漢方加工とは、毒性を効果的に軽減または排除し、TCMの安全性を向上させることができる製薬技術を指します。EFの伝統的な加工方法は、マトン油で炒めることで、EFの毒性を軽減し、腎臓を温め、陽8を促進する効果を高めます。この処理方法は、中国薬局方および各種処理仕様に含まれています1.EFのプロセスは次のようにのみ指定されています:EF100 kgごとに20 kgの羊水(精製)が追加され、均一で光沢がある1になるまで穏やかに焼成されます。上記の標準には厳密なEF処理メソッドパラメータがないため、一貫性を提供するためにローカル処理仕様が統一されていません。したがって、EFプロセスの体系的な研究を行うことは有用でしょう。本論文では、Box-Behnken実験計画応答曲面法を用いてEFの処理技術を最適化した。
Box-Behnken実験計画は、工程の因子を最適化するために通常使用される方法です。抽出パラメータは、重回帰式適合因子と効果値の間の関数関係を確立することによって最適化できます。最近、この方法は、TCM抽出5、6、7および処理9、10、11の研究に広く使用されています。塩加工ソラレ科フルクタス12、ワイン加工フルータス13、ローストシナモミラムルス14など、ボックスベンケン設計に従った塩加工、ワイン加工、炒め物を含むTCM調製方法がさまざまな研究で報告されています。この方法は、テスト時間が短縮され、テスト精度が高く、多要素およびマルチレベルのテストに適しています。この方法は、直交計画試験方法よりも単純であり、均一設計方法15よりも包括的である。得られた関係は、テスト範囲内の任意のテストポイントの予測値を決定することができ、これは大きな利点である。ゼブラフィッシュモデルを使用して、EFが処理後に毒性が低いかどうかをテストできます。
TCM毒性試験において、ゼブラフィッシュモデルには、細胞実験のハイスループットとげっ歯類実験との類似性という2つの利点があります16。このモデルは、サイズが小さく、産卵率が高く、繁殖サイクルが短く、繁殖が容易であることが特徴です。このモデルは、細胞培養プレートでの大規模な同期実験に使用でき、実験薬の投与量が少なく、実験サイクルが短く、コストが低く、実験プロセス全体の観察と操作が容易です17。ゼブラフィッシュの胚は透明で急速に成長します。したがって、異なる発生段階における内臓組織に対する薬物の毒性および催奇形性効果は、顕微鏡下で直接観察することができる18。ゼブラフィッシュとヒトの遺伝子相同性は85%にもなります18。ゼブラフィッシュのシグナル伝達経路はヒトのそれと類似している18。ゼブラフィッシュの生物学的構造と生理学的機能は、哺乳類のものと非常に類似しています18。したがって、薬物検査用のゼブラフィッシュモデルは、信頼性が高く、ヒトに完全に適用可能な実験動物を提供することができます19。
本研究では,イカリイン,エピメジンA,エピメジンB,エピメジンC,バオフオシドIの含有量を評価指標として,EF加工技術で使用するマトン油の量と温度,揚げ温度を最適化するためにBox-Behnken設計応答曲面法を用いた。ゼブラフィッシュモデルを用いて、EFに対する処理の減衰効果を評価するために、処理前後のゼブラフィッシュ胚発生に対するEF水抽出物の効果を予備的に調査した。

optimization of the epimedii folium mutton-oil processing technology and testing its effect on zebrafish embryonic development

伝統的な漢方薬(TCM)として、エピメディフォリウム(EF)は、>2,000年前の医学と食品の歴史があります。臨床的には、マトンオイルで処理されたEFは薬としてよく使用されます。近年、EFを原料とした製品の安全性リスクや副作用の報告が徐々に増加しています。処理により、TCMの安全性を効果的に向上させることができます。TCM理論によれば、マトンオイル処理はEFの毒性を軽減し、腎臓に対するその調子効果を高めることができます。しかし、EFマトンオイル加工技術の体系的な研究と評価は不足しています。この研究では、Box-Behnken実験計画応答曲面法を使用して、複数のコンポーネントの内容を評価することにより、処理技術の主要なパラメーターを最適化しました。その結果、EFの最適なマトン油加工技術は、マトンオイルを120°C±10°Cで加熱し、粗EFを加え、均一に光沢があるまで189°C±10°Cまで穏やかに炒め、取り除いて冷却することであることが示された。EF100 kgごとに、15 kgのマトンオイルを使用する必要があります。粗精製および羊油処理EFの水性抽出物の毒性および催奇形性をゼブラフィッシュ胚発生モデルで比較した。結果は、粗ハーブグループがゼブラフィッシュ奇形を引き起こす可能性が高く、その半分の最大致死EF濃度が低いことを示しました。結論として、最適化されたマトンオイル処理技術は安定していて信頼性が高く、再現性も良好でした。EFの水性抽出物は、特定の用量ではゼブラフィッシュ胚の発生に対して毒性があり、毒性は加工薬よりも生薬の方が強かった。結果は、マトン油処理が粗EFの毒性を低下させることを示した。これらの知見は、マトン油処理EFの品質、均一性、および臨床的安全性を向上させるために使用できます。

本研究では、広く用いられている処理方法を採用し、安定した信頼性の高い持続技術を獲得した。結果は、処理されたEpimediiの毒性が減少したことを示した。応答曲面法は、実験が少なくて済み、サイクルが短いため、従来の数学的統計に存在する問題を回避できるだけでなく、ユーザーが最適な条件を選択するのにも役立ちます。この方法は、化学工業、生物工学、食品産業、漢方薬の調製、漢方薬の抽出などでも広く使用できます。溶液を調製し、クロマトグラフィーを実行した後、影響要因として、羊肉油の量、羊肉油の温度、揚げ物温度など、EF処理の主要なパラメーターを選択します。評価指標として、イカリイン、EA、EB、EC、およびBIコンテンツの包括的なスコアを使用します。応答曲面解析ソフトウェアを使用して、Box-Behnken応答曲面実験を設計し、2次応答曲面を探索し、2次多項式モデルを構築します。新しいBox-Behnken計画を選択し、数値因子オプションを3に設定します。因子A、B、Cを設定し、続行をクリックしてから応答オプションを1に設定し、続行をクリックして計画を完了します。特定のパラメータに従ってEFを処理し、たとえば、注文番号1の場合、精製されたマトンオイルを体積で15%とし、摂氏50度に加熱して溶かします。溶かしたマトンに粗EFを加えます。穏やかに光沢があるまで穏やかな火で炒め、EFを取り出して冷まします。加工品の試験液を調製し、原稿に記載のHPLCで分析します。各化学組成の保持時間とピーク面積を記録し、外部標準曲線に対して各試験溶液中のイカリイン、EA、EB、EC、およびBIの含有量を計算します。総合得点計算式を用いて17の実験群の総合得点を算出する。17グループの実験の包括的なスコアリング結果をデータ分析ソフトウェアにインポートして、実験データを分析します。評価項目で、2次プロセス次数オプションと多項式モデルタイプオプションを選択します。マトン油量と揚げ温度の応答面は他の因子に比べて急峻であり,等高線は楕円形になる傾向があり,これら2因子の交互作用が他の因子間の交互作用よりも有意であることが示された。混合標準物質、粗エピメディフォリウムおよびエピメディフォリウム加工品のHPLCクロマトグラムは、生EF中のBI含有量が低いが、処理後に増加することを示している。粗群は、1ミリリットル当たり150マイクログラムの薬物濃度で明らかな脊椎変形、体の湾曲、心膜浮腫、および肝変形を示すが、これらの変化は処理群の幼魚ではまれであった。生薬群のゼブラフィッシュはすべて、200マイクログラム/ミリリットルの薬物投与後24時間で死亡したが、処理群の死亡率はわずか6.67%であったEF1ミリリットルあたり200マイクログラムの投与は、72時間後に生薬群のすべてのゼブラフィッシュの死を引き起こしたが、処理群では、EFのミリリットルあたり500マイクログラムは、すべてのゼブラフィッシュの死を引き起こした。受精後72時間における2つの実験群の致死濃度の中央値は、LC 50が粗群で151.3マイクログラム/ミリリットル、処理群で219.8マイクログラム/ミリリットルであることを示した。実験で最も重要なことは、応答曲面法の因子を選択し、これらの因子のパラメータを決定することです。
