Epimedii folium (EF)은 Epimedium brevicornu Maxim., Epimedium sagittatum (Sieb. et Zucc.)의 말린 잎입니다. Maxim., Epimedium pubescens Maxim., 또는 Epimedium koreanum Nakai. EF는 골다공증, 갱년기 증후군, 유방 덩어리, 고혈압, 관상 동맥 심장 질환 및 기타 질병을 치료하는 데 사용할 수 있습니다1. 중국 전통 의학(TCM)인 EF는 2,000년 이상의 의학 및 식품 분야의 역사를 가지고 있습니다. 저렴한 가격과 신장을 강화하는 현저한 효과로 인해 의약품 및 건강 식품에 널리 사용됩니다. EF의 과정은 다음과 같이 지정됩니다 : EF 100kg 당 양수 20kg (정제 된)을 첨가하고 균일하고 광택이 날 때까지 온화하게 소성합니다1. 위의 표준에는 엄격한 EF 처리 방법 매개 변수가 없으므로 일관성을 제공하기 위해 로컬 처리 사양이 통합되지 않았습니다. 따라서 EF 프로세스에 대한 체계적인 연구를 수행하는 것이 유용 할 것입니다. 본 논문에서는 EF의 처리 기술을 최적화하기 위해 Box-Behnken 실험 설계-반응 표면 방법을 사용하였다.
Box-Behnken 실험 설계는 공정의 요인을 최적화하는 데 일반적으로 사용되는 방법입니다. 추출 매개변수는 다중 회귀 방정식 피팅 요인과 효과 값 간의 기능적 관계를 설정하여 최적화할 수 있습니다. 최근에, 이 방법은 TCM 추출 5,6,7 및 처리 9,10,11을 연구하는데 널리 이용되고 있다. 다양한 연구에서 소금 가공 Psoraleae fructus12, 와인 가공 Cnidii fructus13 및 볶은 Cinnamomi ramulus14와 같은 Box-Behnken 설계에 따라 소금 가공, 와인 가공 및 볶음을 포함하는 TCM 준비 방법이 보고되었습니다. 이 방법은 테스트 시간을 단축하고 테스트 정확도를 높이며 다단계 및 다단계 테스트에 적합합니다. 이 방법은 직교 설계 시험 방법보다 간단하고 균일 설계 방법15보다 포괄적이다. 얻은 관계는 테스트 범위 내의 모든 테스트 포인트의 예측 값을 결정할 수 있으며 이는 큰 이점입니다. 제브라피쉬 모델은 처리 후 EF의 독성이 덜한지 테스트하는 데 사용할 수 있습니다.
TCM 독성 연구에서, 제브라피쉬 모델은 세포 실험의 높은 처리량과 설치류 실험과의 유사성이라는 두 가지 장점을 가지고 있다16. 이 모델은 작은 크기, 높은 산란 속도, 짧은 번식주기 및 번식 용이성이 특징입니다. 이 모델은 세포 배양 플레이트에서 대규모 동기 실험에 사용할 수 있으며 실험 약물 투여량이 적고 실험 주기가 짧으며 비용이 저렴하며 전체 실험 과정을 관찰하고 작동하기 쉽습니다17. 제브라피쉬 배아는 투명하고 빠르게 발달합니다. 그러므로, 상이한 발달 단계에서 내장 조직에 대한 약물의 독성 및 기형 유발 효과는 현미경으로 직접 관찰할 수 있다18. 제브라피쉬와 인간 사이의 유전자 상동성은 85%에 달합니다18. 제브라피쉬의 신호 전달 경로는 인간의 신호 전달 경로와 유사하다18. 제브라피쉬의 생물학적 구조와 생리적 기능은 포유류의 그것과 매우 유사하다18. 그러므로, 약물 실험을 위한 제브라피쉬 모델은 인간에게 신뢰할 수 있고 완전히 적용할 수 있는 실험 동물을 제공할 수 있다19.
본 연구에서는 Box-Behnken 설계-반응 표면 방법론을 사용하여 EF 가공 기술에 사용되는 양고기 기름의 양과 온도, 튀김 온도를 최적화하였으며, 이카리인, 에피메딘 A, 에피메딘 B, 에피메딘 C, 바오후오사이드 I의 함량을 평가 지표로 하였다. 제브라피쉬 모델은 EF에 대한 처리의 감쇠 효과를 평가하기 위해 처리 전후에 제브라피쉬 배아 발달에 대한 EF 물 추출물의 효과를 예비 적으로 탐색하는 데 사용되었습니다.

중국 전통 의학(TCM)인 에피메디 폴리움(EF)은 2,000년 > 된 의학 및 식품 분야에서 역사를 가지고 있습니다. 임상적으로 양고기 기름으로 가공한 EF는 약으로 사용되는 경우가 많습니다. 최근에는 EF를 원료로 사용하는 제품의 안전 위험과 부작용에 대한 보고가 점차 증가하고 있습니다. 처리는 TCM의 안전성을 효과적으로 향상시킬 수 있습니다. TCM 이론에 따르면 양고기 기름 가공은 EF의 독성을 줄이고 신장에 대한 강장제 효과를 향상시킬 수 있습니다. 그러나 EF 양고기 기름 가공 기술에 대한 체계적인 연구와 평가가 부족하다. 이 연구에서는 Box-Behnken 실험 설계-반응 표면 방법론을 사용하여 여러 구성 요소의 내용을 평가하여 처리 기술의 주요 매개변수를 최적화했습니다. 그 결과 EF의 최적 양고기 기름 가공 기술은 다음과 같다: 양고기 기름을 120°C ± 10°C에서 가열하고, 조잡한 EF를 첨가하고, 고르게 광택이 날 때까지 189°C ± 10°C로 부드럽게 볶은 다음, 그것을 제거하고 식힌다.
