열충격 단백질 90(Hsp90)은 암의 발병 및 진행을 담당하는 단백질의 안정성을 유지하는 분자 샤페론입니다. 또한, 항종양제에 대한 내성 발달을 담당하는 단백질도 Hsp90의 고객이다1. Hsp90은 전체 단백질의 2% 미만을 구성할 수 있는 정상 세포와 비교하여 모든 암세포 유형(세포 단백질의 >90%)에서 유비쿼터스로 과발현됩니다. 더욱이, 암세포의 Hsp90은 공동 샤페론과 복합체에 존재하는 반면, 정상 세포에서는 주로 자유롭고 복합체가 없는 상태로 존재합니다 2,3. 최근 몇 년 동안, 몇몇 Hsp90 억제제는 시험관 내 및 생체 내 연구에서 세놀리틱 효과를 갖는 것으로 입증되었으며, 여기서 마우스의 수명을 크게 향상시켰습니다 4,5,6. 앞서 언급한 모든 발견은 Hsp90 억제제가 부작용이 적고 내성 발생 가능성이 감소하면서 여러 암 유형에 효과적일 수 있다는 사실을 입증합니다. 말라카이트 그린 분석법은 무기 인산염의 검출을 위한 간단하고, 민감하며, 빠르고, 저렴한 절차로, 원하는 표적에 대한 화합물의 자동화 및 고처리량 스크리닝(HTS)에 적합하다11. 이 분석은 HTS 12,13,14,15,16,17뿐만 아니라 소규모 실험실 규모 설정에서 Hsp90 억제제의 스크리닝에 성공적으로 사용되었습니다. 이 분석은 Hsp90의 ATPase 활성으로 인해 형성된 유리 무기 인산염을 결정하는 비색법을 사용합니다. 이 정량화의 기초는 유리 인산염과 몰리브덴 사이에 인산염 분해 화합물이 형성되는 것이며, 이는 이후 말라카이트 그린과 반응하여 녹색을 생성합니다(그림 1). 이 빠른 색 형성은 분광 광도계 또는 플레이트 리더에서 600-660 nm18,19 사이에서 측정됩니다.
본 프로토콜에서, 효모 Hsp90을 이용한 말라카이트 그린 분석을 수행하는 절차 및 샤페론에 대한 억제제의 후속 확인이 기재되어 있다. Hsp90의 약물성이 처음으로 확립된 천연물 분자인 겔다나마이신(geldanamycin, GA)은 진정한 억제제로 간주되었다10. HTS는 테스트를 위해 많은 수의 분자를 사용할 수 있기 때문에 현재 약물 발견 프로그램의 필수적인 부분이 되었습니다. 이 기술은 Covid-19 감염 치료를 위한 약물의 용도 변경이 시급하기 때문에 지난 2년 동안 더 중요해졌습니다20,21. 따라서, 말라카이트 그린 분석법을 채택하여 효모 Hsp90 단백질에 대한 분자의 HTS에 대한 상세한 개요가 제시된다.
