체 흐름 및 기공 크기에 의해 제어됩니다 8,9,10. 기공 크기와 유체 흐름은 모두 영양소 수송과 국소 전단력을 제어합니다. 차례로, 성장하는 생물막은 기공을 막고, 유체(11,12,13)의 속도 분포, 질량 수송, 및 다공성 매질(14,15)의 수압 전도도에 영향을 미친다. 유압 전도도의 변화는 밀폐 시스템(16,17,18,19)의 압력 증가를 통해 반영됩니다. 생물막 발달 및 생물막힘에 대한 현재의 미세유체 연구는 균질한 기하학적 구조(즉, 단일 기공 크기) 또는 이질적인 다공성 매체(12,21,22)에서 유속의 영향을 연구하는 데 중점을 둡니다. 그러나 생물막 발달에 대한 유속 및 공극 크기의 영향과 그에 따른 생물 막힌 다공성 배지의 압력 변화를 풀기 위해서는 다양한 다공성 배지 형상 및 환경 조건을 병렬로 연구할 수 있는 고도로 제어 가능하고 다재다능한 실험 플랫폼이 필요합니다.
본 연구는 압력 측정과 다공성 매질 내에서 진화하는 생물막의 동시 이미징을 결합한 미세유체 플랫폼을 소개합니다. 채널 형상 설계의 가스 투과성, 생체 적합성 및 유연성으로 인해 폴리디메틸실록산(PDMS)으로 만들어진 미세유체 장치는 다공성 매질에서 생물막 발달을 연구하는 데 적합한 도구입니다. 미세유체공학은 물리적, 화학적 조건(예를 들어, 유체 흐름 및 영양소 농도)을 매우 정밀하게 제어하여 미생물 서식지의 환경을 모방할 수 있게 한다23. 또한, 미세유체 장치는 광학 현미경을 사용하여 마이크로미터 해상도로 쉽게 이미지화할 수 있고 온라인 측정(예: 국부 압력)과 결합할 수 있습니다.
