L’activité métabolique, qui peut être définie comme la somme de toute l’activité de l’organisme qui implique de l’énergie, est essentielle pour comprendre la fonction et l’évolution de la vie sur terre. Sur les récifs coralliens, les mesures du métabolisme sont vraiment importantes pour décrire des modèles tels que la symbiose, ou le processus entre un hôte et, par exemple, le symbiote Symbiodiniaceae à l’intérieur du tissu hôte corallien. Et cela peut nous dire non seulement comment l’organisme fonctionne normalement, mais aussi comment cet organisme fonctionne lorsque des facteurs de stress climatiques sont en jeu, par exemple.
Pour commencer l’installation de l’équipement et du corail, connectez les deux plaques de bain-marie à l’aide d’un polypipe bleu et de connecteurs. Assurez-vous que la plaque du moteur est visible à travers les plaques transparentes du bain-marie lorsque les chambres de respirométrie ne sont pas en place. Ensuite, connectez la base du bain-marie à la plaque moteur de base avec les engrenages magnétiques avant de connecter cet ensemble à une source d’alimentation et de mettre l’appareil sous tension.
Pour assembler la chambre de respirométrie, ajoutez la perle magnétique dans la chambre en verre et placez le flux en plastique opaque à travers la base du support dans la chambre en verre. Placez fermement les chambres dans les bains-marie, en vous assurant que les chambres en verre sont en contact avec l’eau à température contrôlée pour l’expérience. À l’aide des boutons de commande de la vanne, modulez le débit d’eau selon vos besoins, en appliquant un débit continu et doux réglé à 75 litres par heure, avec une agitation lente à 30 tours par minute.
Procédez à la connexion des câbles à fibre optique d’oxygène avec les points du capteur d’oxygène, qui détectent et transmettent les signaux de la chambre via le câble. Assurez-vous de connecter la fibre optique aux ports correspondants du module d’oxygène. Insérez les câbles dans les trous percés sur le côté des chambres du couvercle, en assurant le contact avec les taches.
Si nécessaire, fixez le ruban de plomberie blanc, fin et auto-obturant pour que le câble soit bien ajusté et pour le laisser rester fermement à l’intérieur de la chambre à eau. Assurez-vous que le corail individuel peut être vu avec les tentacules bruns tournés vers le haut à l’intérieur de la chambre. Enfin, mesurez la respiration à l’aide du logiciel de mesure de l’oxygène.
Après avoir importé les données de respirométrie à l’aide du package R respR, la fonction d’inspection a été utilisée pour vérifier les anomalies courantes telles que les valeurs non numériques ou manquantes, et tracer la série chronologique de l’oxygène et calculer un taux glissant. Le taux mobile est devenu stable après le point de 3000. La baisse de l’oxygène n’est devenue détectable qu’après la ligne 200 de la série à temps plein.
La fonction de taux de soulignement automatique, qui identifie les régions linéaires des données, a été utilisée pour extraire les taux. Une région linéaire d’environ 3 000 à 5 000 a été identifiée comme la région la plus linéaire. La fonction adjust_rate a été utilisée pour déterminer les taux de fond des expériences de contrôle.
Enfin, les taux ont été convertis en unités de sortie souhaitées à l’aide de divers paramètres. Le taux spécifique de surface, qui est le taux absolu divisé par la surface de l’échantillon, a été obtenu en sortie. Les résultats de la respiration dans l’obscurité à la petite taille de l’échantillon testé ont indiqué l’efficacité de cette méthode pour capturer le seuil de signal bas.
De plus, les valeurs représentatives de la respiration pour les blancs, ainsi qu’une comparaison de contrôle par rapport à la température élevée pour de très petits échantillons ont été obtenues. Malgré l’importance, il y a un manque de méthodes, et donc un manque de données sur les taux métaboliques clés liés aux récifs coralliens, en particulier en ce qui concerne la progéniture corallienne, qui est souvent de très petite taille, ce qui la rend difficile à mesurer. C’est pourquoi nous avons développé ce système de respirométrie physiologique très flexible et peu coûteux conçu pour mesurer les taux de respiration, c’est-à-dire les taux métaboliques, chez de très petits organismes et, dans ce cas, chez les juvéniles coralliens.
