電子顕微鏡は、ナノスケールレベル1で細胞の内部構造を視覚的かつ正確に示すことができる、生体材料の超微細構造を研究するための最も理想的な方法の1つです。しかし、サンプル調製プロセスが複雑でコストが高いため、電子顕微鏡は光学顕微鏡ほど普及していません。近年の電子顕微鏡技術の進歩により、画質の大幅な向上がもたらされ、それと同時に関連する作業負荷も大幅に減少しています。そのため、電子顕微鏡は、さまざまな分野で科学知識を進歩させる上で重要な役割を担っています2。
ショウジョウバエは、標的遺伝子の空間的および時間的発現を正確に制御するための遺伝子操作を行うための優れた動物モデルです3。その上、ショウジョウバエには、哺乳類のモデルと比較して成長期間が短く、飼育が容易であるという利点があります。したがって、ショウジョウバエは形態学研究で広く使用されています4,5。
ショウジョウバエの幼虫では、神経筋接合部(NMJ)ブートンは筋肉に広く分布しており6,7、NMJの免疫染色は、シナプスの量と形態に関する情報を容易に提供できる8,9。A2およびA3セグメントの6番目/7番目の筋肉に位置するNMJボタンは、光学顕微鏡を使用した定量的および形態学的研究に適しています。これは、そのサイズと豊富さ10,11のためです。したがって、ショウジョウバエの幼虫のNMJは、神経科学研究の有用なモデルと考えられている12。

TEMの拡散分布と限られた視覚範囲は、インフラストラクチャ解析に課題をもたらします。このプロトコールは、従来のアプローチを超える革新的で効率的なサンプル調製法を提供します。この高度なTEMサンプル調製方法は、従来の方法と比較して、その後の脱水中にサンプルを平らに保つことにより、サンプルのカールを防ぐのに効果的です。試薬の選択や投与量の調整は、サンプルの種類に応じて必要となります。サンプル調製プロセスで使用される多くの有毒な化学試薬があります。手順を実演するのは、Gan Guangming博士の研究室の修士課程の学生であるSheng Qingyuanです。まず、さまよう後期3齢のショウジョウバエの幼虫をJan溶液で解剖し、標準的な手順に従って全身壁を取得します。幼虫の体壁を解剖室に固定剤で摂氏4度で一晩固定します。翌日、幼虫の体壁を解剖室から1.5ミリリットルの遠心分離チューブに移し、カコジル酸ナトリウム緩衝液で数回すすぎます。幼虫の体壁を1%四酸化オスミウムで2時間固定します。固定後、サンプルを蒸留水で数回洗浄します。2%飽和酢酸小便器溶液をチューブに加え、PHI神経筋接合部を染色します。そして2時間後、サンプルを脱イオン水ですすいでください。次に、はさみを使用して、細孔サイズ270ミクロンの金属メッシュの2つの丸い部分を切り取ります。金属メッシュを平底のボトルの底に置きます。次に、固定された幼虫をメッシュに配置し、続いて別のメッシュを配置します。サンプルをエタノール濃度の増加中で、摂氏4度でそれぞれ20分間脱水します。次に、サンプルをプロピレンオキシドで室温でそれぞれ5分間2回すすぎます。次に、サンプルをプロピレンオキシドとエポキシ樹脂溶液に入れ、続いて純粋なエポキシ樹脂を室温で2時間置きます。埋め込むには、ポリエチレンフィルムを大きな正方形と小さな中空の長方形の支柱にカットします。小さな正方形を大きな正方形に接着します。大きな正方形の中央に十分なエポキシ樹脂を追加します。次に、ステレオスコープをオンにし、筋肉側を上に向けて、事前に配置したエポキシ樹脂にサンプルを配置します。サンプルにエポキシ樹脂を数滴加え、ポリエチレンフィルムで覆います。重合するサンプルを、それぞれ24時間ずつ昇温させます。重合後、鋭利な刃を使用して、A2およびA3セグメントを含む台形を保持しながら、幼虫の体の余分な部分を取り除きます。残った幼虫の体壁から台形を取り出し、AB接着剤で樹脂充填カプセルに貼り付けます。光学顕微鏡下で、A2およびA3セグメントの6番目と7番目の筋肉の位置を確認し、接面を6番目の筋肉と平行に保ちます。次に、A3セグメントの両側に沿ってサンプルの5分の1を切り取ります。ミクロトームを使用して、サンプルの6番目の筋肉から始まる極薄切片を調製します。セクショニング後、各銅メッシュにできるだけ多くのスライスを取り付けます。透過型電子顕微鏡を使用してサンプルを観察します。Lowicryl K4M樹脂とエポキシ樹脂の中で、Lowicryl K4M樹脂はショウジョウバエの体筋のより鮮明で観察しやすい結果を提供しました。共焦点イメージングは、第6筋と第7筋の間の神経筋接合部の視覚化を強化したことを示しました。透過型電子顕微鏡法により、ビーズ状の神経筋接合部が明らかになり、シナプス構造情報が改善されました。サンプルは、金属メッシュの制限により、脱水プロセス全体を通じて平らのままでした。また、プラスチック製品間でサンプルを重合させ、サンプルの折り畳みを防いでいます。将来的には、TEMサンプル調製法を脳および腹側神経索のニューロピーに適用できるようになり、それによって超微細構造研究の可能性が向上します。
