La thrombose est la formation d’un caillot sanguin qui bloque partiellement ou complètement un vaisseau sanguin, entravant la circulation naturelle du sang. Cela conduit à des événements cardiovasculaires graves et mortels, tels que les cardiopathies ischémiques et les accidents vasculaires cérébraux. Les maladies cardiovasculaires sont la principale cause de morbidité et de mortalité, et causent un décès sur quatre dans le monde 1,2,3. Bien que la thrombose se manifeste par un dysfonctionnement du système vasculaire, elle pourrait être le résultat d’une infection microbienne ou virale sous-jacente, d’un trouble immunitaire, d’une tumeur maligne ou d’une affection métabolique. La circulation sanguine est maintenue par l’interaction complexe entre divers composants du système vasculaire, y compris les cellules endothéliales, les globules rouges / blancs, les plaquettes et les facteurs de coagulation4. En cas de lésion vasculaire, les plaquettes interagissent avec les protéines adhésives de la matrice sous-endothéliale et libèrent leur contenu granulaire, ce qui recrute plus de plaquettes5. Parallèlement, la cascade de coagulation est activée, conduisant à la formation et au dépôt de fibrine. En fin de compte, un caillot se forme, contenant des plaquettes et des globules rouges piégés dans un maillage de fibrine6. Bien que des médicaments antiplaquettaires et anticoagulants soient disponibles pour moduler la thrombose, les saignements parasites demeurent une préoccupation majeure avec ces thérapies, nécessitant un réglage fin des dosages et des combinaisons de ces médicaments. Ainsi, il est toujours urgent de découvrir de nouveaux médicaments anti-thrombotiques7.
La thrombose est étudiée à l’aide de plusieurs méthodes pour infliger des lésions vasculaires: mécanique (ligature des vaisseaux), thermique (lésion au laser) et chimique (application FeCl3 / Rose Bengal). La nature de la thrombose varie en fonction de l’emplacement (artériel vs veineux), de la méthode ou de l’étendue de la blessure. Parmi tous ces types, les lésions vasculaires induites par FeCl3 sont la méthode la plus utilisée. Il a été employé chez les souris, les rats, les lapins, les cobayes et les chiens 8,9,10,11,12. La méthode est relativement simple, facile à utiliser et, si les principaux paramètres sont normalisés, elle est sensible et reproductible dans divers systèmes vasculaires (p. ex. artères [carotides et fémorales], veines [jugulaires] et artérioles [cremaster et mésentérique]) (tableau supplémentaire 1).
Ce modèle peut également être utilisé pour approfondir notre compréhension de la mécanique et de la morphologie de la formation des caillots. Cette technique offre uniquement l’avantage d’arrêter la thrombose à différents points de débit, pour étudier les étapes intermédiaires du processus avant qu’il ne devienne occlusif. Les progrès récents de la recherche sur la thrombose ont utilisé ce modèle pour attirer l’attention sur les méthodes non pharmacologiques de thrombolyse13 ou l’administration non invasive d’agents antithrombotiques et/ou fibrinolytiques14,15. Plusieurs groupes ont montré que, lorsque les membranes plaquettaires sont recouvertes de ces traitements, les médicaments peuvent être activés par stimulation thermique pour cibler les caillots16. Les techniques décrites ici peuvent être utiles à des études telles que la validation de leurs résultats au niveau plaquettaire unique. Dans ce manuscrit, le protocole 1 décrit la procédure de base de lésion vasculaire médiée par FeCl3, tandis que le protocole 2 décrit la méthode de collecte et de fixation de l’échantillon de lésion vasculaire pour une analyse plus approfondie par microscopie électronique.

Les maladies cardiovasculaires sont l’une des principales causes de mortalité et de morbidité dans le monde. La thrombose aberrante est une caractéristique commune des affections systémiques comme le diabète et l’obésité, et des maladies inflammatoires chroniques comme l’athérosclérose, le cancer et les maladies auto-immunes. En cas de lésion vasculaire, le système de coagulation, les plaquettes et l’endothélium agissent généralement de manière orchestrée pour prévenir les saignements en formant un caillot sur le site de la blessure. Les anomalies de ce processus entraînent soit des saignements excessifs, soit une thrombose incontrôlée / une activité antithrombotique insuffisante, ce qui se traduit par une occlusion des vaisseaux et ses séquelles. Le modèle de lésion carotidienne induite par FeCl3 est un outil précieux pour sonder comment la thrombose commence et progresse in vivo. Ce modèle implique des dommages endothéliaux/dénudation et la formation subséquente de caillots sur le site blessé. Il fournit un test quantitatif très sensible pour surveiller les dommages vasculaires et la formation de caillots en réponse à différents degrés de dommages vasculaires. Une fois optimisée, cette technique standard peut être utilisée pour étudier les mécanismes moléculaires sous-jacents à la thrombose, ainsi que les changements ultrastructuraux dans les plaquettes dans un thrombus en croissance. Ce test est également utile pour étudier l’efficacité des agents antithrombotiques et antiplaquettaires. Cet article explique comment initier et surveiller la thrombose artérielle induite par FeCl3 et comment prélever des échantillons pour analyse par microscopie électronique.

Avec cette méthode, on peut déterminer quels composants des plaquettes et du système hémostatique sont importants pour la thrombose. De plus, cette méthode peut déterminer comment la croissance et l’architecture du thrombus sont affectées au niveau cellulaire. Le principal avantage de cette technique est qu’elle permet une manière reproductible d’étudier de nombreux stades de thrombose au niveau structurel.Une personne novice dans cette technique devrait éviter de trop manipuler le tissu dans la zone chirurgicale. Cela peut entraîner des saignements et compliquer à l’excès l’identification de l’artère carotide. Commencez à marquer le site de la blessure après avoir exposé l’artère carotide en encerclant lâchement la région entre deux fils chirurgicaux d’une taille de 0,1 millimètre.Placez la sonde sous l’artère distale du filetage inférieur. Insérez le morceau de plastique sous l’artère entre les deux fils pour marquer le site des dommages causés par le chlorure de fer. Prenez des mesures de débit avant d’effectuer une blessure pour noter l’écoulement basal.Effectuez la blessure en plaçant du papier filtre imbibé de chlorure de fer sur l’artère pendant trois minutes. Retirez le papier filtre après trois minutes pour éviter l’étendue variable de la blessure et faciliter le comptage du débit par la sonde. Ajouter une solution saline sur le site de la blessure pour éliminer tout excès de chlorure de fer résiduel.Surveillez la lecture du flux. Lorsque le débit tombe en dessous de 50 % de la valeur initiale, retirez la sonde. Séchez rapidement la zone et ajoutez le fixateur pour fixer la zone de blessure à l’extérieur.Ensuite, tenez rapidement l’artère près de la zone de blessure avec des forceps. Couper en aval du filetage inférieur et en amont du fil supérieur. Placez le tissu sur la boîte de culture de tissus en plastique dans la même orientation que celle qui a été recueillie et ajoutez quelques gouttes du fixateur.À l’aide d’un scalpel, nettoyez le tissu supplémentaire autour de l’artère. Assurez-vous d’enregistrer l’orientation du flux sanguin en coupant une extrémité horizontalement et l’autre effilée ou oblique. Placer l’échantillon dans un fixateur pendant une heure à température ambiante, puis stocker l’échantillon dans du paraformaldéhyde à 1% à 4 degrés Celsius jusqu’à ce qu’il soit envoyé dans de la glace humide pour analyse par microscopie électronique.Les données ont été tracées sous la forme d’une courbe de survie de Kaplan-Meier, un diagramme à points avec des barres montrant le flux sanguin terminal au moment de l’arrêt du flux sanguin ou de la fin d’une expérience, ou sous forme de graphique linéaire. Le flux sanguin a augmenté soudainement dans certains cas après une diminution progressive pendant quelques minutes, indiquant une excrétion partielle du thrombus en croissance et a été considéré comme un événement d’embolisation. La morphologie du thrombus occlusif a également été étudiée à l’aide de cette méthode.Le prélèvement rapide de l’échantillon pendant le processus de thrombose est l’étape la plus importante, car un retard dans ce processus peut entraîner une mauvaise interprétation de notre analyse structurelle.
