さまざまな細菌の形は、17世紀にアントニーファンレーウェンフックによって最初に注目されました1。細菌は古来より球体から枝分かれ細胞まで多種多様な形で存在してきました2。細胞の形状は、細菌分類学者が各細菌種を記述および分類するための基本的な条件であり、主にグラム陽性門とグラム陰性門3の形態学的分離のために。細菌の細胞形態を決定するいくつかの要素が知られており、そのすべてが細胞壁および膜の構成要素として、ならびに細胞骨格において細胞カバーおよび支持体に関与している。このようにして、科学者たちは、細菌の形状を定義する遺伝子のクラスターによって定義される細菌細胞の形態を決定することに関係する化学的、生化学的、物理的メカニズムとプロセスをまだ解明しています2,4。
さらに、科学者たちは、この細胞の形状が細胞の重要なパラメータで最適に見えるため、桿体の形状が細菌細胞の祖先の形態である可能性が高いことを示しました。したがって、球菌、らせん状、ビブリオ状、糸状、およびその他の形態は、さまざまな環境への適応と見なされます。実際、特定の形態は独立して何度も進化しており、細菌の形状が特定の環境に適応している可能性があることを示唆しています3,5。しかし、細菌の細胞のライフサイクルを通じて、細胞の形状が変化し、これは有害な環境条件に対する遺伝的応答としても発生します3。細菌細胞の形状とサイズは、細菌の剛性、堅牢性、および表面対体積比を強く決定し、この特性はバイオテクノロジープロセスに利用できます6。
電子顕微鏡は、光ベースの顕微鏡を超えて到達できる高倍率のために、生物学的サンプルを研究するために使用されます。透過型電子顕微鏡(TEM)と走査型電子顕微鏡(SEM)は、この目的のために最も一般的に使用される技術です。ただし、サンプルは、適切な画像を取得するために、顕微鏡のチャンバーに入れる前にいくつかの処理が必要です。サンプルには金のカバーが必要であり、画像全体の取得に使用される時間は長すぎないようにする必要があります。対照的に、原子間力顕微鏡(AFM)は、表面の分析に広く使用されている技術ですが、生物学的サンプルの研究にも採用されています。
表面分析で使用されるAFMモードには、接触モード、非接触モードまたはタッピング、磁力顕微鏡(MFM)、導電性AFM、圧電力顕微鏡(PFM)、ピーク力タッピング(PFT)、接触共振、力量など、いくつかのタイプがあります。各モードは材料の分析に使用され、材料の表面とその機械的および物理的特性に関するさまざまな情報を提供します。しかしながら、PFTは液体培地7中の細胞に関する地形的および機械的データを得ることを可能にするので、PFTのようないくつかのAFMモードはin vitroでの生物学的試料の分析に使用される。
この作業では、すべての古くてシンプルなAFMモデルに含まれる最も基本的なモードであるコンタクトモードを使用しました。AFMは、鋭利なプローブ(直径約<50 nm)を使用して、100 μm未満の領域をスキャンします。プローブは、サンプルに関連する力場と相互作用するためにサンプルに位置合わせされます。表面はプローブでスキャンされ、力を一定に保ちます。次に、カンチレバーが表面を横切って移動するときのカンチレバーの動きを監視することにより、表面の画像が生成されます。収集された情報は、接着性、弾性、粘度、せん断などの表面のナノ機械的特性を提供します。
AFM接触モードでは、カンチレバーは一定のたわみでサンプル全体をスキャンされます。これにより、サンプルの高さ(Z)を決定することができ、これは他の電子顕微鏡技術よりも優れています。AFMソフトウェアは、先端と試料表面の相互作用による3D画像スキャンの生成を可能にし、先端のたわみはレーザーと検出器を介して試料の高さと相関します。
一定の力を持つ静的モード(接触モード)では、出力は高さ(z地形)とたわみまたはエラー信号の2つの異なる画像を表示します。静的モードは、特に静的モードによって加えられる高負荷とねじり力を処理できる空気中の堅牢なサンプルにとって、貴重でシンプルなイメージングモードです。たわみまたは誤差モードは、定力モードで動作します。しかしながら、トポグラフィ画像は、表面構造に偏向信号を加えることによってさらに強調される。このモードでは、たわみはフィードバックパラメータであるため、たわみ信号はエラー信号とも呼ばれます。このチャネルに現れる特徴や形態は、フィードバックループの「エラー」によるものか、一定のたわみ設定値を維持するために必要なフィードバックループによるものです。
AFMのユニークな設計により、コンパクトになり、卓上に収まるほど小さく、原子ステップを解決するのに十分な高解像度を実現しています。AFM装置は、他の電子顕微鏡用の装置よりも低コストであり、メンテナンスコストは最小限です。顕微鏡は、クリーンルームや隔離されたスペースなどの特別な条件を備えたラボを必要としません。必要なのは振動のないデスクだけです。AFMの場合、サンプルは他の技術(金カバー、痩身)のように精巧な準備をする必要はありません。サンプルホルダーには、乾燥サンプルのみを取り付ける必要があります。
AFM接触モードを用いて、細菌の形態やNPの影響を観察しています。支持体上に固定された細菌の集団および細胞形態、ならびに細菌種上のナノ粒子によって生じる細胞損傷を観察することができる。AFMコンタクトモードで得られた画像は、AFMが強力なツールであり、試薬や複雑な手順に制限されないことを確認し、細菌の特性評価のためのシンプルで高速かつ経済的な方法になります。

電子顕微鏡は、細胞構造を特徴付けるために必要なツールの1つです。しかし、観察のための試料調製のため、手順は複雑で費用がかかります。原子間力顕微鏡(AFM)は、3次元での分解能が高く、真空やサンプルの導電率を必要としないため、非常に有用な特性評価技術です。AFMは、さまざまなトポグラフィーとさまざまな種類の材料でさまざまなサンプルをイメージングできます。
