使用在显微镜下获得的生长拟南芥花粉管的荧光延时测试AMEBaS，使用在花粉特异性LAT52启动子下表达的Ca2+ （CaMeleon）8 或pH（pHluorin）6 比率传感器。每 4 秒拍摄一次来自每个通道的图像，耦合到倒置显微镜、前照式相机（2560 像素× 2160 像素，像素大小 6.45 μm）、荧光照明器和水浸物镜 63x，1.2NA。用于CaMeleon的滤光片设置为：激发波长426-450 nm（CFP）和505-515 nm（YFP），发射波长458-487 nm（CFP）和520-550 nm（YFP），而pHluorin的激发波长为318-390 nm（DAPI）和428-475 nm（FITC），发射波长为435-448 nm（DAPI）和523-536 nm（FITC）。添加了一个完整的数据集，用于在 Zenodo （DOI： 10.5281/zenodo.7975350）14 进行测试。
此外，使用根毛数据测试了管道，其中使用光片显微镜 （SPIM） 进行成像，如前所述 15,16 在 UBQ10 启动子的控制下，拟南芥根毛表达遗传编码的 Ca2+ 报告基因 NES-YC3.617。自制的LabView软件控制了光片显微镜的相机采集、样品平移和快门，可以观察两个cpVenus和CFP通道，还可以实时显示它们的比率。延时摄影的每个比率图像都代表了从间隔 3 μm 的 15 个样品切片中获得的 cpVenus 和 CFP 荧光通道图像之间的最大强度投影 （MIP）。MIPs的延时cpVenus/CFP比值被保存下来，并直接用于AMEBaS分析。
